Impact and inhibitory mechanism of phenolic compounds on the formation of toxic Maillard reaction products

摘要:

As one of the dominant reactions occurring during thermal treatment of food, the Maillard reaction not only leads to the formation of aroma, browning color and taste compounds, but also contributes to the formation of some unpleasant toxic substances including acrylamide, heterocyclic amines and advanced glycation end products. Polyphenols, one of the most abundant antioxidants in the human diet, are contained in different kinds of foods. In this review, some recent studies on the impact of dietary polyphenols on the formation of acrylamide, heterocyclic amines and advanced glycation end products formed during the Maillard reaction are summarized, including the research work conducted with both chemical model systems and real food model systems; the possible inhibitory mechanisms of different polyphenols are also summarized and discussed in this review. Basically we found that some dietary polyphenols not only scavenge free radicals, but also react with reactive carbonyl species, thus lowering the formation of toxic Maillard reaction products. This review provides a useful theoretical foundation for the application of polyphenols in food safety, and suggests some directions for further study of natural products as inhibitors against the formation of toxic substances in thermally processed food.

Design of a cyclic inhibitory CPG controller for the locomotion of a snakelike robot

摘要: The rhythmic locomotion of a creature is a self-excitation behavior of the CPG (central pattern generator), which makes it supremely adapted for environment. Based on this fact, firstly, a snake-ike robot controller with cyclic inhibitory CPG model was designed, and then the stability of a single neuron, CPG model and the NON ( neuron oscillator network) was analyzed. By implementing this control architecture to a simulator based on the mechanical dynamics of a real snake-like robot named Perambulator-I, we presented preliminary rules for parameter setting of the CPG controller to modulate the number of S shapes, the curve of the body shape, locomotion velocity, and the curve of the locomotion trajectory for serpentine locomotion. Moreover, we demonstrated that Perambulator-I can successfully exhibit serpentine locomotion by using the output of the proposed CPG controller. The results of this paper provide a realistic approach for designing an artificial CPG controller.

Design, synthesis and HIV-RT inhibitory activity of novel thiazolidin-4-one derivatives

摘要: A series of 2-aryl-3-(4,5,6-trimethylpyrimidin-2-yl) thiazolidin-4-ones ( – ) and their derivatives bearing a lipophilic substituent, like acetoxy group ( – ), propionyloxy group ( – ), methyl ( and ) at C-5 on thiazolidin-4-one ring were designed, synthesized and evaluated for their HIV-RT inhibitory activity. Using self-catalyzed Pummerer reaction, compounds – and – were obtained in good yield (63.1% 75.2%). Preliminary anti-HIV-RT test of these derivatives indicated that compounds – , (propionyloxy group at C-5) showed moderate HIV-RT inhibitory activity and compounds and with methyl at C-5 showed a weak HIV-RT inhibitory activity. Structure activity relationship analysis suggested that the substituted groups on C-5 would be unfavorable to anti-HIV-RT activity and that the steric effect might play a critical role in the anti-HIV RT activity.

Algal-bloom control by allelopathy of aquatic macrophytes-A review

摘要: Algal-bloom control is an important issue for water environment protection as it induces several negative impacts on the lives of aquatic organisms, aquaculture, landscaping, and human health. The development of an environment-friendly, cost-effective, and convenient alternative for controlling algal bloom has gained much concern. Using the allelopathy of aquatic macrophytes as a novel and safe method for algal-bloom control is a promising alternative. This paper reviews the development and potential application about allelopathy of aquatic plants on algae, including the allelopathic research history, the potential research problems, the research methodology, and the reported aquatic macrophytes and their inhibitory allelochemicals. Potential modes of inhibition action of allelochemicals on algae, possible ways for application, and future development directions of research on algal-bloom control by aquatic macrophytes were also presented.

Allelopathic inhibition on red tide microalgae by five macroalgal extracts

摘要: This study aims to identify effective antialgal allelochemicals from marine macroalgae that inhibit the growth of red tide microalgae. Practically, new algicidal agents were developed to control red tide. The growth inhibitory effects of 5 marine macroalgae , , , , and on were evaluated by adding crude seawater extracts of macroalgal dry tissue into the culture medium containing . The half-effective concentrations at 120 h (EC) of the seawater extracts were 0.6, 0.9, 1.0, 1.0, and 4.7 g/L for the five macroalgae above, respectively. , and showed strong allelopathic effect on the growth of . There have been no previous reports with regard to the allelopathic effects of the former two macroalgae so far. The possible allelochemicals of 21 compounds of the were detected using Gas chromatography-mass spectrometry (GC-MS) analysis. Inhibitory activity of Bifidobacterium adolescent combined with cisplatin on melanoma in mice and its

摘要: The aim of this study is to explore inhibitory activity of Bifidobacterium adolescent combined with cisplatin on the growth of melanoma (B16) in mice and the underlying mechanism. C57 mice were inoculated with B16 cancer cells to construct mouse model of melanoma and treated with bifidobacterium adolescent combined with cisplatin. Ratios of inhibitory activity on the growth of melanoma (B16) were calculated. Pathology changes of the tumor were observed by HE staining. B16 cell cycles were examined on a flow cytometer. Lymphocyte proliferation was measured with MTT assay and the T-cell subset was measured by double marked fluorescence. When bifidobacterium of 10 cfu/L was injected, the ratio of inhibitory activity on the growth of melanoma (B16) reached 54%, which was similar to that of cisplatin group. The ratio of inhibitory activity reached 74.45% when the mice were treated by bifidobacterium combined with cisplatin. HE staining shows that bifidobacterium inhibited B16 cell proliferation and enhanced the cisplatins killing activity on B16 cells. The results of flow cytometry demonstrated that B16 cell proliferation was arrested at G stage after treatment with bifidobacterium. The B16 cell proliferation was arrested at S stage after treatment with cisplatin. The CD4+ percentage increased and the difference was significant compared with the normal group after treatment with bifidobacterium, indicating that T-cell immune activity was enhanced. Treatment with bifidobacterium combined with cisplatin can enhance the inhibitory activity on the growth of melanoma (B16) of cisplatin. The mechanism of the inhibitory activity on B16 cell proliferation is correlated with the enhanced immune activity in mice.

摘要: The fish ( ) was used as the source for probiotics. 46 bacterial isolates were obtained from the skin, gills, guts and intestines of the guppy, (collected from a government model fish farm in Kottayam, India). Of the above isolated strains, four isolates were selected based on their inhibitory spectrum against five indicator strains, 1739, 3906, 2495, 1271 and 1424 (standard cultures collected from Microbial Type Culture Collection (MTCC) Chandigarh, India). Among the resulting isolates, two were gram-positive cocci, namely MBTU-PB2 and MBTU-PB3 and belong to the genus . The other two were gram-negative rods, namely MBTU-PB1 and MBTU-PB4, of the genera and , respectively. The basic probiotic characteristics of these isolates such as the production of bacteriocin like inhibitory substances (BLIS), antibiotic sensitivities and growth profiles were also determined. The above four isolated strains exhibited different antagonisms than the five indicator strains. During incubation, the antibacterial activity gradually increased in the inhibition zone and was influenced by the lag period ( ) and doubling time. The lag periods for most of the four selected strains were shorter than those of the indicator strains and the isolates had different growth rates ( ) than the indicator strains. All four isolates produced BLIS, however, the strains had different BLIS activities against the indicator strains. Treatment of the neutralized cell free supernatants of the selected isolates with proteases eliminated or reduced the BLIS activity, suggesting a proteinaceous nature of the inhibitory compounds. Further, the optimum BLIS activity was observed at neutral pH after 18 h of incubation. The antibiotic sensitivity assay revealed that the isolates were susceptible to routinely used antibiotics, whereas the plasmid profiles showed that the plasmids had no role in the antagonistic properties of the four isolated strains. The results showed that the isolates could be a promising source for biocontrol agents in aquacultures.

Toxic effects of wastewater from various phases of monosodium glutamate production on seed germination and root elongation of crops

摘要: To make a comprehensive assessment on monosodium glutamate (MSG) wastewater pollution, a pollution exposure experiment was carried out on the seed germination and root elongation of wheat, Chinese cabbage and tomato by using the wastewater discharged from different processing phases of MSG production. The results showed that there were significantly positive linear relationships between the inhibitory rates of wheat seed germination and root elongation and the COD of the mother liquor scraps. The toxicity of MSG wastewater to the test crops was in the order of tomato > Chinese cabbage > wheat, indicating that tomato was the most sensitive to the wastewater, and could be considered as an ideal toxic bioindicator. The half-effect concentrations (IC) based on the seed germination and root elongation of the test crops exposed to the wastewater discharged from various processing phases of MSG production was 22.0 32 432 and 17.3 3320 mg/L, respectively.

Influences and mechanisms of nanofullerene on the horizontal transfer of plasmid-encoded antibiotic resistance genes between

摘要: Abstract • Sub-inhibitory levels of nC60 promote conjugative transfer of ARGs. • nC60 can induce ROS generation, oxidative stress and SOS response. • nC60 can increase cell membrane permeability and alter gene expression. • Results provide evidence of nC60 promoting antibiotic resistance dissemination. The spread and development of antibiotic resistance globally have led to severe public health problems. It has been shown that some non-antibiotic substances can also promote the diffusion and spread of antibiotic resistance genes (ARGs). Nanofullerene (nC60) is a type of nanomaterial widely used around the world, and some studies have discovered both the biological toxicity and environmental toxicity of nC60. In this study, cellular and molecular biology techniques were employed to investigate the influences of nC60 at sub-minimum inhibitory concentrations (sub-MICs) on the conjugation of ARGs between the E. coli strains. Compared with the control group, nC60 significantly increased the conjugation rates of ARGs by 1.32‒10.82 folds within the concentration range of 7.03‒1800 mg/L. This study further explored the mechanism of this phenomenon, finding that sub-MICs of nC60 could induce the production of reactive oxygen species (ROS), trigger SOS-response and oxidative stress, affect the expression of outer membrane proteins (OMPs) genes, increase membrane permeability, and thus promote the occurrence of conjugation. This research enriches our understanding of the environmental toxicity of nC60, raises our risk awareness toward nC60, and may promote the more rational employment of nC60 materials.

Highly Aromatic Norditerpenoid Heterodimers and Monomers from Trigonostemon fragilis

摘要: Four new norditerpenoid heterodimers with different dimerization patterns—namely, trigofragiloids A–C (denoted as compounds 1–3) and (+)- and (−)-trigofragiloid D (compound 4)—and three new phenanthrenone norditerpenoids—namely, trigofragiloids E–G (compounds 5–7)—were isolated from Trigonostemon fragilis. Compounds 1 and 2 feature a novel heterodimeric carbon skeleton formed by the conjugation of a tetra-norditerpenoid and an ennea-norditerpenoid; they have been identified as class 2 atropisomers by means of quantum chemical calculations. Compound 3 is an unprecedented phenylpropanoid-norditerpenoid adduct with a new dimerization pattern. Compounds (+)- and (−)-4 are the first example of S-shaped 1,4-dioxane-fused norditerpenoid dimers. Inspired by the structure elucidation of compound 4, two co-occurring analogues, actephilol A and epiactephilol A, were structurally revised as a pair of geometrical isomers and were identified as two pairs of enantiomers, (+)- and (−)-8 and (+)- and (−)-9, respectively. Their structures were characterized using a combined method. Notably, compound 7 exhibits remarkable adenosine triphosphate-citrate lyase (ACLY) inhibition with a half-maximal inhibition concentration (IC50) value of (0.46 ± 0.11) μmol∙L−1, as active as the positive control BMS-303141, and a molecular docking study offers deep insight into the interaction between compound 7 and ACLY.

Effects of phosphatidylinositol 3-kinase inhibitor on human cervical carcinoma cells

摘要: Phosphatidylinositol 3-kinase (PI3K) is a crucial cell survival pathway implicated in tumorigenesis because of its role in stimulating cell proliferation and suppressing apoptosis. This study was to investigate the regulation of proliferation and apoptosis by LY294002, an inhibitor of PI3K in cervical cancer cells and the expression of FLICE-like inhibitory protein (c-FLIP) . Human cervical cancer HeLa cells were used in this experiment and cultured. The cultured cells were treated with LY294002 at different concentrations (10, 25, 50 and 100µmol/L) for 6, 12, 24, and 48h before harvesting for evaluation. Cell viability was measured by 3-(4,5)-dimethylthiazol(-2-y1)-3,5-di-phenyltetrazoliumbromide (MTT) assay. Apoptosis was analyzed by flow cytometry. The expression of c-FLIP was detected by Western blot. Cell viability was inhibited by LY294002 significantly (<0.05). Flow cytometry analysis revealed that cell apoptosis was significantly increased in the presence of LY294002 as compared with the control group. Although the expression of c-FLIP was increased in a short time, the expression of c-FLIP was markedly suppressed after the treatment of LY294002 for 48h. These results suggested that the PI3K/Akt signal pathway might be involved in the regulation of cell apoptosis in cervical cancer cells. Moreover, the regulation of c-FLIP expression through PI3K/Akt signal pathway in cervical cancer cells was observed .
