Efficient production of D-1,2,4-butanetriol from D-xylose by engineered

摘要: We have developed a whole-cell bioconversion system for the production of D-1,2,4-butanetriol (BT) from renewable biomass. A plasmid pETduet- T7- T7- was constructed and transformed to BL21(DE3) to obtain the whole cells of BL21- capable of bioconversion D-xylose to BT. Then, the factors including carbon sources, nitrogen sources, metal ions, and culture conditions (pH, temperature, IPTG) were identified, and their effects on the whole-cell activity for BT production were investigated. To obtain the highest whole-cell activity, the optimal cultivation parameters are: 15 g·L yeast extract, 5 g·L sucrose, 3g·L KH PO , 5 g·L NaCl, 3g·L NH Cl, 0.25 g·L MgSO ?7H O and 1 mL·L the mixture of trace elements. With the optimized whole cells of BL21- , 60 g·L of xylose was converted to 28g·L BT with a molar yield of 66 %, which is higher than those reported in the biotechnological system.

摘要: 随着消费级RGB-D摄像机的快速发展,真实世界的室内三维场景建模和机器人应用越来越受到重视。然而,室内三维场景建模仍具有挑战性,因室内物体结构可能具有较高复杂性,在此情况下,消费者级传感器采集的RGB-D数据质量需进一步提升。近年来,在提高消费者级传感器采集的RGB-D数据质量方面,有很多值得关注的研究。本文介绍了室内场景建模方法的最新进展、室内公共数据集和库以及RGB-D设备的典型应用,包括室内定位和紧急疏散。

PROTECTIVE ROLES OF D1 PROTEIN TURNOVER AND THE XANTHOPHYLL CYCLE IN TOMATO (SOLANUM LYCOPERSICUM) UNDER

摘要:

D1 protein turnover and the xanthophyll cycle (XC) are important photoprotective mechanisms in plants that operate under adverse conditions. Here, streptomycin sulfate (SM) and dithiothreitol (DTT) were used in tomato plants as inhibitors of D1 protein turnover and XC to elucidate their photoprotective impacts under sub-high temperature and high light conditions (HH, 35°C, 1000 µmol·m-2·s-1). SM and DTT treatments significantly reduced the net photosynthetic rate, apparent quantum efficiency, maximum photochemical efficiency, and potential activity of photosystem II, leading to photoinhibition and a decline in plant biomass under HH. The increase in reactive oxygen species levels resulted in thylakoid membrane lipid peroxidation. In addition, there were increased non-photochemical quenching and decreased chlorophyll pigments in SM and DTT application, causing an inhibition of D1 protein production at both transcriptional and translational levels. Overall, inhibition of D1 turnover caused greater photoinhibition than XC inhibition. Additionally, the recovery levels of most photosynthesis indicators in DTT-treated plants were higher than in SM-treated plants. These findings support the view that D1 turnover has a more important role than XC in photoprotection in tomato under HH conditions.

基于三维参数模型重建的心脏病理视觉识别 Research Article

摘要: 心脏图像的视觉识别对于心脏病理诊断和治疗具有重要意义。由于可用标注数据集有限,传统方法通常基于三维心脏图像的二维切片对病理分类特征进行提取,难以确保心脏解剖结构的整体一致性。为此,本文提出一种基于三维参数模型重建的心脏病理分类方法。首先,基于收缩末期和舒张末期时相心脏图像的多个三维心脏成像数据重建三维心脏模型。其次,基于重建的三维心脏模型,通过统计形状模型方法构建三维参数模型。然后,基于三维统计形状模型及其视觉知识约束对心脏数据进行增强。最后,提取不同时相的三维心脏模型的形状和运动特征,对心脏病理进行分类。在STACOM公开挑战赛的ACDC数据集上的实验验证了所提方法的优越性和有效性。

VITHM1 mediated green synthesis of silver nanoparticles: Mechanism and biological applications

摘要: We present the microbial green synthesis of silver nanoparticles (NPs) by VITHM1 strain (MTCC No. 12465). The secondary metabolites in the cell free supernatant of this bacterium when incubated with 1 mmol/L AgNO , mediated the biological synthesis of AgNPs. The synthesized AgNPs were characterized by UV-visible spectrum, X-ray diffraction (XRD), atomic force microscope, scanning electron microscopy equipped with energy dispersive spectroscopy, transmission electron microscopy, FT-IR spectroscopy, dynamic light scattering and zeta potential. They were highly stable and, spherical in shape with the average size of 30?50 nm. The secondary metabolites involved in the formation of AgNPs were identified gas chromatography-mass spectrography. The 3D structure of the unit cell of the synthesized AgNPs was determined using XRD data base. The synthesized AgNPs exhibited significant antibacterial activity against tested bacterial pathogens, and did not show haemolysis on human red blood cells. This green synthesis could provide a new platform to explore and use AgNPs as antibacterial therapeutic agents.

蓄滞洪区洪水演进一、二维数值仿真及其在洼淀联合调度中的应用

摘要:

为了实现蓄滞洪区内各洼淀实时洪水调度,考虑到蓄滞洪区内复杂河网与洼淀交错的情况,建立了河道、滞洪区联合计算的一、二维衔接洪水演进数学模型,在考虑铁路、公路、堤防及桥涵等阻水建筑物等复杂边界条件下,采用有限体积法进行离散和求解。结合蓄滞洪区实时洪水调度仿真系统的总体框架,建立了模拟洪水在蓄滞洪区内任意位置的水位、流速、淹没水深、历时等演进过程的一、二维洪水演进数值仿真系统。结合大清河实测资料对东淀滞洪区洪水演进进行了模型验证,结果基本吻合,并应用该系统对大清河滞洪区五洼联合调度方案进行了实时模拟,分析了洼淀联合调度下出现的倒淀现象,对不同方案进行了比较分析,实现了实时的洪水调度分析。

AML1-ETO driven acute leukemia: insights into pathogenesis and potential therapeutic approaches

摘要:

The AML1-ETO fusion transcription factor is generated by the t(8;21) translocation, which is present in approximately 4%–12% of adult and 12%–30% of pediatric acute myeloid leukemia (AML) patients. Both human and mouse models of AML have demonstrated that AML1-ETO is insufficient for leukemogenesis in the absence of secondary events. In this review, we discuss the pathogenetic insights that have been gained from identifying the various events that can cooperate with AML1-ETO to induce AML in vivo. We also discuss potential therapeutic strategies for t(8;21) positive AML that involve targeting the fusion protein itself, the proteins that bind to it, or the genes that it regulates. Recently published studies suggest that a targeted therapy for t(8;21) positive AML is feasible and may be coming sometime soon.

A simple umbelliferone based fluorescent probe for the detection of nitroreductase

摘要: A simple nitrobenzyl-umbelliferone (NCOU1) was synthesised containing a nitroreductase (NTR) trigger moiety. The presence of NTR, resulted in the fragmentation of the parent molecule and release of the highly emissive fluorophore umbelliferone an NTR-catalyzed reduction of the nitro group. In the presence of the NTR enzyme, NCOU1 gave rise to a 5-fold increase in fluorescence intensity at 455 nm and was selective for NTR over other reductive enzymes. These results indicate that NCOU1 can be used as a simple assay for the detection of NTR.

Construction of a CaHPO4-PGUS1 hybrid nanoflower through protein-inorganic self-assembly, and its applicationin glycyrrhetinic acid 3-O-mono-β-D-glucuronide preparation

摘要: Glycyrrhetinic acid 3- -mono- -D-glucuronide (GAMG), an important pharmaceutical intermediate and functional sweetener, has broad applications in the food and medical industries. A green and cost-effective method for its preparation is highly desired. Using site-directed mutagenesis, we previously obtained a variant of -glucuronidase from Li-3 (PGUS1), which can specifically transform glycyrrhizin (GL) into GAMG. In this study, a facile method was established to prepare a CaHPO -PGUS1 hybrid nanoflower for enzyme immobilization, based on protein-inorganic hybrid self-assembly. Under optimal conditions, 1.2 mg of a CaHPO -PGUS1 hybrid nanoflower precipitate with 71.2% immobilization efficiency, 35.60 mg∙g loading capacity, and 118% relative activity was obtained. Confocal laser scanning microscope and scanning electron microscope results showed that the enzyme was encapsulated in the CaHPO -PGUS1 hybrid nanoflower. Moreover, the thermostability of the CaHPO -PGUS1 hybrid nanoflower at 55°C was improved, and its half-life increased by 1.3 folds. Additionally, the CaHPO -PGUS1 hybrid nanoflower was used for the preparation of GAMG through GL hydrolysis, with the conversion rate of 92% in 8 h, and after eight consecutive runs, it had 60% of its original activity.

China ADS sub-critical experimental assembly—Venus-1 and preliminary experiment

摘要: China s accelerator-driven sub-critical system (ADS) sub-critical experimental assembly Venus-1 and the preliminary experiment is presented. The core of Venus-1 is a coupled one of a fast neutron zone and a thermal neutron zone. The fast neutron zone is at the centre of the core and formed by natural uranium fuel. A fast neutron spectrum field can be produced in the fast neutron zone and used for the transmutation of minor actinides (MAs). The thermal neutron zone surrounds the fast neutron zone and is formed by low-enriched uranium fuel. It is a fission zone. An epithermal neutron zone between the fast neutron zone and the thermal neutron zone can be established for the transmutation of long-lived fission products (LLFP). On July 18, 2005, the first fuel element was loaded into the Venus-1 sub-critical assembly and some preliminary experiments about the sub-critical neutronics were performed. The Venus-1 can be driven by an Am-Be source or other steady neutron source (Cf-252, D-D reaction and D-T reaction) to study the effect of the external neutron source with different energies or a D-T pulsed neutron source on the dynamic characteristics.
