Stem cell niches and endogenous electric fields in tissue repair

Adult stem cells are responsible for homeostasis and repair of many tissues. Endogenous adult stem cells reside in certain regions of organs, known as the stem cell niche, which is recognized to have an important role in regulating tissue maintenance and repair. In wound healing and tissue repair, stem cells are mobilized and recruited to the site of wound, and participate in the repair process. Many regulatory factors are involved in the stem cell-based repair process, including stem cell niches and endogenous wound electric fields, which are present at wound tissues and proved to be important in guiding wound healing. Here we briefly review the role of stem cell niches and endogenous electric fields in tissue repair, and hypothesize that endogenous electric fields become part of stem cell niche in the wound site.

心血管疾病是全世界主要致死原因之一。人们对新的干预性治疗手段的需求与日俱增。虽然药物和手术治疗极大地改善了心血管疾病患者的生活质量,但人们还是需要价格更便宜、副作用更小的治疗手段。天然和合成生物材料无论是作为给药载体,还是替代支架的细胞外基质,在心脏修复和再生中都展现出巨大的潜力。本文探讨了目前治疗心血管疾病的几种方式和应用于上述疾病干预性治疗的生物材料;着重研究了导电聚合物在纠正局部缺血性心脏病引发的传导异常及其在心脏起搏器中应用的可能性,以改善心肌梗死状态下的传导路径。
Generation and repair of AID-initiated DNA lesions in B lymphocytes

Activation-induced deaminase (AID) initiates the secondary antibody diversification process in B lymphocytes. In mammalian B cells, this process includes somatic hypermutation (SHM) and class switch recombination (CSR), both of which require AID. AID induces U:G mismatch lesions in DNA that are subsequently converted into point mutations or DNA double stranded breaks during SHM/CSR. In a physiological context, AID targets immunoglobulin (Ig) loci to mediate SHM/CSR. However, recent studies reveal genome-wide access of AID to numerous non-Ig loci. Thus, AID poses a threat to the genome of B cells if AID-initiated DNA lesions cannot be properly repaired. In this review, we focus on the molecular mechanisms that regulate the specificity of AID targeting and the repair pathways responsible for processing AID-initiated DNA lesions.

关节软骨是位于关节骨表面的一种无血管且富有弹性的结缔组织。软骨损伤常出现于年轻人和经常从事体育活动的人的膝盖中,由于缺乏合适的基于组织工程的人工基质,目前针对软骨损伤的治疗,特别是对于全层软骨缺损和骨-软骨界面的治疗方式,依旧无法充分替代或再生受损的软骨。随着关节软骨组织工程研发的飞速进展,功能化水凝胶因其自身良好的生物力学性能、含水量、膨胀系数、细胞相容性、生物可降解性和润滑能力,而逐渐涌现成为富有前景的软骨基质替代物。功能化水凝胶可以被合理的设计和根据其理化性能被便捷的调整来模拟软骨的胞外基质。本综述先简要介绍了关节软骨的组成、结构、功能以及关节软骨损伤,然后全面回顾了功能化水凝胶用于关节软骨修复的各种精细的(生物)设计和(生物)制造方法。最后,我们总结了基于功能化水凝胶的关节软骨组织工程策略在体内外实验中所遭遇到的一些困难和挑战,同时为未来临床转化指明了方向。
Progress and perspectives of neural tissue engineering

Traumatic injuries to the nervous system lead to a common clinical problem with a quite high incidence and affect the patient’s quality of life. Based on a major challenge not yet addressed by current therapeutic interventions for these diseases, a novel promising field of neural tissue engineering has emerged, grown, and attracted increasing interest. This review provides a brief summary of the recent progress in the field, especially in combination with the research experience of the author’s group. Several important aspects related to tissue engineered nerves, including the theory on their construction, translation into the clinic, improvements in fabrication technologies, and the formation of a regenerative environment, are delineated and discussed. Furthermore, potential research directions for the future development of neural tissue engineering are suggested.
Bile duct injury repair — earlier is not better

Bile duct injury is a common complication of cholecystectomy. The timing of bile duct injury repair remains controversial. A recent review conducted in France reported 39% complications and 64% failure after immediate repair in 194 patients compared with 14% complications and 8% failure after late repair in 133 patients. A national review of 139 consecutive early repairs conducted at five hepatopancreaticobiliary centers in Denmark reported 4% mortality, 36% morbidity, and 42 restrictures (30%) at a median follow-up of 102 months, and only 64 patients (46%) demonstrated uneventful short-term and long-term outcomes. Most patients with bile duct injury present with bile leak and sepsis; thus, early repair is not recommended. Percutaneous drainage of bile and endoscopic stenting are the mainstays of treatment of bile leak because they convert acute bile duct injury into a controlled external biliary fistula. The ensuing benign biliary stricture should be repaired by a biliary surgeon after a delay of 4–6 weeks once the external biliary fistula has closed.
