Repression of CDKN2C caused by PML/RARα binding promotes the proliferation and differentiation block

摘要:

Inappropriate cell proliferation during oncogenesis is often accompanied by inactivation of components involved in the cell cycle machinery. Here, we report that cyclin-dependent kinase inhibitor 2C (CDKN2C) as a member of the cyclin-dependent kinase inhibitors is a target of the PML/RARα oncofusion protein in leukemogenesis of acute promyelocytic leukemia (APL). We found that CDKN2C was markedly downregulated in APL blasts compared with normal promyelocytes. Chromatin immunoprecipitation combined with quantitative polymerase chain reaction demonstrated that PML/RARα directly bound to the CDKN2C promoter in the APL patient-derived cell line NB4. Luciferase assays indicated that PML/RARα inhibited the CDKN2C promoter activity in a dose-dependent manner. Furthermore, all-trans retinoic acid treatment induced CDKN2C expression by releasing the PML/RARα binding on chromatin in NB4 cells. Functional studies showed that ectopic expression of CDKN2C induced a cell cycle arrest at the G0/G1 phase and a partial differentiation in NB4 cells. Finally, the transcriptional regulation of CDKN2C was validated in primary APL patient samples. Collectively, this study highlights the importance of CDKN2C inactivation in the abnormal cell cycle progression and differentiation block of APL cells and may provide new insights into the study of pathogenesis and targeted therapy of APL.

摘要: butterfly wings were replicated by a sol-gel method and a calcination process, which could take advantage of the spatial features of the wing to enhance their photocatalytic properties. Hierarchical structures of -carbon-TiO (PP-C-TiO ) were confirmed by SEM observations. By applying the Brunauer-Emmett-Teller method, it was concluded that in the presence of wings the product shows higher surface area with respect to the pure TiO made in the absence of the wings. The higher specific surface area is also beneficial for the improvement of photocatalytic property. Furthermore, the conduction and valence bands of the PP-C-TiO are more negative than the corresponding bands of pure TiO , allowing the electrons to migrate from the valence band to the conduction band upon absorbing visible light. That is, the presence of C originating from wings in the PP-C-TiO could extend the photoresponsiveness to visible light. This strategy provides a simple method to fabricate a high-performance photocatalyst, which enables the simultaneous control of the morphology and carbon element doping.

Effect of noble metal nanoparticle size on C–N bond cleavage performance in hydrodenitrogenation: a study

摘要: Breakage of the C–N bond is a structure sensitive process, and the catalyst size significantly affects its activity. On the active metal nanoparticle scale, the role of catalyst size in C–N bond cleavage has not been clearly elucidated. So, Ru catalysts with variable nanoparticle sizes were obtained by modulating the reduction temperature, and the catalytic activity was evaluated using 1,2,3,4-tetrahydroquinoline and o-propylaniline with different C–N bond hybridization patterns as reactants. Results showed a 13 times higher reaction rate for sp3-hybridized C–N bond cleavage than sp2-hybridized C–N bond cleavage, while the reaction rate tended to increase first and then decrease as the catalyst nanoparticle size increased. Different concentrations of terrace, step, and corner sites were found in different sizes of Ru nanoparticles. The relationship between catalytic site variation and C–N bond cleavage activity was further investigated by calculating the turnover frequency values for each site. This analysis indicates that the variation of different sites on the catalyst is the intrinsic factor of the size dependence of C–N bond cleavage activity, and the step atoms are the active sites for the C–N bond cleavage. When Ru nanoparticles are smaller than 1.9 nm, they have a strong adsorption effect on the reactants, which will affect the catalytic performance of the Ru catalyst. Furthermore, these findings were also confirmed on other metallic Pd/Pt catalysts. The role of step sites in C–N bond cleavage was proposed using the density function theory calculations. The reactants have stronger adsorption energies on the step atoms, and step atoms have d-band center nearer to the Fermi level. In this case, the interaction with the reactant is stronger, which is beneficial for activating the C–N bond of the reactant.

NiCo2O4@quinone-rich N–C core–shell nanowires as composite electrode for electric double layer capacitor

摘要: The bind-free carbon cloth-supported electrodes hold the promises for high-performance electrochemical capacitors with high specific capacitance and good cyclic stability. Considering the close connection between their performance and the amount of carbon material loaded on the electrodes, in this work, NiCo2O4 nanowires were firstly grown on the substrate of active carbon cloth to provide the necessary surface area in the longitudinal direction. Then, the quinone-rich nitrogen-doped carbon shell structure was formed around NiCo2O4 nanowires, and the obtained composite was used as electrode for electric double layer capacitor. The results showed that the composite electrode displayed an area-specific capacitance of 1794 mF∙cm–2 at the current density of 1 mA∙cm–2. The assembled symmetric electric double layer capacitor achieved a high energy density of 6.55 mW∙h∙cm–3 at a power density of 180 mW∙cm–3. The assembled symmetric capacitor exhibited a capacitance retention of 88.96% after 10000 charge/discharge cycles at the current density of 20 mA∙cm–2. These results indicated the potentials in the preparation of the carbon electrode materials with high energy density and good cycling stability.

New insight into effect of potential on degradation of Fe-N-C catalyst for ORR

摘要: In recent years, Fe-N-C catalyst is particularly attractive due to its high oxygen reduction reaction (ORR) activity and low cost for proton exchange membrane fuel cells (PEMFCs). However, the durability problems still pose challenge to the application of Fe-N-C catalyst. Although considerable work has been done to investigate the degradation mechanisms of Fe-N-C catalyst, most of them are simply focused on the active-site decay, the carbon oxidation, and the demetalation problems. In fact, the 2e pathway in the ORR process of Fe-N-C catalyst would result in the formation of H O , which is proved to be a key degradation source. In this paper, a new insight into the effect of potential on degradation of Fe-N-C catalyst was provided by quantifying the H O intermediate. In this case, stability tests were conducted by the potential-static method in O saturated 0.1 mol/L HClO . During the tests, H O was quantified by rotating ring disk electrode (RRDE). The results show that compared with the loading voltage of 0.4 V, 0.8 V, and 1.0 V, the catalysts being kept at 0.6 V exhibit a highest H O yield. It is found that it is the combined effect of electrochemical oxidation and chemical oxidation (by aggressive radicals like H O /radicals) that triggered the highest H O release rate, with the latter as the major cause.

Durability of plasma-sprayed Cr3C2-NiCr coatings under rolling contact conditions

The aim of this paper was to address the rolling contact fatigue (RCF) failure mechanisms of plasma-sprayed Cr3C2-NiCr coatings under different tribological conditions of contact stress. Weibull distribution plots of fatigue lives of the coated specimens at different contact stresses were obtained. The failure modes of coatings were identified on the basis of wore surface observations of the failed coatings. Results showed that the RCF failure modes can be classified into four main categories, i.e., surface abrasion, spalling, cohesive delamination, and interfacial delamination. The probabilities of the surface abrasion and spalling type failures were relatively high at low contact stress. When the coatings were subjected to abrasion and spalling type failures, the failure of the coating was depended on the microstrcture of the coating. The stress concentration near the micro-defects in the coating may be the may reason for the formation of spall. The coatings were prone to fail in delamination under higher contact stresses. However, the delamination of coating may be related to distribution of shear stress amplitude within coating. The location of maximum shear stress amplitude can be used as a key parameter to predict the initiation of subsurface cracks within coating in rolling contact.

Plasma soluble C-type lectin-like receptor-2 is associated with the risk of coronary artery disease

摘要: Accumulating evidence suggests that C-type lectin-like receptor-2 (CLEC-2) plays an important role in atherothrombosis. In this case-control study, we investigated the association between CLEC-2 and incidence of coronary artery disease (CAD). A total of 216 patients, including 14 cases of stable angina pectoris (SAP, non-ACS) and 202 cases of acute coronary syndrome (ACS), and 89 non-CAD control subjects were enrolled. Plasma levels of soluble CLEC-2 (sCLEC-2) were measured using the enzyme-linked immunosorbent assay (ELISA). Compared with the control group (65.69 (55.36–143.22) pg/mL), the plasma levels of sCLEC-2 were significantly increased in patients with CAD (133.67 (88.76–220.09) pg/mL) and ACS (134.16 (88.88–225.81) pg/mL). The multivariate adjusted odds ratios (95% confidence interval) of CAD reached 2.01 (1.52–2.66) ( <0.001) for each 1-quartile increase in sCLEC-2. Restricted cubic splines showed a positive dose-response association between sCLEC2 and CAD incidence ( <0.001). The addition of sCLEC-2 to conventional risk factors improved the C statistic (0.821 vs. 0.761, = 0.004) and reclassification ability (net reclassification improvement: 57.45%, <0.001; integrated discrimination improvement: 8.27%, <0.001) for CAD. In conclusion, high plasma sCLEC-2 is independently associated with CAD risk, and the prognostic value of sCLEC-2 may be evaluated in future prospective studies.

A state-of-the-art review of solar passive building system for heating or cooling purpose

摘要: The major portion of energy in a building is consumed by heating, ventilating, and air-conditioning (HVAC). The traditional heating and cooling systems contribute greatly to the emission of greenhouse gases, especially carbon dioxide. Four different ways, i.e., Trombe wall, solar chimney, unglazed transpired solar façade, and solar roof, are adopted for solar heating. Similarly, two major ways, i.e., evaporative cooling and building integrated evaporative cooling are adopted for cooling of the building. Therefore, an attempt has been made in this paper to compile the developments of solar heating and cooling technologies in a building.

In situ DRIFTS study of photocatalytic CO 2 reduction under UV irradiation

摘要: Photocatalytic reduction of CO on TiO and Cu/TiO photocatalysts was studied by in situ diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) under UV irradiation. The photocatalysts were prepared by sol-gel method via controlled hydrolysis of titanium (IV) butoxide. Copper precursor was loaded onto TiO during sol-gel procedure. A large amount of adsorbed HO and surface OH groups was detected at 25°C on the TiO photocatalyst after being treated at 500°C under air stream. Carbonate and bicarbonate were formed rapidly due to the reaction of CO with oxygen-vacancy and OH groups, respectively, on TiO surface upon CO adsorption. The IR spectra indicated that, under UV irradiation, gas-phase CO further combined with oxygen-vacancy and OH groups to produce more carbonate or bicarbonate. The weak signals of reaction intermediates were found on the IR spectra, which were due to the slow photocatalytic CO reduction on photocatalysts. Photogenerated electrons merge with H ions to form H atoms, which progressively reduce CO to form formic acid, dioxymethylene, formaldehyde and methoxy as observed in the IR spectra. The well-dispersed Cu, acting as the active site significantly increases the amount of formaldehyde and dioxymethylene, thus promotes the photoactivity of CO reduction on Cu/TiO. A possible mechanism of the photocatalytic CO reduction is proposed based on these intermediates and products on the photocatalysts.

摘要: A bio-electrochemical fuel cell reactor with cathodic Fe /TiO generates electricity. It destroys recalcitrant pollutants in cathode chamber without photocatalysis. Fe /TiO generates reactive oxygenated species in the dark or under photocatalysis. Cathodic produced ROS (hydroxy radical/superoxide radical) can degrade tetracycline or dyes. Electricity generation is enhanced by semiconductor catalyzed cathodic degradation of pollutants. In this study, a new water treatment system that couples (photo-) electrochemical catalysis (PEC or EC) in a microbial fuel cell (MFC) was configured using a stainless-steel (SS) cathode coated with Fe /TiO . We examined the destruction of methylene blue (MB) and tetracycline. Fe /TiO was prepared using a chemical reduction-deposition method and coated onto an SS wire mesh (500 mesh) using a sol technique. The anode generates electricity using microbes (bio-anode). Connected via wire and ohmic resistance, the system requires a short reaction time and operates at a low cost by effectively removing 94% MB (initial concentration 20 mg·L ) and 83% TOC/TOC under visible light illumination (50 W; 1.99 mW·cm for 120 min, MFC-PEC). The removal was similar even without light irradiation (MFC-EC). The of the MFC-PEC system was approximately 0.675 kWh·m ·order , whereas that of the MFC-EC system was zero. The system was able to remove 70% COD in tetracycline solution (initial tetracycline concentration 100 mg·L ) after 120 min of visible light illumination; without light, the removal was 15% lower. The destruction of MB and tetracycline in both traditional photocatalysis and photoelectrocatalysis systems was notably low. The electron spin-resonance spectroscopy (ESR) study demonstrated that ·OH was formed under visible light, and ·O was formed without light. The bio-electricity-activated O and ROS (reactive oxidizing species) generation by Fe /TiO effectively degraded the pollutants. This cathodic degradation improved the electricity generation by accepting and consuming more electrons from the bio-anode.

Greenhouse gas emissions during co-composting of cattle feedlot manure with construction and demolition (C&

摘要: Manure management strategies should reflect current animal feeding practices and encourage recycling of organic waste to help protect our environment. This research investigated greenhouse gas (GHG) emissions during cattle manure stockpiling or composting with and without construction and demolition (C&D) waste. Manure was collected from cattle fed a typical finishing diet (CK manure) and from cattle on diets which included 30% dried distillers grains with solubles (DG manure). The CK and DG manures were co-composted with (4:1) C&D waste (treatments: CK_CD DG_CD), composted alone (treatments: CK and DG) in 13 m bins or stockpiled without C&D waste (treatments: CK_ST and DG_ST) for 99 days. Manure type (CK vs. DG manure) had no effect on GHG emissions over the 99 day manure composting or stockpiling. Composting with C&D waste produced similar CO emissions, about double that from manure stockpiling (7.0 kgC·m ). In contrast, CH emissions were reduced by the inclusion of C&D waste (64 gC·m with C&D vs. 244 gC·m without C&D) while the manure stockpile emitted the greatest amount of CH (464 gC·m ). Additionally, only 0.48% of C was emitted in CH form with C&D waste, compared to 1.68% when composting without C&D waste and 7.00% when cattle manure was stockpiled. The N O emissions (12.4 to 18.0 gN·m ) were similar across all treatments. The lower CH emissions with C&D waste are beneficial in reducing overall GHG emissions from manure composting, while reducing the amount of material entering landfills.

摘要: 2R-1C模型的参数估计常运用需要高性能处理单元的迭代方法,从而激励我们研究更省时且更节能的参数估计方法。这些低复杂度的算法将更适于便携式微机设备的运行。运用一组封闭表达式从测量阻抗的实部和虚部计算2R-1C模型的参数。此外,比较了QINIPE和复杂非线性最小二乘法(CNLS)对2R-1C模型参数的估计。结果表明,虽然QINIPE估计精度稍低于CNLS,其依然适合许多实际应用,且运行时间降至原来的1/45至1/30。
