HPV prevalence and genotyping in the cervix of Chinese women

摘要: The role of human papillomavirus (HPV) as an etiologic factor of cervical cancer has been firmly established, and prophylactic vaccines are now available and have been approved in many countries. Vaccination implies a promising future for cervical cancer prevention especially for countries with very limited access to screening. However, the vaccines are not accessible in mainland China at the moment, and much needs to be understood about the potential benefit when HPV vaccines are applied to Chinese women, and to make the vaccines more specific to Chinese women. This article reviews advanced multi-center, hospital/population-based studies of most recent years, and aims to draw a definitive conclusion on HPV prevalence and genotyping in cervical cancer and precancerous lesions in China from the aspect of study population, geographic areas and time period.

Using RAPD in genotyping and transmission detection

摘要: The aim of this paper is to develop an applicable Random Amplified Polymorphic DNAs (RAPD) method for genotyping strain, and discuss the possibility of using the RAPD method to trace strain transmission route. Four different pretreatment methods were used on the genomic DNA component, and the best adaptive extract method was selected for RAPD. Different RAPD primers sequence was used for amplification and their differentiating capabilities for strains were compared. Applicable RAPD primer was selected for genotyping and then applied into transmission detection. The results show that the so called cetyltrimethylammonium bromide (CTAB) method for extracting genomic DNA could give integrated genomic DNA and give out relatively better RAPD fingerprint maps, subsequently, using selected RAPD primer could give out a group of amplification polymerase chain reaction bands. The fingerprint maps from different strains were distinctive. Some main segments were common to all the strains tested. Some segments were different among the strains. According to the fingerprint maps and similarity index of different isolates, isolates from a pair of sex-partners were very similar. Based on these findings, the best extracting method and suitable RAPD primer were chosen. The RAPD fingerprint maps could type effectively and could be used as an additional approach in molecular epidemiology for tracing infection sources.

摘要: Efficient evaluation of crop phenotypes is a prerequisite for breeding, cultivar adoption, genomics and phenomics study. Plant genotyping is developing rapidly through the use of high-throughput sequencing techniques, while plant phenotyping has lagged far behind and it has become the rate-limiting factor in genetics, large-scale breeding and development of new cultivars. In this paper, we consider crop phenotyping technology under three categories. The first is high-throughput phenotyping techniques in controlled environments such as greenhouses or specifically designed platforms. The second is a phenotypic strengthening test in semi-controlled environments, especially for traits that are difficult to be tested in multi-environment trials (MET), such as lodging, drought and disease resistance. The third is MET in uncontrolled environments, in which crop plants are managed according to farmer’s cultural practices. Research and application of these phenotyping techniques are reviewed and methods for MET improvement proposed.
