Brazilian biosafety law and the new breeding technologies

Globally, the area of land cultivated with genetically modified (GM) crops has increased a thousand-fold over the last two decades. Although this technology has become important for food production, the regulatory frameworks that underpin these outcomes are based on a list of requirements for a risk assessment that differ from country to country. In recent years, policymakers have had the opportunity to learn from the controversies over transgenics to create effective regulatory milestones for emerging technologies, allowing them to reach their potential for a more sustainable agriculture, ensuring food security. In Brazil, Law No11.105 of 24 March 2005 established a framework with four main organizations responsible for risk assessment and management. However, most of new breeding technologies did not exist at that time and were not considered in this law. In 2016, Normative Resolution No 16 of the National Biosafety Technical Commission (CTNBio) was established to address this gap based on the evaluation of the products obtained through these techniques (termed Innovative Precision Improvement Techniques in the resolution), in a case-by-case consultation system. Briefly, if the product is designated to be a genetically modified, the developer will have to go through the biosafety requirements and will be approved only after CTNBio risk assessment. If the product is designated not to be GM (for the purposes of the legislation), then it can be registered using the existing procedures. Currently, 152 GM products are commercially approved in Brazil. In 2018, CTNBio assessed the first consultation on commercial release of plants generated using the new breeding technologies and has subsequently approved six products. It is expected that many institutions would be able to participate in Brazilian and world markets, developing and introducing new biotechnological solutions and products through a more sustainable approach and without facing public disapproval, a common issue for GM crops.

Functional magnetic nanoparticles for facile viable but nonculturable bacteria separation and purification

摘要: The functional surface-modified MNPs were capable of capture with high efficiency. After induced to VBNC state by chlorination, cells could be separated by MNPs with an additional incubation process. This study provides a facile and economic method for VBNC cell enrichment and purification. GRAPHIC ABSTRACT Viable But Nonculturable (VBNC) Bacteria, which represent a unique population of microorganisms in drinking water systems, have become a potential threat to human health. Current studies on VBNC cells usually fail to obtain pure VBNC state bacteria, which may lead to inaccurate results. We therefore introduce a novel method of VBNC cell separation and purification in this paper. PAH-coated magnetic nanoparticles (MNPs) were synthesized and found to be capable of capturing and releasing bacterial cells with high efficiency. With the aid of an additional incubation step, VBNC cells were easily isolated and purified from normal bacteria using functional MNPs. Our method represents a new technique that can be utilized in studies of VBNCs.

Potential shift of bacterial community structure and corrosion-related bacteria in drinking water distribution pipeline driven by water source switching

摘要: Abstract • Bacterial release from aged pipe sections under extreme conditions was quantified. • Released bacterial community structure exhibited large variation after transition. • Risks from transition reduced significantly with cleaner source. As a result of pollution in the present water sources, cities have been forced to utilize cleaner water sources. There are few reports regarding the potential shift of bacterial community structure driven by water source switching, especially that of corrosion-related bacteria. Three types of finished water were used for simulation, the polluted source water from the Qiantang and Dongtiaoxi Rivers (China) was replaced by cleaner water from Qiandao Lake (China). Here, we discussed the transition effects through three simulated reactors. The bacterial characteristics were identified using the high-throughput sequencing and heterotrophic plate count method. It was observed that the level of culturable bacteria declined by 2–3 orders of magnitude after water source switching. The bacterial community released from the pipeline reactor was significantly different under different finished water, and it exhibited large variation at the genus level. Porphyrobacter (58.2%) and Phreatobacter (14.5%) clearly replaced Novosphingobium, Aquabacterium, and Cupriavidus as new dominant genera in system A, which could be attributed to the lower carbon and nitrogen content of the new water source. Although corrosion-inhibiting bacteria decreased after switching, they still maintained dominant in three reactors (6.6%, 15.9%, and 19.7%). Furthermore, potential opportunistic pathogens such as Sphingomonas were detected. Our study shows that after transition to a high quality water source, the total culturable bacteria released was in a downtrend, which leads to a great reduction in the risk of bacterial leakage in the produced drinking water.

Process stability and microbial community composition in pig manure and food waste anaerobic co-digesters operated at low HRTs

摘要: This study assessed the effects of reducing hydraulic retention times (HRTs) from 21 days to 10.5 days when anaerobically co-digesting pig manure and food waste. Continuously stirred tank reactors of 3.75 L working volume were operated in triplicate at 42°C. Digester HRT was progressively decreased from 21 to 15 days to 10.5 days, with an associated increase in organic loading rate (OLR) from 3.1 kg volatile solids (VS)·m ·day to 5.1 kg VS·m ·day to 7.25 kg VS·m ·day . Reducing HRT from 21 days to 15 days caused a decrease in specific methane yields and VS removal rates. Operation at a HRT of 10.5 days initially resulted in the accumulation of isobutyric acid in each reactor. High throughput 16S rRNA gene sequencing revealed that this increase coincided with a shift in acidogenic bacterial populations, which most likely resulted in the increased isobutyric acid concentrations. This may in turn have caused the increase in relative abundance of bacteria, which syntrophically degrade non-acetate volatile fatty acids (VFAs) into H and CO . This, along with the increase in abundance of other syntrophic VFA oxidizers, such as suggests that VFA oxidation plays a role in digester operation at low HRTs. Reducing the HRT to below 21 days compromised the ability of the anaerobic digestion system to reduce enteric indicator organism counts below regulatory limits.
