Antibiotics-mediated intestinal microbiome perturbation aggravates tacrolimus-induced glucose disorders

Abstract: Both immunosuppressants and antibiotics (ABX) are indispensable for transplant patients. However, the former increases the risk of new-onset diabetes, whereas the latter impacts intestinal microbiota (IM). It is still unclear whether and how the interaction between immunosuppressants and ABX alters the IM and thus leads to glucose metabolism disorders. This study examined the alterations of glucose and lipid metabolism and IM in mice exposed to tacrolimus (TAC) with or without ABX. We found that ABX further aggravated TAC-induced glucose tolerance and increased insulin secretion. Combined treatment resulted in exacerbated lipid accumulation in the liver. TAC-altered microbial community was further amplified by ABX administration, as characterized by reductions in phylum Firmicutes, family Lachnospiraceae, and genus . Analyses based on the metagenomic profiles revealed that ABX augmented the effect of TAC on microbial metabolic function mostly related to lipid metabolism. The altered components of gut microbiome and predicted microbial functional profiles showed significant correlation with hepatic lipid accumulation and glucose disorders. In conclusion, ABX aggravated the effect of TAC on the microbiome and its metabolic capacities, which might contribute to hepatic lipid accumulation and glucose disorders. These findings suggest that the ABX-altered microbiome can amplify the diabetogenic effect of TAC and could be a novel therapeutic target for patients.

Multiscale stochastic finite element method on random field modeling of geotechnical problems – a fast computing procedure

Abstract: The Green-function-based multiscale stochastic finite element method (MSFEM) has been formulated based on the stochastic variational principle. In this study a fast computing procedure based on the MSFEM is developed to solve random field geotechnical problems with a typical coefficient of variance less than 1. A unique fast computing advantage of the procedure enables computation performed only on those locations of interest, therefore saving a lot of computation. The numerical example on soil settlement shows that the procedure achieves significant computing efficiency compared with Monte Carlo method.

Reactivity of triacetone triperoxide and diacetone diperoxide: Insights from nuclear Fukui function

Abstract: Triacetone triperoxide (TATP) is more sensitive than diacetone diperoxide (DADP) in the solid-state explosion. To explain this reactivity difference, we analyzed the electronic structures and properties of the crystals of both compounds by using method to calculate the structures of their individual molecules as well as their lattice structures and particularly calculating Nuclear Fukui function to gain insight into the sensitivity of the initial, rate-determining step of their decomposition. Our results indicate that TATP and DADP crystal structures exhibit significantly different electronic properties. Most notably, the electronic structure of the TATP crystal shows asymmetry among its reactive oxygen atoms as supported by magnitudes of their nuclear Fukui functions. The greater explosion sensitivity of crystalline TATP may be attributed to the properties of its electronic structure. The electronic calculations provided valuable insight into the decomposition sensitivity difference between TATP and DADP crystals.

Stable Reliability—Based Optimization Designof Compressive Bar

Based on the stable reliability design theory and the optimization design method,  the stable reliability-based optimization of compressive bar is extensively discussed and a numerical method for stable reliability-based optimization is proposed.  On the condition of known first two moments of basic random variables,  the respective program can be used to obtain the stable reliability-based optimization information of compressive bar accurately and quickly.

Block coordinate descent with time perturbation for nonconvex nonsmooth problems in real-world studies Research Article

Abstract: The era of big data in healthcare is here, and this era will significantly improve medicine and especially oncology. However, traditional machine learning algorithms need to be promoted to solve such large-scale real-world problems due to a large amount of data that needs to be analyzed and the difficulty in solving problems with nonconvex nonlinear settings. We aim to minimize the composite of a smooth nonlinear function and a block-separable nonconvex function on a large number of block variables with inequality constraints. We propose a novel parallel first-order optimization method, called asynchronous block coordinate descent with (ATP), which adopts a technique that escapes from saddle points and sub-optimal local points. The details of the proposed method are presented with analyses of convergence and iteration complexity properties. Experiments conducted on real-world machine learning problems validate the efficacy of our proposed method. The experimental results demonstrate that enables ATP to escape from saddle points and sub-optimal points, providing a promising way to handle nonconvex optimization problems with inequality constraints employing asynchronous block coordinate descent. The asynchronous parallel implementation on shared memory multi-core platforms indicates that the proposed algorithm, ATP, has strong scalability.
