Association of miRNA-122-binding site polymorphism at the interleukin-1 α gene and its interaction with

摘要:

This study was designed to investigate the contribution of miRNA-122-binding site polymorphism at the IL-1A gene and its multiplicative interactions with hepatitis B virus (HBV) mutations in the risk of hepatocellular carcinoma (HCC). A total of 1021 healthy controls, 302 HBV surface antigen (HBsAg) seroclearance subjects, and 2011 HBsAg-positive subjects (including 1021 HCC patients) were enrolled in this study. Quantitative PCR was used to genotype rs3783553. HBV mutations were determined by direct sequencing. Multivariate logistic regression analyses were performed to test the associations of rs3783553, mutations, and their interactions with the risk of HCC. No significant association was found between rs3783553 and the risk of HCC among healthy controls, HBsAg seroclearance subjects, HBsAg-positive subjects without HCC, and all controls. Additionally, rs3783553 was not significantly associated with chronic HBV infection, liver cirrhosis, HBV e antigen seroconversion, abnormal alanine aminotransferase, and high viral load (>104 copies/ml). However, the TTCA insertion allele of rs3783553 was significantly associated with an increased frequency of HBV C7A mutation compared with homozygous TTCA deletion carriers [(del/ins+ ins/ins) vs. del/del, adjusted odds ratio (OR)=1.48, 95% confidence interval (CI)=1.09-2.02, P=0.013]. Multiplicative interaction of rs3783553 with HBV preS deletion significantly reduced the risk of HCC in males, with an adjusted OR of 0.64 (95% CI=0.42-0.98; P=0.041) after age and HBV genotype were adjusted. Although rs3783553 did not significantly affect genetic susceptibility to HBV-related HCC, its variant allele may predispose the host to selecting HBV C7A mutation during evolution and significantly reduce the risk of HCC caused by HBV preS deletion. This study provides an insight into the complex host-virus interaction in HBV-induced hepatocarcinogenesis and is helpful in determining HBsAg-positive subjects who are likely to develop HCC.

Predictive values of plasma TNFα and IL-8 for intracranial hemorrhage in patients with acute promyelocytic

摘要: In patients with acute promyelocytic leukemia (APL), intracranial hemorrhage (ICH), if not identified promptly, could be fatal. It is the leading cause of failure of induction and early death. Thus, biomarkers that could promptly predict severe complications are critical. Here, cytokine differences between patients with APL with and without ICH were investigated to develop predictive models for this complication. The initial cytokine profiling using plasma samples from 39 patients and 18 healthy donors found a series of cytokines that were remarkedly different between patients with APL and healthy controls. The APL patients were subsequently divided into high and low white blood cell count groups. Results showed that tumor necrosis factor α and interleukin 8 (IL-8) were vital in distinguishing patients with APL who did or did not develop ICH. In addition, verification in 81 patients with APL demonstrated that the two cytokines were positively correlated with the cumulative incidence of ICH. Finally, in-vitro and in-vivo experimental evidence were provided to show that IL-8 influenced the migration of APL-derived NB4 cells and impaired the blood–brain barrier in PML/RARα positive blast-transplanted FVB/NJ mice. These assessments may facilitate the early warning of ICH and reduce future mortality levels in APL.

Effect of IL-10 on formation of foam cell induced by ox-LDL

摘要: Atherosclerosis is a chronic disease that causes various cardiovascular complications. It has been realized that cellular and humoral immunity plays crucial roles in atherogenic lesion formation. In this study the effects of lipopolysaccharide (LPS) and interleukin-10 (IL-10) on the formation of foam cells during the early stages of atherosclerosis have been investigated. Macrophage was induced by phorbol myristate acetate (PMA) treatment on THP-1 cells. The cells were further stimulated by ox-LDL, ox-LDL plus LPS, ox-LDL plus IL-10 and LPS. By using an oil red O staining technique, the formation of foam cells was evaluated by lipid granules formation in the cells. The ratio of foam cell formation was increased from (9.77 ± 1.70)% to (16.27 ± 2.27)% after 24 h stimulation with ox-LDL, and the increase was observed with incubating time. The foam cells were significantly increased in the presence of LPS in a dose-dependent manner. The maximum increase of about 40% was observed. However, the significant elevation by LPS was abrogated when IL-10 was added. These results indicated that IL-10 can effectively prevent the formation of foam cells induced by ox-LDL with or without LPS. This study demonstrates that ox-LDL can cause foam cell formation from macrophages . LPS can significantly accelerate this event. IL-10, an anti-inflammatory cytokine, can inhibit the effect of ox-LDL and LPS. These results indicate that inflammatory effects in blood vessels can speed up foam cell formation. The inhibitive effect of IL-10 is an important factor for delaying atherosclerosis processes.

Emergence in cyber-physical systems: potential and risk

摘要: (CPSs) are distributed assemblages of computing, communicating, and physical components that sense their environment, algorithmically assess the incoming information, and affect their physical environment. Thus, they share a common structure with other , and therefore share both the possible benefits and the probable harmful effects of emergent phenomena. Emergence is an often unexpected pattern that arises from the interactions among the individual system components and the environment. In this paper we focus on three major problems concerning emergence in the context of CPSs: how to successfully exploit emergence, how to avoid its detrimental effects in a single CPS, and how to avoid harmful emergence that arises due to unexpected interaction among several independently developed CPSs that are operating in the same environment. We review the state of the research with regard to these problems and outline several approaches that could be used to address them.

Decomposition of aqueous chlorinated contaminants by UV irradiation with H

摘要: In the present study, the decomposition rates of carbon tetrachloride (CCl ) and 2,4-dichlorophenol (2,4-DCP) in water by the ultraviolet (UV) light irradiation alone and H O /UV were experimentally investigated. The detailed experimental studies have been conducted for examining treatment capacities of the two different ultraviolet light sources (low and medium pressure Hg arc) in H O /UV processes. The low or medium UV lamp alone resulted in a 60%–90% decomposition of 2,4-DCP while a slight addition of H O resulted in a drastic enhancement of the 2,4-DCP decomposition rate. The decomposition rate of 2,4-DCP with the medium pressure UV lamp alone was about 3–6 times greater than the low pressure UV lamp alone. In the direct photolysis of aqueous CCl , the medium pressure UV lamp had advantage over the low pressure UV lamp because the molar extinction coefficient of CCl at shorter wavelength (210–220 nm) is about 20 to 50 times higher than that at 254 nm. However, adding H O to the medium pressure UV lamp system rendered a negative oxidation rate because H O acted as a UV absorber being competitive with CCl due to negligible reaction between CCl and OH radicals. The results from the present study indicated significant influence of the photochemical properties of the target contaminants on the photochemical treatment characteristics for designing cost-effective UV-based degradation of toxic contaminants.

铰接车辆的主动转向控制策略研究 Article

摘要: 为了提高铰接车辆的机动性及高速稳定性,基于线性二次型最优控制理论提出了一种用于拖车和挂车的主动转向控制策略。首先建立了3自由度的带挂车主动转向的拖挂车简化模型。运用基于模拟退火的粒子群优化算法对简化模型在特定速度和方向盘角度下的车辆关键参数进行辨识,简化模型的参数可通过当前车辆工况查表和插值获得。仿真结果表明简化模型的输出结果与TruckSim一致性较好,这样简化模型可准确为控制器提供理想的横摆角速度。设计了基于线性二次型最优控制的主动转向控制器。控制器时刻跟踪理想的横摆角速度并同时尽量降低拖车和挂车的质心侧偏角。最后,基于TruckSim-Simulink联合仿真分别进行了低速和高速试验。仿真试验结果表明主动转向控制策略能显著改善拖挂车低速的机动性和高速稳定性。控制策略不仅适用于弯度不大的转弯路况,还适用于急转弯路况。

Cytokine storm and translating IL-6 biology into effective treatments for COVID-19

摘要: As of May 3, 2023, the coronavirus disease 2019 (COVID-19) pandemic has resulted in more than 760 million confirmed cases and over 6.9 million deaths. Several patients have developed pneumonia, which can deteriorate into acute respiratory distress syndrome. The primary etiology may be attributed to cytokine storm, which is triggered by the excessive release of proinflammatory cytokines and subsequently leads to immune dysregulation. Considering that high levels of interleukin-6 (IL-6) have been detected in several highly pathogenic coronavirus-infected diseases, such as severe acute respiratory syndrome in 2002, the Middle East respiratory syndrome in 2012, and COVID-19, the IL-6 pathway has emerged as a key in the pathogenesis of this hyperinflammatory state. Thus, we review the history of cytokine storm and the process of targeting IL-6 signaling to elucidate the pivotal role played by tocilizumab in combating COVID-19.

Effects of heavy rainfall on the composition of airborne bacterial communities

摘要: Wet deposition scavenges particles and particle-associated bacteria from the air column, but the impact of raindrops on various surfaces on Earth causes emission of surface-associated bacteria into the air column. Thus, after rainfall, these two mechanisms are expected to cause changes in airborne bacterial community composition (BCC). In this study, aerosol samples were collected at a suburban site in Seoul, Korea before and after three heavy rainfall events in April, May, and July 2011. BCC was investigated by pyrosequencing the 16S rRNA gene in aerosol samples. Interestingly, the relative abundance of non-spore forming operational taxonomic units (OTUs) was always higher in post-rain aerosol samples. In particular, the absolute and relative abundances of airborne always increased after rainfall, whereas those of airborne , including and , consistently decreased. Marine bacterial sequences, which were temporally important in aerosol samples, also decreased after rainfall events. Further, increases in pathogen-like sequences were often observed in post-rain air samples. Rainfall events seemed to affect airborne BCCs by the combined action of the two mechanisms, with potentially adverse effects on human and plant health.

Adsorption properties of polycarboxylate ether-based superplasticizer on cement particles and their resultant dispersion

摘要: The cement dispersion performance of a polycarboxylate (PCE)-based superplasticizer is highly related to their adsorption behaviors as a function of time. This study evaluated effects of PCEs on rheological properties of cementitious materials. First, characteristics of PCEs were characterized via permeation chromatography (GPC) and Fourier-transform infrared spectrometry (FT-IR). The adsorption behavior of single and blended PCEs on cementitious composites was identified using total organic carbon analyzer (TOC). Based on the measurement of PCE adsorption, the changes of rheological properties of cementitious materials as well as the number of dispersed cement particles were characterized using a rheometer and laser spectroscopy, respectively. The experimental results support the systematic mechanism of PCE adsorption, cement dispersion, and the decrease in viscosity of cementitious materials.

Bavachin enhances NLRP3 inflammasome activation induced by ATP or nigericin and causes idiosyncratic hepatotoxicity

摘要: Psoraleae Fructus (PF) is a well-known traditional herbal medicine in China, and it is widely used for osteoporosis, vitiligo, and other diseases in clinical settings. However, liver injury caused by PF and its preparations has been frequently reported in recent years. Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury (IDILI), but the mechanism underlying its hepatotoxicity remains unclear. This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity. Bavachin boosts the secretion of IL-1β and caspase-1 caused by ATP or nigericin but not those induced by poly(I:C), monosodium urate crystal, or intracellular lipopolysaccharide. Bavachin does not affect AIM2 or NLRC4 inflammasome activation. Mechanistically, bavachin specifically increases the production of nigericin-induced mitochondrial reactive oxygen species among the most important upstream events in the activation of the NLRP3 inflammasome. Bavachin increases the levels of aspartate transaminase and alanine aminotransferase in serum and hepatocyte injury accompanied by the secretion of IL-1β via a mouse model of lipopolysaccharide-mediated susceptibility to IDILI. These results suggest that bavachin specifically enhances the ATP- or nigericin-induced activation of the NLRP3 inflammasome. Bavachin also potentially contributes to PF-induced idiosyncratic hepatotoxicity. Moreover, bavachin and PF should be evaded among patients with diseases linked to the ATP- or nigericin-mediated activation of the NLRP3 inflammasome, which may be a dangerous factor for liver injury.

采用背景人声简化特征集的说话人识别直方图均衡化方法 Article

摘要: 本文提出了一种用于说话人识别技术的直方图均衡化方法。该方法采用了一套增补简化特征集,用以在训练数据和测试数据较短时改进说话人识别的效果。该增补特征集采用选择算法或聚类算法从背景人声中派生得到。当输入语音数据样本不足时,本文提出的方法可作为构建直方图的特征归一化方法使用。另外,该方法作为一种i-vector归一化方法,源于一种目前较为先进的基于i-vector的概率线性判别分析(Probabilistic linear discriminant analysis, PLDA)说话人识别系统。在输入语音和增补集中,用于直方图均衡化的样本值序号均按升序进行估计。新的序列号则按不同种类的序号之和进行排列。随后,该方法采用最新的序列号得出了测试语音样本的累积分布函数。本文将这一方法与倒谱均值归一化(Cepstral mean normalization, CMN)方法、倒谱均值和方差归一化(Cepstral mean and variance normalization, MVN)方法、直方图均衡化(Histogram equalization, HEQ)方法和欧洲电信标准协会模拟前端方法进行了比较。此外,在一具体算例中将该方法性能与采用模糊C-means和K-means算法的贪婪选择算法进行了比较。采用YOHO和ETRI数据库对特征空间进行评估。测试集采用Opus VoIP编码器进行了模拟。本文还采用了2008美国国家标准技术研究所说话人识别评测语料库对该i-vector系统进行了评测。试验结果表明,与传统特征归一化方法相比,当采用所提出的方法时,平均系统性能可得到有效提提升。

Th17 Cells in autoimmune diseases

摘要:

Th17 cells are a new subset of CD4+ T cells involved in the clearance of extracellular pathogens and fungi. Accumulating evidence suggests that Th17 cells and their signature cytokines have a pivotal role in the pathogenesis of multiple autoimmune-mediated inflammatory diseases. Here, we summarize recent research progress on Th17 function in the development and pathogenesis of autoimmune diseases. We also propose to identify new small molecule compounds to manipulate Th17 function for potential therapeutic application to treat human autoimmune diseases, including rheumatoid arthritis, systemic lupus erythematosus, Sj?gren’s syndrome, inflammatory bowel disease, and multiple sclerosis.

灰色预测模型GM()的适用性分析及在火灾风险预测中的应用

Metabolic interventions combined with CTLA-4 and PD-1/PD-L1 blockade for the treatment of tumors: mechanisms

摘要: Immunotherapies based on immune checkpoint blockade (ICB) have significantly improved patient outcomes and offered new approaches to cancer therapy over the past decade. To date, immune checkpoint inhibitors (ICIs) of CTLA-4 and PD-1/PD-L1 represent the main class of immunotherapy. Blockade of CTLA-4 and PD-1/PD-L1 has shown remarkable efficacy in several specific types of cancers, however, a large subset of refractory patients presents poor responsiveness to ICB therapy; and the underlying mechanism remains elusive. Recently, numerous studies have revealed that metabolic reprogramming of tumor cells restrains immune responses by remodeling the tumor microenvironment (TME) with various products of metabolism, and combination therapies involving metabolic inhibitors and ICIs provide new approaches to cancer therapy. Nevertheless, a systematic summary is lacking regarding the manner by which different targetable metabolic pathways regulate immune checkpoints to overcome ICI resistance. Here, we demonstrate the generalized mechanism of targeting cancer metabolism at three crucial immune checkpoints (CTLA-4, PD-1, and PD-L1) to influence ICB therapy and propose potential combined immunotherapeutic strategies co-targeting tumor metabolic pathways and immune checkpoints.
