Involvement of p38 mitogen-activated protein kinase in the regulation of platelet-derived growth factor

摘要: The aim of this study was to investigate the role of p38 mitogen-activated protein kinase (MAPK) in cell migration induced by platelet-derived growth factor (PDGF). Western blot was performed to detect the phosphorylation of p38 in NIH3T3 cells treated with PDGF. A Transwell cell migration system was used to determine the effects of PDGF treatment on the migration of NIH3T3 cells and the influence of deficiency on this process in a gene knockout (p38) mouse embryonic fibroblast cell line. On the stimulation of PDGF, the migration of NIH3T3 cells was significantly increased (〈0.001) compared to the control and p38 MAP kinase was simultaneously phosphorylated. Furthermore, the PDGF-induced cell migration was significantly blocked in gene knockout (p38) mouse embryonic fibroblasts (MEFs) (〈0.001) as compared with the wild type cells (p38). p38 MAPK plays an important role in the regulation of cell migration induced by PDGF.

摘要: Mitogen-activated protein kinase (MAPK) signaling pathway, one of the most important signaling pathways in eukaryotic organism, is involved in multiple cellular events such as cell growth, differentiation, and apoptosis. MAPK is of great importance to the normal function of organisms, while its dysfunction results in various diseases. So far, inhibitors specifically against each subfamilies of MAP kinase have been developed, while more endeavors are needed to discover the compounds selectively targeting a particular subfamily member. Most of the kinase inhibitors exert their functions in an ATP-competitive way or a non-ATP-competitive way. Further studies on the effective mechanism of the MAPK inhibitors and their therapeutic roles in the treatment of diseases are helpful for the illumination of MAP kinase function, the development of novel inhibitors, and the therapy of diseases caused by the dysfunction of the MAPK pathway.

Human menstrual blood-derived stem cells alleviate autoimmune hepatitis via JNK/MAPK signaling pathway and

摘要: Autoimmune hepatitis (AIH) is a severe globally distributed liver disease that could occur at any age. Human menstrual blood-derived stem cells (MenSCs) have shown therapeutic effect in acute lung injury and liver failure. However, their role in the curative effect of AIH remains unclear. Here, a classic AIH mouse model was constructed through intravenous injection with concanavalin A (Con A). MenSCs were intravenously injected while Con A injection in the treatment groups. The results showed that the mortality by Con A injection was significantly decreased by MenSCs treatment and liver function tests and histological analysis were also ameliorated. The results of phosphoproteomic analysis and RNA-seq revealed that MenSCs improved AIH, mainly by apoptosis and c-Jun N-terminal kinase/mitogen-activated protein signaling pathways. Apoptosis analysis demonstrated that the protein expression of cleaved caspase 3 was increased by Con A injection and reduced by MenSCs transplantation, consistent with the TUNEL staining results. An AML12 co-culture system and JNK inhibitor (SP600125) were used to verify the JNK/MAPK and apoptosis signaling pathways. These findings suggested that MenSCs could be a promising strategy for AIH.

Mechanisms of connective tissue formation and blocks of mitogen activated protein kinase

摘要: Ninety male Wistar rats were selected under the “Guide for the Care and Use of Laboratory Animals” for skin-muscle wound models. Three groups of animals were examined respectively for inoculation of inhibitor of p38 MAPK (mitogen activated protein kinase) SB 203580 and JNK inhibitor SP 600125, and a control. Light microscopy, immunohistochemistry, and tensometry revealed that the inhibition of p38 or JNK cascades have modified the formation of the connective tissue scar. The degree of connective tissue growth in the area of surgical wound had been significantly reduced by the end of observation (30 d) as the SB 203580 was applied (% volume of collagen 43.60 (41.05 – 60.15) 73.54 (66.87 – 78.01) in control, = 0.002). Conversely, when we have applied the JNK blocker, the density of collagen in scar tissue increased (78.14 (72.77 – 81.14), = 0.022 control). SB203580 inhibits the expression of p38, c-Jun and c-Fos. When we have used the JNK blocker, the expression of c-Fos and c-Jun decreased, but the expression of p38 increased. This determines the high functional activity of fibroblasts after using SP 600125. Obtained results show the importance of studying regulators of cell differentiation as potential drugs, which significantly affect the outcome of the pathological processes.

Adsorption of benzene and toluene from waste gas using activated carbon activated by ZnCl

摘要: A series of activated carbons with high surface area were prepared from walnut shell using chemical activation with ZnCl . In this research the carbonization stage was carried out at 500°C. The performance of the synthesized carbons evaluated in adsorption of benzene and toluene from waste gas. The influence of impregnation ratio on the characteristics of synthesized activated carbons as well as their adsorption capacity was investigated. The ratio of activation agent to walnut shell was selected in the range of 0.5–2.0 wt/wt. The synthesized activated carbons were characterized using XRD, SEM, BET and FTIR techniques. The highest activated carbon production yield was obtained at impregnation ratio of 1.5 wt/wt. The XRD analysis illustrated that peaks intensity decreased with increasing impregnation ratio showing that amorphous property of samples was increased. The SEM analysis revealed successful pore development in synthesized activated carbons obtained at high impregnation ratios. The surface area of the activated carbons increased with increasing impregnation ratio and its maximum value reached 2643 m ?g at impregnation ratio of 2/1. FTIR analysis indicated that the relative amount of different acidic surface groups on synthesized carbons was a function of impregnation ratio. Experimental results for benzene and toluene adsorption showed a high potential of employing synthesized impregnated activated carbon for treatment of waste gas. Generally, the amount of VOC adsorbed on the surface was affected by physicochemical properties of synthesized activated carbons.

Expression of protease-activated receptors on platelets in healthy individuals

摘要: This study aimed to investigate the expression of protease-activated receptors (PARs) on platelets in healthy individuals and preliminarily elucidate physiological functions of PARs. Thirty healthy volunteers, who did not take any anticoagulants and antiplatelet agents within 10 days before the examination, were recruited. Fasting venous blood (5 mL) was taken from the medial cubital vein in each individual and platelet-rich plasma (PRP) was prepared. The expression of PAR1 mRNA and PAR4 mRNA in PRP was determined by RT-PCR analysis. The results showed that the average levels of PAR1 mRNA and PAR4 mRNA on platelets in healthy individuals were 0.1601 ± 0.0269 and 0.1073 ± 0.0194 respectively. In combination with literature analysis, it was concluded that the thrombin signaling pathway plays a vital role in the development of hemostasis and thrombosis, and the selective PARs antagonist has the potential for extensive application in clinical practice.

Removal of phenol by powdered activated carbon adsorption

摘要: In this study, the adsorption performance of powdered activated carbon (PAC) on phenol was investigated in aqueous solutions. Batch adsorption studies were performed to evaluate the effects of various experimental parameters like PAC type, PAC dose, initial solution pH, temperature and pre-oxidation on the adsorption of phenol by PAC and establish the adsorption kinetics, thermodynamics and isothermal models. The results indicated that PAC adsorption is an effective method to remove phenol from water, and the effects of all the five factors on adsorption of phenol were significant. The adsorption rate of phenol by PAC was rapid, and more than 80% phenol could be absorbed by PAC within the initial 10 min. The adsorption process can be well described by pseudo-second-order adsorption kinetic model with rate constant amounted to 0.0313, 0.0305 and 0.0241 mg·μg ·min with coal, coconut shell and bamboo charcoal. The equilibrium data of phenol absorbed onto PAC were analyzed by Langmuir, Freundlich and Tempkin adsorption isotherms and Freundlich adsorption isotherm model gave the best correlation with the experimental data. Thermodynamic parameters such as the standard Gibbs free energy (? ), enthalpy (? ) and entropy (? ) obtained in this study indicated that the adsorption of phenol by PAC is spontaneous, exothermic and entropy decreasing.

Kinetics and thermodynamics of the phosphine adsorption on the modified activated carbon

摘要: The kinetics and the thermodynamics of phosphine (PH ) adsorption on the modified activated carbon have been explained for the adsorption process of PH . This study investigated the kinetic and thermodynamic properties of PH adsorption on the activated carbon impregnated with 5% HCl solution. The thermodynamic properties including PH adsorption isotherm and adsorption heat were separately investigated at 20°C, 70°C, 90°C. The results showed that the Freundlich-type isotherm equation described the isotherms well. The adsorption capacity increased with increasing temperature between 20°C and 70°C. Between 70°C and 90°C, the adsorption capacity decreased obviously with increasing temperature. The adsorption capacity reached the maximum at 70°C. By analyzing the results of the kinetics and the thermodynamics, we found that the adsorption of PH was dominated by physical adsorption at the lower temperature (20°C). Then with increasing temperature, chemical adsorption gradually dominated in the adsorption process. The adsorption capacity decreased at above 70°C is due to the exothermic effects in the process of adsorption.

A hybrid membrane process for simultaneous thickening and digestion of waste activated sludge

摘要: A hybrid membrane process for simultaneous sludge thickening and digestion (MSTD) was studied. During one cycle (15 d) of operation under a hydraulic retention time of 1 d, the concentration of mixed liquor suspended solids (MLSS) continuously increased from about 4 g·L to 34 g·L, and the mixed liquor volatile suspended solids (MLVSS) increased from about 3 g·L to over 22 g·L. About 42% of the MLVSS and 39% of the MLSS reduction were achieved. The thickening and digestion effects in the MSTD were further analyzed based on a mass balance analysis. Test results showed that biopolymers and cations of biomass were gradually released to the bulk solution during the process. It was also found that the capillary suction time, colloidal chemical oxygen demand, soluble microbial products, viscosity, and MLSS had significant positive correlations with the membrane fouling rate, whereas extracellular polymeric substances, polysaccharides, and proteins extracted from biomass had negative impacts on membrane fouling.

Permeability and thermal conductivity of host compressed natural graphite for consolidated activated

摘要: Permeability and thermal conductivity test units were set up to study the heat and mass transfer performance of the host material, i.e. expanded natural graphite (ENG), for consolidated activated carbon (AC) adsorbent. The permeability was tested with nitrogen as the gas source, and the thermal conductivity was studied using steady-state heat source method. The results showed that the values of permeability and thermal conductivity were 10 to 10 m and 1.7 to 3.2 W/(m·K), respectively, while the density compressed expanded natural graphite (CENG) varied from 100 to 500 kg/m . The permeability decreased with the increasing density of CENG, whereas the thermal conductivity increased with the increasing density of CENG. Then the thermal conductivity and permeability of granular AC were researched. It was discovered that the thermal conductivity of samples with different grain size almost kept constant at 0.36 W/(m·K) while the density was approximately 600 kg/m . This means that the thermal conductivity was not related to the grain size of AC. The thermal conductivity of CENG was improved by 5 to 10 times compared with that of granular AC. Such a result showed that CENG was a promising host material for AC to improve the heat transfer performance, while the mass transfer performance should be considered in different conditions for utilization of adsorbent.

Tuning porosity of coal-derived activated carbons for CO2 adsorption

摘要: A simple method was developed to tune the porosity of coal-derived activated carbons, which provided a model adsorbent system to investigate the volumetric CO2 adsorption performance. Specifically, the method involved the variation of the activation temperature in a K2CO3 induced chemical activation process which could yield activated carbons with defined microporous (< 2 nm, including ultra-microporous < 1 nm) and meso-micro-porous structures. CO2 adsorption isotherms revealed that the microporous activated carbon has the highest measured CO2 adsorption capacity (6.0 mmol∙g–1 at 0 °C and 4.1 mmol∙g–1 at 25 °C), whilst ultra-microporous activated carbon with a high packing density exhibited the highest normalized capacity with respect to packing volume (1.8 mmol∙cm−3 at 0 °C and 1.3 mmol∙cm–3 at 25 °C), which is significant. Both experimental correlation analysis and molecular dynamics simulation demonstrated that (i) volumetric CO2 adsorption capacity is directly proportional to the ultra-micropore volume, and (ii) an increase in micropore sizes is beneficial to improve the volumetric capacity, but may lead a low CO2 adsorption density and thus low pore space utilization efficiency. The adsorption experiments on the activated carbons established the criterion for designing CO2 adsorbents with high volumetric adsorption capacity.
