Biosensor-based assay of exosome biomarker for early diagnosis of cancer

Abstract: Cancer imposes a severe threat to people’s health and lives, thus pressing a huge medical and economic burden on individuals and communities. Therefore, early diagnosis of cancer is indispensable in the timely prevention and effective treatment for patients. Exosome has recently become an attractive cancer biomarker in noninvasive early diagnosis because of the unique physiology and pathology functions, which reflects remarkable information regarding the cancer microenvironment, and plays an important role in the occurrence and evolution of cancer. Meanwhile, biosensors have gained great attention for the detection of exosomes due to their superior properties, such as convenient operation, real-time readout, high sensitivity, and remarkable specificity, suggesting promising biomedical applications in the early diagnosis of cancer. In this review, the latest advances of biosensors regarding the assay of exosomes were summarized, and the superiorities of exosomes as markers for the early diagnosis of cancer were evaluated. Moreover, the recent challenges and further opportunities of developing effective biosensors for the early diagnosis of cancer were discussed.

Abstract: The aim of this paper was to explore the relationship between energy metabolism and the meridian phenomenon. The manner of change of oxygen partial pressure in acupoints and no-acupoints of the urinary bladder meridian of goats was observed with a needle-type tissue oxygen tension sensor after the acupuncture effect was blocked by Ca complexation with ethylenediamine tetraacetic acid (EDTA)-Na . The results showed that: (1) the concentration of calcium ion in urinary bladder meridian acupoints was higher than that in no-acupoints ( <0.01 or <0.05); (2) the tissue oxygen tension in urinary bladder meridian acupoints was higher than that in no-acupoints ( <0.01); (3) the oxygen partial pressure in acupoints decreased significantly after injection with EDTA-Na , compared with injection of either normal sodium or nothing ( <0.01); (4) the oxygen partial pressure in acupoints was higher than that in no-acupoints after EDTA-Na injection ( <0.01). Acupuncture could reduce oxygen partial pressure in acupoints by regulating microcirculation and increasing metabolic levels.

Genetic biosensors for small-molecule products: Design and applications in high-throughput screening

Abstract: Overproduction of small-molecule chemicals using engineered microbial cells has greatly reduced the production cost and promoted environmental protection. Notably, the rapid and sensitive evaluation of the concentrations of the desired products greatly facilitates the optimization process of cell factories. For this purpose, many genetic components have been adapted into biosensors of small molecules, which couple the intracellular concentrations of small molecules to easily detectable readouts such as fluorescence, absorbance, and cell growth. Such biosensors allow a high-throughput screening of the small-molecule products, and can be roughly classified as protein-based and RNA-based biosensors. This review summarizes the recent developments in the design and applications of biosensors for small-molecule products.

Shrink-induced graphene sensor for alpha-fetoprotein detection with low-cost self-assembly and label-free assay

Abstract:

Combination of shrink induced nano-composites technique and layer-by-layer (LbL) self-assembled graphene challenges controlling surface morphology. Adjusting shrink temperature achieves tunability on graphene surface morphology on shape memory polymers, and it promises to be an alternative in fields of high-surface-area conductors and molecular detection. In this study, self-assembled graphene on a shrink polymer substrate exhibits nanowrinkles after heating. Induced nanowrinkles on graphene with different shrink temperature shows distinct surface roughness and wettability. As a result, it becomes more hydrophilic with higher shrink temperatures. The tunable wettability promises to be utilized in, for example, microfluidic devices. The graphene on shrink polymer also exhibits capability of being used in sensing applications for pH and alpha-fetoprotein (AFP) detection with advantages of label free and low cost, due to self-assembly technique, easy functionalization, and antigen-antibody reaction on graphene surface. The detection limit of AFP detection is down to 1 pg/mL, and therefore the sensor also has a significant potential for biosensing as it relies on low-cost self-assembly and label-free assay.

Microbial fuel cell with high content solid wastes as substrates: a review

Abstract: Fundamentals and configuration design of MFCs fueled by HCSW were reviewed. HCSWs including sewage sludge, biomass and biowaste treated in MFCs were summarized. HCSW based MFCs technologies covered the types of sediment, soil, wetland and plant. Activated sludge process and composting could be coupled with HCSW-MFCs. HCSW-MFCs could be applied in bioremediation and biosensing. With the increasing concern about the serious global energy crisis and high energy consumption during high content solid wastes (HCSWs) treatment, microbial fuel cell (MFC) has been recognized as a promising resource utilization approach for HCSW stabilization with simultaneous electrical energy recovery. In contrast to the conventional HCSW stabilization processes, MFC has its unique advantages such as direct bio-energy conversion in a single step and mild reaction conditions (viz., ambient temperature, normal pressure, and neutral pH). This review mainly introduces some important aspects of electricity generation from HCSW and its stabilization in MFC, focusing on: (1) MFCs with different fundamentals and configurations designed and constructed to produce electricity from HCSW; (2) performance of wastes degradation and electricity generation; (3) prospect and deficiency posed by MFCs with HCSW as substrates. To date, the major drawback of MFCs fueled by HCSW is the lower power output than those using simple substrates. HCSW hydrolysis and decomposition would be a major tool to improve the performance of MFCs. The optimization of parameters is needed to push the progress of MFCs with HCSW as fuel.
