Abstract:

Two decades have passed since the first bacterial whole-genome sequencing, which provides new opportunity for microbial genome. Consequently, considerable genetic diversity encoded by bacterial genomes and among the strains in the same species has been revealed. In recent years, genome sequencing techniques and bioinformatics have developed rapidly, which has resulted in transformation and expedited the application of strategy and methodology for bacterial genome comparison used in dissection of infectious disease epidemics. Bacterial whole-genome sequencing and bioinformatic computing allow genotyping to satisfy the requirements of epidemiological study in disease control. In this review, we outline the significance and summarize the roles of bacterial genome sequencing in the context of bacterial disease control and prevention. We discuss the applications of bacterial genome sequencing in outbreak detection, source tracing, transmission mode discovery, and new epidemic clone identification. Wide applications of genome sequencing and data sharing in infectious disease surveillance networks will considerably promote outbreak detection and early warning to prevent the dissemination of bacterial diseases.

approach for fabricating photoluminescent carbon dots and its potential application as an efficient sensor probe

Abstract: The past decade has witnessed a variety of members of the carbon family along with exposure of carbon dots due to their magnificent properties in sensing, bioimaging, catalytic applications, biomedical fields, and so on. Herein, we report the simple hydrothermal method to fabricate photoluminescent doped carbon quantum dots for the detection of noxious lead(II) ions. Lead(II) ion is very venomous for both the environment and human health for which its detection is demanded area in the research field. The as-prepared carbon dots show excellent photostability, low toxicity and significant photoluminescence properties along with good water solubility. Along with these properties, carbon dots have a quantum yield of approximately 15%. In the practical field of application, these carbon dots have been used as sensing probes for the detection of lead(II) ions with a detection limit of 60 nmol·L–1. The fluorescence intensity of carbon dots was remarkably quenched in the presence of the lead(II) ion selectively among all the tested metal ions. Furthermore, we have studied the Stern–Volmer relationship for lead(II) quenching along with the explanation of the probable quenching mechanism. Ability of the doped carbon dots in heavy metal ions sensing in an environmental sample was demonstrated.

A simple umbelliferone based fluorescent probe for the detection of nitroreductase

Abstract: A simple nitrobenzyl-umbelliferone (NCOU1) was synthesised containing a nitroreductase (NTR) trigger moiety. The presence of NTR, resulted in the fragmentation of the parent molecule and release of the highly emissive fluorophore umbelliferone an NTR-catalyzed reduction of the nitro group. In the presence of the NTR enzyme, NCOU1 gave rise to a 5-fold increase in fluorescence intensity at 455 nm and was selective for NTR over other reductive enzymes. These results indicate that NCOU1 can be used as a simple assay for the detection of NTR.

Abstract: A series of dehydroabietic acid-based diarylamines have been synthesized in order to investigate their fluorescent properties, photostability, cell toxicity and fluorescence imaging. The geometries as well as their molecular properties were optimized at the B3LYP/6-31G* level using Gaussian 03. The results indicate that molecular geometry, HOMO and LUMO energies, and energy gaps are important to predict absorption and fluorescent properties. Five of the compounds can be effectively taken up by human cervical carcinoma, human hepatocellular carcinoma SMMC-7721, human gastric cancer SGC-7901 and human lung adenocarcinoma A549 cells and strong blue fluorescent signals are detected in these cells. These compounds are potential candidates for fluorescent probes in biological diagnosis.
