carcinoma progression by repressing translation through the interaction with eukaryotic initiation factor 4A-I

摘要: tRNA-derived small RNAs (tsRNAs) are novel non-coding RNAs that are involved in the occurrence and progression of diverse diseases. However, their exact presence and function in hepatocellular carcinoma (HCC) remain unclear. Here, differentially expressed tsRNAs in HCC were profiled. A novel tsRNA, tRNAGln-TTG derived 5′-tiRNA-Gln, is significantly downregulated, and its expression level is correlated with progression in patients. In HCC cells, 5′-tiRNA-Gln overexpression impaired the proliferation, migration, and invasion in vitro and in vivo, while 5′-tiRNA-Gln knockdown yielded opposite results. 5′-tiRNA-Gln exerted its function by binding eukaryotic initiation factor 4A-I (EIF4A1), which unwinds complex RNA secondary structures during translation initiation, causing the partial inhibition of translation. The suppressed downregulated proteins include ARAF, MEK1/2 and STAT3, causing the impaired signaling pathway related to HCC progression. Furthermore, based on the construction of a mutant 5′-tiRNA-Gln, the sequence of forming intramolecular G-quadruplex structure is crucial for 5′-tiRNA-Gln to strongly bind EIF4A1 and repress translation. Clinically, 5′-tiRNA-Gln expression level is negatively correlated with ARAF, MEK1/2, and STAT3 in HCC tissues. Collectively, these findings reveal that 5′-tiRNA-Gln interacts with EIF4A1 to reduce related mRNA binding through the intramolecular G-quadruplex structure, and this process partially inhibits translation and HCC progression.

Metabolic interventions combined with CTLA-4 and PD-1/PD-L1 blockade for the treatment of tumors: mechanisms

摘要: Immunotherapies based on immune checkpoint blockade (ICB) have significantly improved patient outcomes and offered new approaches to cancer therapy over the past decade. To date, immune checkpoint inhibitors (ICIs) of CTLA-4 and PD-1/PD-L1 represent the main class of immunotherapy. Blockade of CTLA-4 and PD-1/PD-L1 has shown remarkable efficacy in several specific types of cancers, however, a large subset of refractory patients presents poor responsiveness to ICB therapy; and the underlying mechanism remains elusive. Recently, numerous studies have revealed that metabolic reprogramming of tumor cells restrains immune responses by remodeling the tumor microenvironment (TME) with various products of metabolism, and combination therapies involving metabolic inhibitors and ICIs provide new approaches to cancer therapy. Nevertheless, a systematic summary is lacking regarding the manner by which different targetable metabolic pathways regulate immune checkpoints to overcome ICI resistance. Here, we demonstrate the generalized mechanism of targeting cancer metabolism at three crucial immune checkpoints (CTLA-4, PD-1, and PD-L1) to influence ICB therapy and propose potential combined immunotherapeutic strategies co-targeting tumor metabolic pathways and immune checkpoints.

Synthesis, insecticidal activities and DFT study of pyrimidin-4-amine derivatives containing the 1,2,4

摘要: Twenty six novel pyrimidin-4-amine derivatives containing the 1,2,4-oxadiazole motif were synthesized. Their chemical structures were confirmed by 1H nuclear magnetic resonance (NMR), 13C NMR, and high-resolution mass spectrography. The insecticidal activity results indicated that some of them possessed excellent insecticidal activity (100%) against Mythimna separate, especially for compounds 6d, 6f, 6o, 6w, 6y and 6z. These compounds exhibited no activity against the insects Aphis medicagini and Tetranychus cinnabarinus. The structure- insecticidal activity relationships are discussed. Density functional theory analysis can potentially be used to design more active compounds. These results provide useful insecticide design information for further optimization.

摘要: 亚波长结构以及双谐振特性确保了透射单元在28.4%的1-dB透射带宽内具有稳定的相移能力。基于此,加工测试了一款由喇叭天线馈电的方形口径透射阵。得益于蜂窝状布阵方式,所设计的透射阵可在28.5 GHz到38 GHz的宽带范围中辐射l=–1且模态纯度高于0.93的涡旋波束。测试的峰值增益为22.3 dBic,3-dB轴比带宽为27.6%。

摘要: 2-aryl-3-(naphthalene-1 or 2-yl)-1,3-thiazolidin-4-ones and were synthesized in 41%-67% yield by using microwave-assisted one-pot protocol. The structures of the new compounds , , , , , , and - were confirmed by IR, NMR, MS, and elemental analysis. The antimicrobial activities of the compounds against pv. lachrymans (Smith et Bryan) Young, Dye & Wilkie, Pers., and Blum. were examined. Some of the compounds showed good antifungical activity against Blum.

4-1BBL expressed by eukaryotic cells activates immune cells and suppresses the progression of murine

摘要: The interaction by co-stimulatory molecules 4-1BB and 4-1BB ligand (4-1BBL) plays an important role in the activation, proliferation and differentiation of T lymphocytes. The function of 4-1BB/4-1BBL expressed by the immune cells has been the focus for many tumor immunotherapy efforts. In this study, 4-1BBL was expressed in non-immune cells and non-tumor cells, and the role of 4-1BBL in lymphocyte activation and tumor suppression was investigated. The plasmid p4-1BBL containing the full length of mouse 4-1BBL cDNA sequence was constructed, and the plasmid was transfected into baby hamster kidney (BHK) cells and murine muscle cells by means of lipofectin-mediated or naked plasmid DNA injection into the muscle directly. The study demonstrated that the molecule 4-1BBL expressed by BHK cells could enhance the proliferation and cytotoxicity of lymphocytes, and it could increase the expression level of IL-2 and IFN-γ. The treatment with plasmid p4-1BBL revealed that the number of CD8 T cells in the peri-tumoral tissue increased markedly, and the growth rate of the tumor was significantly lower than that of control group. These findings suggest that expression of 4-1BBL by normal cells in the tumor microenvironment can enhance the proliferation and other functions of T lymphocytes. This therapeutic method may provide a promising approach for tumor immunotherapy.

摘要:

近年来,人们对信息处理的需求日益增长,脑启发式神经形态器件得到了广泛的关注。突触器件作为一类重要的神经形态器件,在短短几年内迅速升温。在用于制备突触器件的各种材料中,半导体纳米晶体(NCs)因其优异的电学和光学性能而成为首选材料之一。本综述论文首先介绍了基于半导体纳米晶体的突触器件的研究背景及半导体纳米晶体的基本性质。然后,根据器件有源层所用纳米晶体种类的不同,分类介绍了基于纳米晶体的突触器件的最新研究进展。最后,讨论了基于半导体纳米晶体的突触器件目前仍面临的问题和挑战。

基于二硫化钛可饱和吸收体的被动锁模掺铒光纤激光器 Research Articles

摘要: 本文采用液相剥离法和旋涂法合成调制深度为5.08%、饱和强度为10.62 MW/cm2的二硫化钛聚乙烯醇薄膜型可饱和吸收体。由于二硫化钛可饱和吸收体具有很强的非线性饱和吸收特性,在掺铒锁模光纤激光器中观测到两种类型的光孤子。当泵浦功率达到67.3 mW时,产生重复率为1.716 MHz、脉宽为6.57 ps的传统锁模脉冲串,其输出光谱中心为1556.98 nm、半高全宽为0.466 nm,且有明显对称的Kelly边带。通过调整偏振控制器,得到另一种锁模脉冲,在517.2 mW泵浦功率下,其最大输出功率为3.92 mW,脉冲能量为2.28 nJ。实验证明层状二维材料二硫化钛具有优异的非线性饱和吸收特性,在超快光子学领域具有广阔应用前景。

摘要: 基底微结构制作的薄膜衍射光学元件具备超轻质量(面密度小于0.1 kg/m2)和宽松表面形状公差(厘米级表面精度需求)两个重要特性,将其作为大口径望远镜的主镜可实现超大口径,超轻量化,同时降低发射成本。本文对国内外基于衍射光学的空间大口径望远系统的研究进展进行归纳和总结。首先阐述衍射望远系统的成像理论与组成结构,然后介绍衍射望远系统研究进展,最后总结衍射技术作为未来高分辨率空间侦查系统的发展趋势,提出我国应着重开展的相关工作。

Hepatocyte nuclear factor 1 alpha (HNF1A), hepatocyte nuclear factor 4 alpha (HNF4A), and forkhead box protein A2 (FOXA2) are key transcription factors that regulate a complex gene network in the liver, creating a regulatory transcriptional loop. The Encode and ChIP-Atlas databases identify the recognition sites of these transcription factors in many glycosyltransferase genes. Our in silico analysis of HNF1A, HNF4A, and FOXA2 binding to the 10 candidate glyco-genes studied in this work confirms a significant enrichment of these transcription factors specifically in the liver. Our previous studies identified HNF1A as a master regulator of fucosylation, glycan branching, and galactosylation of plasma glycoproteins. Here, we aimed to functionally validate the role of the three transcription factors on downstream glyco-gene transcriptional expression and the possible effect on glycan phenotype. We used the state-of-the-art clustered regularly interspaced short palindromic repeats/dead Cas9 (CRISPR/dCas9) molecular tool for the downregulation of the HNF1A, HNF4A, and FOXA2 genes in HepG2 cells—a human liver cancer cell line. The results show that the downregulation of all three genes individually and in pairs affects the transcriptional activity of many glyco-genes, although downregulation of glyco-genes was not always followed by an unambiguous change in the corresponding glycan structures. The effect is better seen as an overall change in the total HepG2 N-glycome, primarily due to the extension of biantennary glycans. We propose an alternative way to evaluate the N-glycome composition via estimating the overall complexity of the glycome by quantifying the number of monomers in each glycan structure. We also propose a model showing feedback loops with the mutual activation of HNF1A–FOXA2 and HNF4A–FOXA2 affecting glyco-genes and protein glycosylation in HepG2 cells.

摘要: A series of new benzimidazole derivatives were designed and synthesized. Their chemical structures were testified by H NMR, infrared spectroscopy (IR), mass spectrography (MS), and elemental analysis. Their potent antiviral properties indicated the prospect of new drugs. Compound , , , , , , and were identified as novel antivirus with much better selective activity and inhibitory activity than the comparable ribavirin against Coxsackie virus B in VERO cells.

摘要: 研究发现羧酸酯酶1(CES1)作为肝内标志物和二肽基肽酶4(DPP-IV)作为肝外标志物可反映肝脏损伤的不同病理生理状态。CES1 和DPP-IV 水平可甄别肝损伤本身和炎症损伤之间的差异。尽管血清与组织中γ-谷氨酰基转肽酶(γ-GT)升降水平的变化方向相反,但其持续时间远短于CES1,并迅速恢复到正常水平。在上述生物标志物中,只有CES1 能够明确排除炎症干扰下的肝细胞损伤。CES1 还能准确评估熊去氧胆酸(UDCA;单成分药物)和清肺排毒汤(QFPDD;多组分药物)的抗胆汁淤积作用。本文数据强调了联合使用CES1(作为肝内肝损伤标志物)和DPP-IV(作为肝外炎症作用标志物)可准确评估和追踪特异性肝损伤,并可区分肝损伤和炎症性肝损伤的差异。
