Abstract: Lung transplantation is increasingly practiced for patients with end-stage lung disease. The successful outcome of solid organ transplantation today is severely impeded by the production of alloantibodies, mainly directed against the protein products of the HLA complex of the organ donor. While the association between antibody mediated rejection and allograft damage has been well established in renal and heart transplantation, it has not yet been well characterized in lung transplantation. This review addresses the question of HLA matching in lung transplantation and current knowledge of the allogenicity of different HLA class I and II antigens. The role of the antibody mediated immune response is discussed as well as the importance of pre-transplant or post-transplant circulating antibodies. Finally, potential mechanisms, which may act individually or in combination, of antibody mediated damage to solid organ transplants are considered.

Abstract: Growing evidence suggests that somatic hypermutational status and programmed cell death-1 overexpression are potential predictive biomarkers indicating treatment benefits from immunotherapy using immune checkpoint inhibitors. However, biomarker-matched trials are still limited, and many of the genomic alterations remain difficult to target. To isolate the potential somatic hypermutational tumor from microsatellite instability low/microsatellite stability (MSI-L/MSS) cases, we employed two commercial kits to determine MSI and forensic short tandem repeat (STR) alternations in 250 gastrointestinal (GI) tumors. Three types of forensic STR alternations, namely, allelic loss, Aadd, and Anew, were identified. 62.4% (156/250) of the patients with GI exhibited STR alternation, including 100% (15/15) and 60% (141/235) of the microsatellite high instability and MSI-L/MSS cases, respectively. 30% (75/250) of the patients exhibited STR instability with more than 26.32% (26.32%–84.21%) STR alternation. The cutoff with 26.32% of the STR alternations covered all 15 MSI cases and suggested that it might be a potential threshold. Given the similar mechanism of the mutations of MSI and forensic STR, the widely used forensic identifier STR kit might provide potential usage for identifying hypermutational status in GI cancers.

Abstract: As an increasingly popular flow metering technology, exhibits high measurement accuracy under single-phase flow condition and is widely used in the industry. However, under complex flow conditions, such as two-phase flow, the measurement accuracy is greatly decreased due to various factors including improper signal processing methods. In this study, three s—the (QD) method, Hilbert method, and sliding discrete time Fourier transform method—are analyzed for their applications in processing sensor signals and providing measurement results under gas-liquid . Based on the analysis, specific improvements are applied to each method to deal with the signals under . For simulation, sensor signals under single- and s are established using a random walk model. The phase difference tracking performances of these three methods are evaluated in the simulation. Based on the digital signal processor, a converter program is implemented on its evaluation board. The converter program is tested under single- and s. The improved signal processing methods are evaluated in terms of the measurement accuracy and complexity. The QD algorithm has the best performance under the single-phase flow condition. Under the , the QD algorithm performs a little better in terms of the indication error and repeatability than the improved Hilbert algorithm at 160, 250, and 420 kg/h flow points, whereas the Hilbert algorithm outperforms the QD algorithm at the 600 kg/h flow point.
