Functional ferritin nanoparticles for biomedical applications

摘要: Ferritin, a major iron storage protein with a hollow interior cavity, has been reported recently to play many important roles in biomedical and bioengineering applications. Owing to the unique architecture and surface properties, ferritin nanoparticles offer favorable characteristics and can be either genetically or chemically modified to impart functionalities to their surfaces, and therapeutics or probes can be encapsulated in their interiors by controlled and reversible assembly/disassembly. There has been an outburst of interest regarding the employment of functional ferritin nanoparticles in nanomedicine. This review will highlight the recent advances in ferritin nanoparticles for drug delivery, bioassay, and molecular imaging with a particular focus on their biomedical applications.

Evaluation of soil microbial toxicity of waste foundry sand for soil-related reuse

摘要: The relationship between the chemical contaminants and soil microbial toxicity of waste foundry sand (WFS) was investigated. Five different types of WFS from typical ferrous, aluminum, and steel foundries in China were examined for total metals, leachable metals, and organic contaminants. The soil microbial toxicity of each WFS was evaluated by measuring the dehydrogenase activity (DHA) of a blended soil and WFS mixture and then comparing it to that of unblended soil. The results show that the five WFSs had very different compositions of metal and organic contaminants and thus exhibited very different levels of soil microbial inhibition when blended with soil. For a given WFS blended with soil in the range of 10 wt.%–50 wt.% WFS, the DHA decreased almost linearly with increased blending ratio. Furthermore, for a given blending ratio, the WFSs with higher concentrations of metal and organic contaminants exhibited greater microbial toxicity. Correlation analysis shows that the relationship between ecotoxicity and metal and organic contaminants of WFSs can be described by an empirical logarithmic linear model. This model may be used to control WFS blending ratios in soil-related applications based on chemical analysis results to prevent significant inhibition of soil microbial activity.

Improvement of the assimilable organic carbon (AOC) analytical method for reclaimed water

摘要: Microbial growth is an issue of concern that may cause hygienic and aesthetic problems during the transportation and usage of reclaimed water. Assimilable organic carbon (AOC) is an important parameter which determines the heterotrophic bacterial growth potential of water. P17 and sp. NOX are widely used to measure AOC in drinking water. The AOC values of various reclaimed water samples determined by P17 and NOX were compared with those determined by the new strains isolated from reclaimed water in this study. It showed that the conventional test strains were not suitable for AOC measurement of reclaimed water in certain cases. In addition to P17 and NOX, sp. ZJ2, G3 and sp. G6, were selected as test strains for AOC measurement of reclaimed water. Key aspects of the bioassay including inoculum cell density, incubation temperature, incubation time and the pH of samples were evaluated for the newly selected test strains. Higher inoculum density (10 CFU·mL ) and higher incubation temperature (25°C) could reduce the time required for the tests. The AOC results of various collected samples showed the advantages of the method proposed based on those five strains in evaluating the biologic stability of reclaimed water.

Evaluation of endocrine disruption and dioxin-like effects of organic extracts from sewage sludge in autumn in Beijing, China

摘要: Study on effective disposal and utilization of sewage sludge has recently been the target of growing interest in China. However, potential risks are associated with the use of sludge due to its contamination with toxic organics, heavy metals and pathogenic microorganisms. In this study, a screening assessment was conducted on sewage sludge samples collected from 17 different sewage treatment plants in Beijing, based on a batch of in vitro bioassays, including a set of recombinant gene yeast assays for endocrine disruption, and an ethoxy resorufin-O-deethylase (EROD) assay using H4IIE cells for aryl hydrocarbon receptor (Ah-R) agonistic activities. Our results suggested that moderate levels of estrogen receptor agonistic activities (0.9 ng E2·g to 6.8 ng E2·g , dw), but relative higher androgen receptor antagonistic activities (nd to 45%), progestin receptor antagonistic activities (nd to 80%) and Ah-R agonistic activities (1390 to 6740 pg 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)·g , dw) were found in sewage sludge samples. However, there were no significant correlations between the toxic effects of sewage sludge and the sewage treatment processes. In addition, the 17β-estradiol (E2) equivalent quantity (EEQ) level of the sewage sludge was increased after the composting treatment, whereas the 2,3,7,8-tetrachlorodibenzo- -dioxin toxic equivalent quantity (TEQ) level of sewage sludge composted was much lower than that of sewage sludge.
