Application of StrucGP in medical immunology: site-specific -glycoproteomic analysis of macrophages

摘要: The structure of N-glycans on specific proteins can regulate innate and adaptive immunity via sensing environmental signals. Meanwhile, the structural diversity of N-glycans poses analytical challenges that limit the exploration of specific glycosylation functions. In this work, we used THP-1-derived macrophages as examples to show the vast potential of a N-glycan structural interpretation tool StrucGP in N-glycoproteomic analysis. The intact glycopeptides of macrophages were enriched and analyzed using mass spectrometry (MS)-based glycoproteomic approaches, followed by the large-scale mapping of site-specific glycan structures via StrucGP. Results revealed that bisected GlcNAc, core fucosylated, and sialylated glycans (e.g., HexNAc4Hex5Fuc1Neu5Ac1, N4H5F1S1) were increased in M1 and M2 macrophages, especially in the latter. The findings indicated that these structures may be closely related to macrophage polarization. In addition, a high level of glycosylated PD-L1 was observed in M1 macrophages, and the LacNAc moiety was detected at Asn-192 and Asn-200 of PD-L1, and Asn-200 contained Lewis epitopes. The precision structural interpretation of site-specific glycans and subsequent intervention of target glycoproteins and related glycosyltransferases are of great value for the development of new diagnostic and therapeutic approaches for different diseases.

Achievements in burn surgery over the past 50 years in China

摘要: This paper reflects on the advancements of clinical and scientific research in the field of burn surgery in China. It includes emergency care of massive burns, resuscitation, anti-infection, prevention and cure of internal organ injuries, metabolic and nutritional support, wound repair and rehabilitation, and special types of burns; it also covers pathology, microbiology, immunology, cell biology, molecular biology, and tissue engineering.

Mechanism of arterial remodeling in chronic allograft vasculopathy

摘要: Chronic allograft vasculopathy (CAV) remains a major obstacle for long-term survival of grafts even though therapeutic strategies have improved considerably in recent years. CAV is characterized by concentric and diffuse neointimal formation, medial apoptosis, infiltration of lymphocyte or inflammatory cells, and deposition of extracellular matrix both in arteries and veins. Recent studies have shown that stem cells derived from the recipient contribute to neointimal formation under the regulation of chemokines and cytokines. Arterial remodeling in allografts eventually causes ischemic graft failure. The pathogenesis is multi-factorial with both immunologic and non-immunological factors being involved. The immunological factors have been discussed extensively in other articles. This review focuses mainly on the arterial remodeling that occurs in 3 layers of vessel walls including intimal injury, accumulation of smooth muscle-like cells in the neointimal, medial smooth muscle cell apoptosis, adventitial fibrosis, and deposition of extracellular matrix.

肝内区域免疫对肝移植后肿瘤复发的影响 Review

摘要:

肝脏恶性肿瘤是肝移植的一个主要适应症,但是肝移植后肿瘤复发却是影响受体长期生存的一个严峻临床挑战。肿瘤生物学特征、分期和移植后的免疫抑制状态一直被认为是肝癌复发的危险因素。而越来越多的证据表明,肝脏缺血再灌注(IR)对同种异体移植物的损伤则为肝移植后的肿瘤细胞侵袭性、转移性
提供了有利的免疫微环境。在活体肝移植中,边缘移植物(如小体积或脂肪移植物)的严重损伤与较低的无复发生存率之间的相关性,证实了IR 损伤与肿瘤复发之间的关联。IR 可引起肝内免疫微环境重构,包括恶化移植物损伤的促炎反应和加快组织修复的抗炎反应。然而,肝内区域免疫对移植后肿瘤复发的作
用尚不清晰。本文详述了IR 损伤诱导的肝内体液微环境和调节性区域免疫微环境,以及它们如何影响肝移植后肿瘤复发的最新研究进展。综合理解移植后肝内区域免疫,将为移植后肿瘤复发提供精准诊断、治疗和预后预测的新策略。
