The critical role of autophagy in plant responses to abiotic stresses

摘要: Autophagy is an evolutionary conserved recycling process in eukaryotes whereby intracellular components are engulfed by autophagosomes, which are subsequently transferred to the vacuoles for further degradation and reuse. In organisms like yeast and metazoans, autophagy is actively engaged during environmental perturbation either by degrading denatured proteins and organelles or by interfacing with stress related signaling molecules. Studies over the last decade have also revealed numerous important mechanisms where autophagy is widely involved in plant abiotic stress responses. Autophagy serves as a pivotal route for nutrient remobilization by the degradation of superfluous or damaged cellular cytoplasmic material and organelles. It is also reported to regulate the accumulation of reactive oxygen species, to maintain the cellular redox balance of plants under stressful conditions. Furthermore, autophagy is essential in regulating cellular toxicity by removing aggregated and/or denatured proteins and thereby improving plant stress tolerance. In this review, recent advances in our understanding of autophagy, along with pathways and regulatory networks through which it influences many aspects of plant growth and development in response to nutrient starvation, oxidative stress, osmotic stress and extreme temperatures are discussed.

SUPEROXIDE DISMUTASE FAMILY GENES IN WATERMELON AND THEIR RESPONSES TO DIFFERENT ABIOTIC STRESSES

Superoxide dismutase (SOD) is an important enzyme in the antioxidant system of plants and plays a vital role in stress responses by maintaining the dynamic balance of reactive oxygen species (ROS) concentrations. Genome-wide analysis of the SOD gene family in various plant species has been conducted but little is known about this gene family in watermelon (Citrullus lanatus). Here, eight SOD genes were identified in the watermelon genome and are designated ClCSD1-5ClFSD1-2 and ClMSD according to their metal cofactors. Phylogenetic analysis shows that SOD proteins from various plant species can be classified into five groups and members in the same group possess the same metal cofactor and similar subcellular localizations. Expression analysis of the ClSOD genes indicates that they had tissue-specific expression patterns with high expression in different tissues including the leaves, flowers and fruit. In addition, the expression of ClSOD genes differed appreciably under salinity, drought and abscisic acid (ABA) treatments, indicating that they may be involved in ROS scavenging under different abiotic stresses via an ABA-dependent signaling pathway. These results lay the foundation for elucidating the function of ClSOD genes in stress tolerance and fruit development in watermelon.

摘要: β-amylase (BAM) is an important enzyme involved in conversion of starch to maltose in multiple biological processes in plants. However, there is currently insufficient information on the BAM gene family in the important fruit crop banana. This study identified 16 genes in the banana genome. Phylogenetic analysis showed that MaBAMs were classified into four subfamilies. Most in each subfamily shared similar gene structures. Conserved motif analysis showed that all identified MaBAM proteins had the typical glyco hydro 14 domains. Comprehensive transcriptomic analysis of two banana genotypes revealed the expression patterns of in different tissues, at various stages of fruit development and ripening, and in responses to abiotic stresses. Most s showed strong transcript accumulation changes during fruit development and late-stage ripening. Some showed significant changes under cold, salt, and osmotic stresses. This finding indicated that might be involved in regulating fruit development, ripening, and responses to abiotic stress. Analysis of five hormone-related and seven stress-relevant elements in the promoters of further revealed that BAMs participated in various biological processes. This systemic analysis provides new insights into the transcriptional characteristics of the genes in banana and may serve as a basis for further functional studies of such genes.

An Implantable and Self-Powered Sensing System for the In Vivo Monitoring of Dynamic H2O2 Level in Plants

摘要: The real-time monitoring of hydrogen peroxide (H2O2) is significant for understanding the working mechanism of signal molecules, breeding for stress tolerance, and diagnosing plant health. However, it remains a challenge to realize real-time monitoring of the dynamic H2O2 level in plants. Here, we report an implantable and self-powered sensing system for the continuous monitoring of H2O2 level in plants. A photovoltaic (PV) module is integrated into a sensing system to collect sunlight or artificial light in the planting environment in order to continuously power an implantable microsensor. The transmission process of the H2O2 signal was monitored and analyzed in vivo, and the time and concentration specificity of the H2O2 signal for abiotic stress were resolved. This implantable system provides a promising analysis tool for key signal molecules in plants and might be extended to the real-time monitoring of signaling molecules in other crops.

Abiotic association of phthalic acid esters with humic acid of a sludge landfill

摘要: The abiotic association between phthalic acid esters (PAEs) and humic substances (HS) in sludge landfill plays an important role in the fate and stability of PAEs. An equilibrium dialysis combined with C-labeling was used to study the abiotic association of two abundant PAEs (diethyl phthalate and di- -butyl phthalate) with humic acid (HA) isolated from a sludge landfill with different stabilization times and different molecular weights. Elemental analysis and Fourier Transform Infrared Spectrophotometer (FTIR) suggested that high value of HA was related to the high aromatic content and large molecular weight of HA. The results indicated that the association strength of PAEs with HA depended on both the properties of the PAEs and the characteristics of HA. The values of the association were strongly dependent on solution pH, and decreased dramatically as the pH was increased from 3.0 to 9.0. The results suggested that non-specific hydrophobic interaction between PAEs and HA was the main contributor to the association of the PAEs with HA. The interactive hydrogen-bonds between the HA and the PAEs molecules may also be involved in the association.

Comparison of evapotranspiration and energy partitioning related to main biotic and abiotic controllers

Knowledge of evapotranspiration (ET) and energy partitioning is useful for optimizing water management, especially in areas where water is scarce. A study was undertaken in a furrow-irrigated vineyard (2015) and a drip-irrigated vineyard (2017) in an arid region of north-west China to compare vineyard ET and energy partitioning and their responses to soil water content (SWC) and leaf area index (LAI). ET and soil evaporation (E) and transpiration (T) were determined using eddy covariance, microlysimeters, and sap flow. Seasonal average E/ET, T/ET, crop coefficient (Kc), evaporation coefficient (Ke), and basal crop coefficient (Kcb) were 0.50, 0.50, 0.67, 0.35, and 0.29, respectively, in the furrow-irrigated vineyard and 0.42, 0.58, 0.57, 0.29, and 0.43 in the drip-irrigated vineyard. The seasonal average partitioning of net radiation (Rn) into the latent heat flux (LE), sensible heat flux (H) and soil heat flux (G) (LE/Rn, H/Rn, and G/Rn), evaporative fraction (EF) and Bowen ratio (β) were 0.57, 0.26, 0.17, 0.69 and 0.63, respectively, in the furrow-irrigated vineyard and 0.46, 0.36, 0.17, 0.57 and 0.97 in the drip-irrigated vineyard. The LE/Rn, H/Rn, EF, and β were linearly correlated with LAI. The E, Kc, Ke, E/ET, LE/Rn, LEs/Rn (ratio of LE by soil E to Rn), H/Rn, EF and β were closely correlated with topsoil SWC (10 cm depth). Responses of ET and energy partitioning to the LAI and SWC differed under the two irrigation methods. Drip irrigation reduced seasonal average E/ET and increased average T/ET. From the perspective of energy partitioning, seasonal average H/Rn increased whereas LE/Rn, especially LEs/Rn, decreased. Compared with furrow irrigation, drip irrigation decreased the proportion of unproductive water consumption thereby contributing to enhanced water use efficiency and accumulation of dry matter.

Magnetostriction is investigated to evaluate the stress relief caused by pulsed magnetic field treatment, because this physical property is closely associated with residual stress. Magnetostriction of different stressed samples is measured in this paper. The stress variations caused by pulsed magnetic treatment are also compared. It is found that magnetostriction variations are closely associated with stress changes. Thermodynamic potential is used to find the relationship between them. Based on several assumptions, we find that the product of magnetostriction amplitude and stress magnitude is nearly a constant during magnetic field treatment, which is valuable for stress relief evaluation and optimizing processing parameters. This conclusion is testified by stress measurements, and the calculated values are in accordance with the experiment results.

Elastic modulus and thermal stress in coating during heat cycling with different substrate shapes

The elastic modulus of a deposit (Ed) can be obtained by monitoring the temperature (?T) and curvature (?k) of a one-side coated long plate, namely, a one-dimensional (1D) deformation model. The aim of this research is to design an experimental setup that proves whether a 1D deformation model can be scaled for complex geometries. The setup includes a laser displacement sensor mounted on a robotic arm capable of scanning a specimen surface and measuring its deformation. The reproducibility of the results is verified by comparing the present results with Stony Brook University Laboratory’s results. The ?k-?T slope error is less than 8%, and the Ed estimation error is close to 2%. These values reveal the repeatability of the experiments. Several samples fabricated with aluminum as the substrate and 100MXC nanowire (Fe and Cr alloy) as the deposit are analyzed and compared with those in finite element (FE) simulations. The linear elastic behavior of 1D (flat long plate) and 2D (squared plate) specimens during heating/cooling cycles is demonstrated by the high linearity of all ?k-?T curves (over 97%). The Ed values are approximately equal for 1D and 2D analyses, with a median of 96 GPa and standard deviation of 2 GPa. The correspondence between the experimental and simulated results for the 1D and 2D specimens reveals that deformation and thermal stress in coated specimens can be predicted regardless of specimen geometry through FE modeling and by using the experimental value of Ed. An example of a turbine-blade-shaped substrate is presented to validate the approach.

Parametric equations for notch stress concentration factors of rib–deck welds under bending loading

摘要: The effective notch stress approach for evaluating the fatigue strength of rib–deck welds requires notch stress concentration factors obtained from complex finite element analysis. To improve the efficiency of the approach, the notch stress concentration factors for three typical fatigue-cracking modes (i.e., root–toe, root–deck, and toe–deck cracking modes) were thoroughly investigated in this study. First, we developed a model for investigating the effective notch stress in rib–deck welds. Then, we performed a parametric analysis to investigate the effects of multiple geometric parameters of a rib–deck weld on the notch stress concentration factors. On this basis, the multiple linear stepwise regression analysis was performed to obtain the optimal regression functions for predicting the notch stress concentration factors. Finally, we employed the proposed formulas in a case study. The notch stress concentration factors estimated from the developed formulas show agree well with the finite element analysis results. The results of the case study demonstrate the feasibility and reliability of the proposed formulas. It also shows that the fatigue design curve of FAT225 seems to be conservative for evaluating the fatigue strength of rib–deck welds.

Analysis on annealing-induced stress of blind-via TSV using FEM

Copper-filled through silicon via (TSV) is a promising material owing to its application in high-density three-dimensional (3D) packaging. However, in TSV manufacturing, thermo-mechanical stress is induced during the annealing process, often causing reliability issues. In this paper, the finite element method is employed to investigate the impacts of via shape and SiO2 liner uniformity on the thermo-mechanical properties of copper-filled blind-via TSV after annealing. Top interface stress analysis on the TSV structure shows that the curvature of via openings releases stress concentration that leads to ~60 MPa decrease of normal stresses, sxx and syy, in copper and ~70 MPa decrease sxx of in silicon. Meanwhile, the vertical interface analysis shows that annealing-induced stress at the SiO2/Si interface depends heavily on SiO2 uniformity. By increasing the thickness of SiO2 linear, the stress at the vertical interface can be significantly reduced. Thus, process optimization to reduce the annealing-induced stress becomes feasible. The results of this study help us gain a better understanding of the thermo-mechanical behavior of the annealed TSV in 3D packaging.

The formation mechanism and the influence factor of residual stress in machining

Residual stresses generated in cutting process have important influences on workpiece performance. The paper presents a method of theoretical analysis in order to explicate the formation mechanism of residual stresses in cutting. An important conclusion is drawn that the accumulated plastic strain is the main factor which determines the nature and the magnitude of surface residual stresses in the workpiece. On the basis of the analytical model for residual stress, a series of simulations for residual stress prediction during cutting AISI 1045 steel are implemented in order to obtain the influences of cutting speed, depth of cut and tool edge radius on surface residual stress in the workpiece. And these influences are explained from the perspective of formation mechanism of residual stress in cutting. The conclusions have good applicability and can be used to guide the parameters selection in actual production.

An investigation on stress distribution effect on multi- piezoelectric energy harvesters

摘要: With the fast development of piezoelectric materials and due to its green and renewable characteristics, the piezoelectric energy harvesting technology has been paid more and more attention by pavement engineers. The stress distribution will significantly affect the piezoelectric material performance. In this paper, the effects of multiple piezoelectric elements on the generation of electrical energy and output power are studied. In the case of constant external load, the number of the piezoelectric units does not necessarily produce more energy. When the same multi piezoelectric units work together, if the stress state of the piezoelectric units is different, the total output energy affected by the connection mode. For uneven stress distribution, the optimal output mode is that each of the piezoelectric units rectified before connected in parallel.

摘要:

针对石化特种设备中硫化氢(H2S)应力腐蚀问题现状,综合比较了国际权威机构NACE,EFC以及ISO发布的最新标准、规范,对H2S应力腐蚀的环境界定、试验方法、影响因素、应力腐蚀评价准则及对材料的要求等做了扼要的综述。同时对焊接残余应力的产生、测定方法、消除措施等进行了论述,列举了实际参与研究的发生应力腐蚀容器的实例及残余应力的影响。
