Exploring the coupling relationship of industrial agglomeration and low-carbon economy considering spatiotemporaldifferentiation: An empirical study in China’s construction machinery industry

摘要: Although China’s construction machinery thrives to meet the needs of construction, a number of challenges still remain to be overcome, such as lack of thorough knowledge of regional disparities and several limitations in terms of carbon emissions and economic development. Meanwhile, a low-carbon economy was proposed and implemented in China. This research aims to investigate the differences in industrial agglomeration of construction machineries and further explore the relationship between industrial agglomeration and low-carbon economy. On this basis, spatiotemporal analysis was performed to evaluate the levels of industrial agglomeration in different regions based on the situations of China’s construction machinery industry. Furthermore, this study explored the interaction between industrial agglomeration and low-carbon economy utilizing the coupling coordination analysis method. Results showed that the coupling coordination of the two subsystems was extremely unbalanced in 2006, and it maintained an increasing trend, reaching a relatively high level in 2018. Finally, suggestions, such as establishing a policy guarantee system and implementing variable policies in different regions, were proposed to provide guidelines for the government decision-making and promote the sustainable development of China’s construction machinery industry.

Monocyte subsets and their differentiation tendency after burn injury

摘要:

Monocytes are critical effectors and regulators of immune response. Studying the nomenclature of monocyte subsets may be beneficial for understanding the complex function of monocytes in steady and inflammatory states. A monocyte has the potential to differentiate into dendritic cells or macrophages, and this behavior significantly changes in severely burned patients and mice. The findings in the present study may help enhance understanding on the perturbation of the immune system after severe burn injury.

GID complex regulates the differentiation of neural stem cells by destabilizing TET2

摘要: Brain development requires a delicate balance between self-renewal and differentiation in neural stem cells (NSC), which rely on the precise regulation of gene expression. Ten-eleven translocation 2 (TET2) modulates gene expression by the hydroxymethylation of 5-methylcytosine in DNA as an important epigenetic factor and participates in the neuronal differentiation. Yet, the regulation of TET2 in the process of neuronal differentiation remains unknown. Here, the protein level of TET2 was reduced by the ubiquitin-proteasome pathway during NSC differentiation, in contrast to mRNA level. We identified that TET2 physically interacts with the core subunits of the glucose-induced degradation-deficient (GID) ubiquitin ligase complex, an evolutionarily conserved ubiquitin ligase complex and is ubiquitinated by itself. The protein levels of GID complex subunits increased reciprocally with TET2 level upon NSC differentiation. The silencing of the core subunits of the GID complex, including WDR26 and ARMC8, attenuated the ubiquitination and degradation of TET2, increased the global 5-hydroxymethylcytosine levels, and promoted the differentiation of the NSC. TET2 level increased in the brain of the Wdr26+/− mice. Our results illustrated that the GID complex negatively regulates TET2 protein stability, further modulates NSC differentiation, and represents a novel regulatory mechanism involved in brain development.

Abivertinib inhibits megakaryocyte differentiation and platelet biogenesis

摘要: Abivertinib, a third-generation tyrosine kinase inhibitor, is originally designed to target epidermal growth factor receptor (EGFR)-activating mutations. Previous studies have shown that abivertinib has promising antitumor activity and a well-tolerated safety profile in patients with non-small-cell lung cancer. However, abivertinib also exhibited high inhibitory activity against Bruton’s tyrosine kinase and Janus kinase 3. Given that these kinases play some roles in the progression of megakaryopoiesis, we speculate that abivertinib can affect megakaryocyte (MK) differentiation and platelet biogenesis. We treated cord blood CD34+ hematopoietic stem cells, Meg-01 cells, and C57BL/6 mice with abivertinib and observed megakaryopoiesis to determine the biological effect of abivertinib on MK differentiation and platelet biogenesis. Our in vitro results showed that abivertinib impaired the CFU-MK formation, proliferation of CD34+ HSC-derived MK progenitor cells, and differentiation and functions of MKs and inhibited Meg-01-derived MK differentiation. These results suggested that megakaryopoiesis was inhibited by abivertinib. We also demonstrated in vivo that abivertinib decreased the number of MKs in bone marrow and platelet counts in mice, which suggested that thrombopoiesis was also inhibited. Thus, these preclinical data collectively suggested that abivertinib could inhibit MK differentiation and platelet biogenesis and might be an agent for thrombocythemia.

A dynamic simulation analysis of spatiotemporal heterogeneity

摘要: There is notable variability in carbon emission reduction efforts across different provinces in China, underscoring the need for effective strategies to implement carbon emission allowance auctions. These auctions, as opposed to free allocations, could be more aligned with the principle of “polluter pays.” Focusing on three diverse regions — Ningxia, Beijing, and Zhejiang — this study employs a system dynamics simulation model to explore markets for carbon emissions and green certificates trading. The aim is to determine the optimal timing and appropriate policy intensities for auction introduction. Key findings include: (1) Optimal auction strategies differ among the provinces, recommending immediate implementation in Beijing, followed by Ningxia and Zhejiang. (2) In Ningxia, there’s a potential for a 6.20% increase in GDP alongside a 21.59% reduction in carbon emissions, suggesting a feasible harmony between environmental and economic objectives. (3) Market-related policy variables, such as total carbon allowances and Renewable Portfolio Standards, significantly influence the optimal auction strategies but have minimal effect on carbon auction prices.

摘要: The secondary laticifer, a specific tissue in the secondary phloem of rubber tree, is differentiated from the vascular cambia. The number of the secondary laticifer in the trunk bark of rubber tree is positively correlated with rubber yield. Although jasmonates have been demonstrated to be crucial in the regulation of secondary laticifer differentiation, the mechanism for the jasmonate-induced secondary laticifer differentiation remains to be elucidated. By using an experimental morphological technique, the present study revealed that trichostatin A (TSA), an inhibitor of histone deacetylation, could induce the secondary laticifer differentiation in a concentration-dependent manner. The results suggest that histone acetylation is essential for the secondary laticifer differentiation in rubber tree.

Repression of CDKN2C caused by PML/RARα binding promotes the proliferation and differentiation block

Inappropriate cell proliferation during oncogenesis is often accompanied by inactivation of components involved in the cell cycle machinery. Here, we report that cyclin-dependent kinase inhibitor 2C (CDKN2C) as a member of the cyclin-dependent kinase inhibitors is a target of the PML/RARα oncofusion protein in leukemogenesis of acute promyelocytic leukemia (APL). We found that CDKN2C was markedly downregulated in APL blasts compared with normal promyelocytes. Chromatin immunoprecipitation combined with quantitative polymerase chain reaction demonstrated that PML/RARα directly bound to the CDKN2C promoter in the APL patient-derived cell line NB4. Luciferase assays indicated that PML/RARα inhibited the CDKN2C promoter activity in a dose-dependent manner. Furthermore, all-trans retinoic acid treatment induced CDKN2C expression by releasing the PML/RARα binding on chromatin in NB4 cells. Functional studies showed that ectopic expression of CDKN2C induced a cell cycle arrest at the G0/G1 phase and a partial differentiation in NB4 cells. Finally, the transcriptional regulation of CDKN2C was validated in primary APL patient samples. Collectively, this study highlights the importance of CDKN2C inactivation in the abnormal cell cycle progression and differentiation block of APL cells and may provide new insights into the study of pathogenesis and targeted therapy of APL.

未来产业颠覆性技术创新的时空现象学研究

未来产业是塑造新质生产力的关键方面,其发展高度依赖颠覆性技术创新突破;我国目前的基础研究与科技创新能力不显强健,核心症结在于未能从哲学根源出发把握本质规律。本文基于现象学本质还原与先验还原的分析方法,针对颠覆性技术创新瓶颈内涵的复杂哲学问题进行理论诊断,溯源技术哲学、科学哲学的脉络谱系,厘清现代技术与科学研究之间的辩证关系,将技术创新问题投影至本源时空间范畴。研究发现,传统动力学时空观坚持身心二元的哲学思想,将时空间视为超越任何参考系的绝对抽象概念,忽视身体与实践活动发挥的关键作用。为此,集成现代科学与时空现象学理论,推动“数据表征、知识流动、具身耦联”的时空间重构;响应未来产业战略需求与科学逻辑框架,建立“技性科学、智能科学、具身科学”的未来科学研究范式演进架构;融入系统科学思想方法,构建“工程应用、智能集成、具身协同”的颠覆性技术创新系统。进一步地,基于“哲学 – 科学 – 技术 – 产业”的四元现象结构、“新质 – 新知”工程创新战略,提出增强发展组织能力、建立和健全服务保障机制、提高企业技术创新能力等应对建议。

Analysis and Zonation of Freeze-Thaw Action in the Chinese Plateau Region Considering Spatiotemporal

摘要: Concerns about the durability of transportation infrastructure due to freeze-thaw (F-T) cycles are particularly significant in the Chinese plateau region, where concrete aging and performance deterioration pose substantial challenges. The current national standards for the frost resistance design of concrete structures are based predominantly on the coldest monthly average temperature and do not adequately address the comprehensive effects of the spatiotemporal variance, amplitude, and frequency of F-T cycles. To address this issue, this study introduced a spatiotemporal distribution model to analyze the long-term impact of F-T action on concrete structures by employing statistical analysis and spatial interpolation techniques. Cluster analysis was applied to create a nationwide zonation of F-T action level from data on the freezing temperature, temperature difference, and the number of F-T cycles. Furthermore, this study explored the similarity between natural environmental conditions and laboratory-accelerated tests using hydraulic pressure and cumulative damage theories. A visualization platform that incorporates tools for meteorological data queries, environmental characteristic analyses, and F–T action similarity calculations was designed. This research lays theoretical groundwork and provides technical guidance for assessing service life and enhancing the quantitative durability design of concrete structures in the Chinese plateau region.

一种基于非线性时空效应的个性化下一个兴趣点推荐方法

摘要: 下一个兴趣点(POI)推荐是基于位置的社交网络(LBSN)的一项重要任务,其目标是使用历史签到数据在特定情境下为用户推荐下一个兴趣点。现有研究将用户时空信息线性离散化,然后使用基于循环神经网络(RNN)的方法进行建模。但是这些研究忽略了时空信息对用户偏好的非线性影响以及用户轨迹和候选兴趣点之间的时空相关性。为解决这些问题,本文提出一种时空轨迹(STT)模型。该模型使用具有注意力机制的长短期记忆网络(LSTM)作为基本框架,并将时空信息以编码形式引入模型。在编码信息过程中,使用指数型衰减因子刻画用户兴趣随时间和距离的非线性漂移特性。此外,本文在目标召回过程中设计一个时空匹配模块,该模块通过测量用户历史轨迹与候选集之间的相关性来为用户筛选最有可能的下一个兴趣点。本文使用4个真实数据集评估STT模型性能。实验结果表明,本文所提方法的推荐效果比主流的推荐模型有显著提升。

Cannabidiol prevents depressive-like behaviors through the modulation of neural stem cell differentiation

摘要: Chronic stress impairs radial neural stem cell (rNSC) differentiation and adult hippocampal neurogenesis (AHN), whereas promoting AHN can increase stress resilience against depression. Therefore, investigating the mechanism of neural differentiation and AHN is of great importance for developing antidepressant drugs. The nonpsychoactive phytocannabinoid cannabidiol (CBD) has been shown to be effective against depression. However, whether CBD can modulate rNSC differentiation and hippocampal neurogenesis is unknown. Here, by using the chronic restraint stress (CRS) mouse model, we showed that hippocampal rNSCs mostly differentiated into astrocytes under stress conditions. Moreover, transcriptome analysis revealed that the FoxO signaling pathway was involved in the regulation of this process. The administration of CBD rescued depressive-like symptoms in CRS mice and prevented rNSCs overactivation and differentiation into astrocyte, which was partly mediated by the modulation of the FoxO signaling pathway. These results revealed a previously unknown neural mechanism for neural differentiation and AHN in depression and provided mechanistic insights into the antidepressive effects of CBD.

Industrial eco-efficiency and its spatial-temporal differentiation in China

摘要: The aim of this paper is to study the spatial-temporal differentiation of industrial eco-efficiency in China. Using methods based on the data envelopment analysis (DEA) model and exploratory spatial data analysis (ESDA) and data from 1985, 1995, 2005, and 2008 of 30 provinces in China, the spatial-temporal pattern changes in industrial eco-efficiency are discussed. The results show that: first, the patterns of industrial eco-efficiency are dominated by clustering of relatively low efficiency provinces; second, spatial relationships between the industrial eco-efficiencies of different provinces changed slightly throughout the period and the provinces persistently exhibit spatial concentration of relatively low industrial eco-efficiency; finally, there is an obvious trend in the polarization of industrial eco-efficiency, i.e., the higher level spatial units are concentrated in eastern China, and the lower level spatial units are mainly in western and central China.

Wastewater Surveillance Provides Spatiotemporal SARS-CoV-2 Infection Dynamics

摘要: Wastewater surveillance can leverage its wide coverage, population-based sampling, and high monitoring frequency to capture citywide pandemic trends independent of clinical surveillance. Here we conducted a 9-month daily wastewater surveillance for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) from 12 wastewater treatment plants (WWTPs), covering approximately 80% of the population, to monitor infection dynamics in Hong Kong. We found that the SARS-CoV-2 virus concentration in wastewater was correlated with the daily number of reported cases and reached two pandemic peaks three days earlier during the study period. In addition, two different methods were established to estimate the prevalence/incidence rates from wastewater measurements. The estimated results from wastewater were consistent with findings from two independent citywide clinical surveillance programmes (rapid antigen test (RAT) surveillance and serology surveillance), but higher than the cases number reported by the Centre for Health Protection (CHP) of Hong Kong. Moreover, the effective reproductive number (Rt) was estimated from wastewater measurements to reflect both citywide and regional transmission dynamics. Our findings demonstrate that large-scale intensive wastewater surveillance from WWTPs provides cost-effective and timely public health information, especially when the clinical surveillance is inadequate and costly. This approach also provides insights into pandemic dynamics at higher spatiotemporal resolutions, facilitating the formulation of effective control policies and targeted resource allocation.
