Safety issues of methylglyoxal and potential scavengers

Shiming LI, Siyu LIU, Chi-Tang HO

Frontiers of Agricultural Science and Engineering 2018, Volume 5, Issue 3,   Pages 312-320 doi: 10.15302/J-FASE-2017174

Abstract:

The health safety of methylglyoxal (MGO) has been recognized as a key issue owing to its ultra-high reactivity toward some key biomolecules such as amino acids, proteins, DNA, sulfhydryl- and basic nitrogen-containing compounds, including amino-bearing neurotransmitters. In this review, we have summarized the endo- and exogenous sources of MGO and its accumulation inside the body due to high intake, abnormal glucose metabolism and or malfunctioning glyoxalases, and review the debate concerning the adverse functionality of MGO ingested from foods. Higher than normal concentrations of MGO in the circulatory system and tissues have been found to be closely associated with the production of advanced glycation end products (AGEs), increased oxidative stress, elevated inflammation and RAGE (AGE receptors) activity, which subsequently progresses to a pathological stage of human health, such as diabetes complications, cancer, cardiovascular and degenerative diseases. Having illustrated the mechanisms of MGO trapping in vivo, we advocate the development of efficient and efficacious MGO scavengers, either assisting or enhancing the activity of endogenous glyoxalases to facilitate MGO removal, or providing phytochemicals and functional foods containing them, or pharmaceuticals to irreversibly bind MGO and thus form MGO-complexes that are cleared from the body.

Current advances for bone regeneration based on tissue engineering strategies

Rui Shi, Yuelong Huang, Chi Ma, Chengai Wu, Wei Tian

Frontiers of Medicine 2019, Volume 13, Issue 2,   Pages 160-188 doi: 10.1007/s11684-018-0629-9

Abstract: Bone tissue engineering (BTE) is a rapidly developing strategy for repairing critical-sized bone defects to address the unmet need for bone augmentation and skeletal repair. Effective therapies for bone regeneration primarily require the coordinated combination of innovative scaffolds, seed cells, and biological factors. However, current techniques in bone tissue engineering have not yet reached valid translation into clinical applications because of several limitations, such as weaker osteogenic differentiation, inadequate vascularization of scaffolds, and inefficient growth factor delivery. Therefore, further standardized protocols and innovative measures are required to overcome these shortcomings and facilitate the clinical application of these techniques to enhance bone regeneration. Given the deficiency of comprehensive studies in the development in BTE, our review systematically introduces the new types of biomimetic and bifunctional scaffolds. We describe the cell sources, biology of seed cells, growth factors, vascular development, and the interactions of relevant molecules. Furthermore, we discuss the challenges and perspectives that may propel the direction of future clinical delivery in bone regeneration.

Simulation and analysis of grinding wheel based on Gaussian mixture model

Yulun CHI, Haolin LI

Frontiers of Mechanical Engineering 2012, Volume 7, Issue 4,   Pages 427-432 doi: 10.1007/s11465-012-0350-3

Abstract:

This article presents an application of numerical simulation technique for the generation and analysis of the grinding wheel surface topographies. The ZETA 20 imaging and metrology microscope is employed to measure the surface topographies. The Gaussian mixture model (GMM) is used to transform the measured non-Gaussian field to Gaussian fields, and the simulated topographies are generated. Some numerical examples are used to illustrate the viability of the method. It shows that the simulated grinding wheel topographies are similar with the measured and can be effective used to study the abrasive grains and grinding mechanism.

Performance and mechanism for cadmium and lead adsorption from water and soil by corn straw biochar

Tong Chi,Jiane Zuo,Fenglin Liu

Frontiers of Environmental Science & Engineering 2017, Volume 11, Issue 2, doi: 10.1007/s11783-017-0921-y

Abstract: Corn straw biochar prepared under 400 °C was the best Cd and Pb adsorption capacity Maximum adsorption capacity of Cd and Pb were 43.48 and 22.73 mg·g , respectively The dominant mechanism of Cd and Pb adsorption was precipitation Biochar could reduce the bioavailability of heavy metals when mixed with soil Cadmium (Cd) and lead (Pb) in water and soil could be adsorbed by biochar produced from corn straw. Biochar pyrolyzed under 400°C for 2 h could reach the ideal removal efficiencies (99.24% and 98.62% for Cd and Pb, respectively) from water with the biochar dosage of 20 g·L and initial concentration of 20 mg·L . The pH value of 4–7 was the optimal range for adsorption reaction. The adsorption mechanism was discussed on the basis of a range of characterizations, including X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), Fourier transform infrared spectroscopy (FTIR) and Raman analysis; it was concluded as surface complexation with active sorption sites (-OH, -COO-), coordination with π electrons (C= C, C= O) and precipitation with inorganic anions (OH , CO , SO ) for both Cd and Pb. The sorption isotherms fit Langmuir model rather than Freundlich model, and the saturated sorption capacities for Cd and Pb were 38.91 mg·g and 28.99 mg·g , respectively. When mixed with soil, biochar could effectively increase alkalinity and reduce bioavailability of heavy metals. Thus, biochar derived from corn straw would be a green material for both removal of heavy metals and amelioration of soil.

Comparative lipidomic analysis of

Bin QIAO, Hong-Chi TIAN, Ying-Jin YUAN

Frontiers of Chemical Science and Engineering 2012, Volume 6, Issue 4,   Pages 461-469 doi: 10.1007/s11705-012-1223-3

Abstract: Variations in the composition and level of phospholipids (PLs) in yeast cells during industrial ethanol fermentation processes were analyzed. A comparative lipidomic method was used to investigate the changes in total cellular PLs during continuous and fed-batch/batch processes. The phospholipid metabolism in yeast changed during both processes, mainly due to the presence of long-chain poly unsaturated fatty acids (PUFA) that contained phosphatidyglycerol (PG), phosphatidylethanolamine (PE) and phosphatidylserine (PS). The complexity of the media affected the growth of the yeast and the membrane composition. Yeast incorporated lots of exogenous saturated and PUFAs from the feedstock during the fermentations. During the continuous fermentation, there was an increase in PLs with shorter chains as the fermentation progressed and early in process there were more long-chains. During the fed-batch/batch process, the PG species increased as the fermentation progressed. This is probably due to an inositol deficiency in the earlier part of the fermentation.

Modulatory effect on dyslipidemia and anti-atherosclerotic function of in newly diagnosed type 2 diabetes patients

SUN Mingxiao, WANG Yao, CHI Jiamin

Frontiers of Medicine 2008, Volume 2, Issue 2,   Pages 174-177 doi: 10.1007/s11684-008-0032-z

Abstract: The modulatory effect of on dyslipidemia and the preventive effect on atherosclerosis in newly diagnosed type 2 diabetic patients were studied. A prospective clinical trial was conducted to test the effectiveness of in selected 201 newly diagnosed type 2 diabetic patients. All patients were randomly divided into two groups: 108 with therapy and 93 without therapy. The mean follow-up period was 22 months. The blood glucose and blood pressure of all patients were under control. Serum levels of total cholesterol (TC), triglyceride (TG), and low density lipoprotein cholesterol (LDL-C) were significantly lowered and their decreased percentages were significantly higher in therapy group ( < 0.05). The number of patients with arteria iliaca intima thickening was significantly lower in group of therapy ( = 0.024). With stepwise multivariate logistic regression analysis, the decreased percentage of TG was significantly and independently related with the decreased number of patients with arteria iliaca intima thickening ( = 0.005). The study demonstrates that therapy is effective on modulating dyslipidemia in newly diagnosed type 2 diabetes patients, and may be related with the improvement of early atherosclerosis.
