EXTRACTION AND EVALUATION OF EDIBLE OIL FROM SCHIZOCHYTRIUM SP. USING AN AQUEOUS ENZYMATIC METHOD

Abstract:

Schizochytrium sp., a marine microalga, is a potential source of edible oil due to its short growth cycle and rapid lipid accumulation, especially of docosahexaenoic acid. An approach to isolate edible microalgal oil from Schizochytrium sp. using aqueous enzymatic extraction (AEE) was developed. Parameters were optimized by single-factor experiments followed by Box-Behnken design. Proteases were effective in extracting oil. The maximum free oil recovery (49.7%±0.58%) and total oil recovery (68.1%±0.94%) were obtained under optimum conditions of liquid-to-solid ratio of 4.8:1, a 2.5% enzyme concentration of papain and an extraction time of 2.2 h. There was a significant difference (P<0.05) in polyunsaturated fatty acid composition between microalgal oil obtained by AEE and by Soxhlet extraction, with the former having superior physiochemical properties and higher concentrations of bioactive components including total phenolic compounds and total tocopherols. These findings indicate a potential application of AEE for extraction of oil from Schizochytrium sp.

Abstract: Canadian goldenrod ( ) is one of the most destructive invasive weeds in South-eastern China. To evaluate its potential application as dietary supplement in red swamp crayfish ( ), the antibacterial properties of aqueous and ethanol extracts of this plant against three major pathogenic bacteria in crayfish aquaculture were examined. Inhibition zone tests and determination of minimum inhibitory concentration revealed that the extracts had lower antibacterial activity than extracts from two traditional medicinal plants that possess antibacterial properties, garlic ( ) and cortex phellodendri ( ). However, they did exhibit greater antibacterial effects than extracts from another widely used medicinal plant, , and an aquatic weed, . Aqueous extracts of Canadian goldenrod gave greater inhibition than the ethanol extracts. Crayfish fed a diet with 2% these aqueous extracts exhibited significantly higher enzyme activity of alkaline phosphatase, catalase and phenoloxidase ( <0.05). Based on the results of this study, we conclude that aqueous extracts of Canadian goldenrod are highly promising for the development of new dietary supplement for use in crayfish aquaculture.

Protective effects of lignin fractions obtained from grape seeds against bisphenol AF neurotoxicity via antioxidative effects mediated by the Nrf2 pathway

Abstract: Lignin exhibits antioxidative and various other biological properties. However, its neuroprotection capability has rarely been studied. In this study, three types of lignin with different structures were prepared from grape seeds by using different isolation techniques. The antioxidative and neuroprotective effects of the lignin fractions were evaluated with the apoptosis model of murine neuroectodermal (NE-4C) neural stem cells stimulated with bisphenol AF. The results demonstrated that the half maximal inhibitory concentration for scavenging 2,2-diphenyl-1-picrylhydrazyl with water-soluble lignin (L-W, 58.19 μg·mL–1) was lower than those of lignin in the autohydrolyzed residue of grape seeds (84.27 μg·mL–1) and original lignin in grape seeds (99.44 μg·mL–1). BPAF exposure had negative effects on the reactive oxygen species, malondialdehyde content, and superoxide dismutase and glutathione peroxidase activities in NE-4C cells, which can be reversed by using the prepared lignin to reduce oxidative stress. An immunofluorescence assay demonstrated that grape seed lignin induced protective effects on BPAF-injured NE-4C cells via the nuclear factor erythroid 2-related Factor 2 pathway. In addition, correlational analyses showed that lignin (L-W) with lower molecular weights and noncondensed phenolic hydroxyl group content and higher contents of COOH groups effectively prevented cell apoptosis, scavenged reactive oxygen species, and ensured protection from nerve injury. This study demonstrated that grape seed lignin can be used as a neuroprotective agent and serves as a demonstration of active lignin production from grape seed waste.
