NES1/KLK10 and hNIS gene therapy enhanced iodine-131 internal radiation in PC3 proliferation inhibition

摘要: gene is thought to be a tumor-suppressor gene. Our previous study found that overexpression of gene in PC3 cell line could slow down the tumor proliferation rate, associated with a mild decrease in expression. The decrease could increase the sensitivity of radiotherapy to tumors. Thus, we supposed to have an “enhanced firepower” effect by combining overexpressed gene therapy and I radiation therapy uptake by overexpressed hNIS protein. We found a weak endogenous expression of hNIS protein in PC3 cells and demonstrated that the low expression of hNIS protein in PC3 cells might be the reason for the low iodine uptake. By overexpressing in PC3, the radioactive iodine uptake ability was significantly increased. Results of and tumor proliferation experiments and F-fluorothymidine ( F-FLT) micro-positron emission tomography/computed tomography (micro-PET/CT) imaging showed that the combined gene therapy and I radiation therapy mediated by overexpressed hNIS protein had the best tumor proliferative inhibition effect. Immunohistochemistry showed an obvious decrease of expression and the lowest expression. These data suggest that via inhibition of expression, overexpressed might enhance the effect of radiation therapy of I uptake in overexpressed PC3 cells.

Disabled homolog 2 is required for migration and invasion of prostate cancer cells

摘要:

Disabled homolog 2 (DAB2) is frequently deleted or epigenetically silenced in many human cancer cells. Therefore, DAB2 has always been regarded as a tumor suppressor gene. However, the role of DAB2 in tumor progression and metastasis remains unclear. In this study, DAB2 expression was upregulated along with human prostate cancer (PCa) progression. DAB2overexpression or knockdown effects in LNCaP and PC3 cell lines were verified to address the biological functions of DAB2 in PCa progression and metastasis. LNCaP and PC3 cell lines were generated from human PCa cells with low and high metastatic potentials, respectively. The results showed that DAB2 shRNA knockdown can inhibit the migratory and invasive abilities of PC3 cells, as well as the tumorigenicity, whereas DAB2 overexpression enhanced LNCaP cell migration and invasion. Further investigation showed that DAB2 regulated the cell migration associated genes in PC3 cells, and the differential DAB2 expression between LNCaP and PC3 cells was partly regulated by histone 4 acetylation. Therefore, DAB2 may play an important role in PCa progression and metastasis.

Human microbiome and prostate cancer development: current insights into the prevention and treatment

摘要: The huge communities of microorganisms that symbiotically colonize humans are recognized as significant players in health and disease. The human microbiome may influence prostate cancer development. To date, several studies have focused on the effect of prostate infections as well as the composition of the human microbiome in relation to prostate cancer risk. Current studies suggest that the microbiota of men with prostate cancer significantly differs from that of healthy men, demonstrating that certain bacteria could be associated with cancer development as well as altered responses to treatment. In healthy individuals, the microbiome plays a crucial role in the maintenance of homeostasis of body metabolism. Dysbiosis may contribute to the emergence of health problems, including malignancy through affecting systemic immune responses and creating systemic inflammation, and changing serum hormone levels. In this review, we discuss recent data about how the microbes colonizing different parts of the human body including urinary tract, gastrointestinal tract, oral cavity, and skin might affect the risk of developing prostate cancer. Furthermore, we discuss strategies to target the microbiome for risk assessment, prevention, and treatment of prostate cancer.

Detection and monitoring prostate specific antigen using nanotechnology approaches to biosensing

摘要: Prostate cancer has a high incidence in men and remains the second cause of mortality due to cancer. As the development of the disease is greatly correlated to age, the identification of novel detection methods reliable, efficient, and cost effective is a matter of significant importance in the ageing population of western societies. The detection of the prostate specific antigen (PSA) in blood samples has been the preferred method for the detection and monitoring of prostate cancer over the past decades. Despite the indications against its use in massive population screening, PSA still remains the best studied biomarker for prostate cancer and the detection of its different forms and incorporation in multiplexed designs with other biomarkers still remains a highly valuable indicator in the theranostics of prostate cancer. The latest developments in the use of nanomaterials towards the construction of PSA biosensors are reviewed hereby. The incorporation of gold nanoparticles, silica nanoparticles and graphene nanostructures to biosensing devices has represented a big leap forward in terms of sensitivity, stability and miniaturization. Both electrochemical and optical detection methods will be reviewed herein for the detection of PSA.

Effects of miR-200c on the migration and invasion abilities of human prostate cancer Du145 cells and

microRNAs (miRNAs) have played a key role in human tumorigenesis, tumor progression, and metastasis. On the one hand, miRNAs are aberrantly expressed in many types of human cancer; on the other hand, miRNAs can function as tumor suppressors or oncogenes that target many cancer-related genes. This study aimed to investigate the effects of miRNA-200c (miR-200c) on the biological behavior and mechanism of proliferation, migration, and invasion in the prostate cancer cell line Du145. In this study, Du145 cells were transfected with miR-200c mimics or negative control miR-NC by using an X-tremeGENE siRNA transfection reagent. The relative expression of miR-200c was measured by RT-PCR. The proliferation, migration, and invasion abilities of Du145 cells were detected by CCK8 assays, migration assays and invasion assays, respectively. The expressions of ZEB1, E-cadherin, and vimentin were observed by western blot. Results showed that DU145 cells exhibited a high expression of miR-200c compared with immortalized normal prostate epithelial cell RWPE-1. Du145 cells were then transfected with miR-200c mimics and displayed lower abilities of proliferation, migration, and invasion than those transfected with the negative control. The protein levels of ZEB1 and vimentin were expressed at a low extent in Du145 cells, which were transfected with miR-200c mimics; by contrast, E-cadherin was highly expressed. Hence, miR-200c could significantly inhibit the proliferation of the prostate cancer cell line Du145; likewise, miR-200c could inhibit migration and invasion by epithelial-mesenchymal transition.

Neoadjuvant radiohormonal therapy for oligo-metastatic prostate cancer: safety and efficacy outcomes

摘要: To evaluate the safety and efficacy of neoadjuvant radiohormonal therapy for oligometastatic prostate cancer (OMPC), we conducted a 3 + 3 dose escalation, prospective, phase I/II, single-arm clinical trial (CHiCTR1900025743), in which long-term neoadjuvant androgen deprivation was adopted 1 month before radiotherapy, comprising intensity modulated radiotherapy to the pelvis, and stereotactic body radiation therapy to all extra-pelvic bone metastases for 4‒7 weeks, at 39.6, 45, 50.4, and 54 Gy. Robotic-assisted radical prostatectomy was performed after 5‒14 weeks. The primary outcome was treatment-related toxicities and adverse events; secondary outcomes were radiological treatment response, positive surgical margin (pSM), postoperative prostate-specific antigen (PSA), pathological down-grading and tumor regression grade, and survival parameters. Twelve patients were recruited from March 2019 to February 2020, aging 66.2 years in average (range, 52‒80). Median baseline PSA was 62.0 ng/mL. All underwent RARP successfully without open conversions. Ten patients recorded pathological tumor down-staging (83.3%), and 5 (41.7%) with cN1 recorded negative regional lymph nodes on final pathology. 66.7% (8/12) recorded tumor regression grading (TRG) –I and 25% (3/12) recorded TRG-II. Median follow-up was 16.5 months. Mean radiological progression-free survival (RPFS) was 21.3 months, with 2-year RPFS of 83.3%. In all, neoadjuvant radiohormonal therapy is well tolerated for oligometastatic prostate cancer.

Ultrasound-guided prostate percutaneous intervention robot system and calibration by informative particle

摘要: Applying a robot system in ultrasound-guided percutaneous intervention is an effective approach for prostate cancer diagnosis and treatment. The limited space for robot manipulation restricts structure volume and motion. In this paper, an 8-degree-of-freedom robot system is proposed for ultrasound probe manipulation, needle positioning, and needle insertion. A novel parallel structure is employed in the robot system for space saving, structural rigidity, and collision avoidance. The particle swarm optimization method based on informative value is proposed for kinematic parameter identification to calibrate the parallel structure accurately. The method identifies parameters in the modified kinematic model stepwise according to parameter discernibility. Verification experiments prove that the robot system can realize motions needed in targeting. By applying the calibration method, a reasonable, reliable forward kinematic model is built, and the average errors can be limited to 0.963 and 1.846 mm for insertion point and target point, respectively.

酮基-脱氧壬酮糖酸(Kdn)是哺乳动物中相当少见的唾液酸。然而,已经发现游离或偶联Kdn浓度升高与人类癌症进展之间存在关联。迄今为止,还缺乏确凿的证据表明Kdn发生在(特定的)人类糖蛋白(偶联的Kdn)上。本文首次报道了Kdn 在来自人类精浆和尿液的前列腺特异性抗原(PSA)N-连接聚糖上表达。有趣的是,仅在触角半乳糖α2,3-连接结构中发现了Kdn,表明Kdn具有高度特异性的生物合成。在与前列腺癌(PCa)相关的尿PSA队列中也发现了这种不寻常的糖基化特征,尽管在PCa和非PCa患者之间没有发现差异。需要进一步研究游离Kdn和偶联Kdn在人类中的发生、生物合成、生物作用和生物标志物的潜力。

This work is devoted to the problem of planning of freight railway transportation. We define a special conflict graph on the basis of a set of acceptable train routes. The investigation aims to solve the classical combinatorial optimization problem in relation to the maximum independent set of vertices in undirected graphs. The level representation of the graph and its tree are introduced. With use of these constructions, the lower and upper bounds for the number of vertices in the maximum independent set are obtained.

Independent cover meshless particle method for complex geotechnical engineering

摘要: A new Independent Cover Meshless Particle (ICMP) method is proposed for the analysis of complex geotechnical engineering. In the ICMP method, the independent rectangular cover regardless of the shape of the analysis model is employed as the influence domain of each discrete node, the general polynomial is employed as the meshless interpolation function of the independent nodal cover, and the Cartesian Transformation Method (CTM) is used for the numerical integration of the nodal covers cut by material interfaces, joints, cracks and faults. The present method has a simple formulation and a low computational cost, and is easy for the numerical analysis and modeling of complex geotechnical engineering. Several typical numerical examples are presented to demonstrate the accuracy and robustness of the proposed method.

Cancer stem cell-immune cell crosstalk in the tumor microenvironment for liver cancer progression

摘要: Crosstalk between cancer cells and the immune microenvironment is determinant for liver cancer progression. A tumor subpopulation called liver cancer stem cells (CSCs) significantly accounts for the initiation, metastasis, therapeutic resistance, and recurrence of liver cancer. Emerging evidence demonstrates that the interaction between liver CSCs and immune cells plays a crucial role in shaping an immunosuppressive microenvironment and determining immunotherapy responses. This review sheds light on the bidirectional crosstalk between liver CSCs and immune cells for liver cancer progression, as well as the underlying molecular mechanisms after presenting an overview of liver CSCs characteristic and their microenvironment. Finally, we discuss the potential application of liver CSCs-targeted immunotherapy for liver cancer treatment.

Topology-independent end-to-end learning model for improving the voltage profile in microgrids-integrated

摘要: With multiple microgrids (MGs) integrated into power distribution networks in a distributed manner, the penetration of renewable energy like photovoltaic (PV) power generation surges. However, the operation of power distribution networks is challenged by the issues of multiple power flow directions and voltage security. Accordingly, an efficient voltage control strategy is needed to ensure voltage security against ever-changing operating conditions, especially when the network topology information is absent or inaccurate. In this paper, we propose a novel data-driven voltage profile improvement model, denoted as system-wide composite adaptive network (SCAN), which depends on operational data instead of network topology details in the context of power distribution networks integrated with multiple MGs. Unlike existing studies that realize topology identification and decision-making optimization in sequence, the proposed end-to-end model determines the optimal voltage control decisions in one shot. More specifically, the proposed model consists of four modules, Pre-training Network and modified interior point methods with adversarial networks (Modified IPMAN) as core modules, and discriminator generative adversarial network (Dis-GAN) and Volt convolutional neural network (Volt-CNN) as ancillary modules. In particular, the generator in SCAN is trained by the core modules in sequence so as to form an end-to-end mode from data to decision. Numerical experiments based on IEEE 33-bus and 123-bus systems have validated the effectiveness and efficiency of the proposed method.
