RGS16 regulated by let-7c-5p promotes glioma progression by activating PI3K-AKT pathway

摘要: Gliomas are the most common central nervous system tumours; they are highly aggressive and have a poor prognosis. RGS16 belongs to the regulator of G-protein signalling (RGS) protein family, which plays an important role in promoting various cancers, such as breast cancer, pancreatic cancer, and colorectal cancer. Moreover, previous studies confirmed that let-7c-5p, a well-known microRNA, can act as a tumour suppressor to regulate the progression of various tumours by inhibiting the expression of its target genes. However, whether RGS16 can promote the progression of glioma and whether it is regulated by miR let-7c-5p are still unknown. Here, we confirmed that RGS16 is upregulated in glioma tissues and that high expression of RGS16 is associated with poor survival. Ectopic deletion of RGS16 significantly suppressed glioma cell proliferation and migration both in vitro and in vivo. Moreover, RGS16 was validated as a direct target gene of miR let-7c-5p. The overexpression of miR let-7c-5p obviously downregulated the expression of RGS16, and knocking down miR let-7c-5p had the opposite effect. Thus, we suggest that the suppression of RGS16 by miR let-7c-5p can promote glioma progression and may serve as a potential prognostic biomarker and therapeutic target in glioma.

Characterization and catalytic properties of mesoporous CuO/SBA-16 prepared by different impregnation

摘要: CuO/SBA-16 catalysts were prepared by two different routes – the conventional impregnation method and the modified impregnation method with pH adjustment. These catalysts were characterized by X-ray diffraction (XRD), atomic absorption spectrometry (AAS), N physisorption and hydrogen temperature programmed reduction (H-TPR) measurements which reveal that the cubic cage-like () pore structure of the parent SBA-16 molecule sieves was well maintained throughout the synthesis. After introduction of Cu, a different CuO dispersion exists on these catalysts. The CuO/SBA-16 prepared by modified impregnation method has a single highly dispersed CuO which is considered as a highly efficient species for hydroxylation of phenol with HO. CuO/SBA-16 prepared by the conventional impregnation method shows the presence of bulk CuO species which is undesirable for this reaction.

Zfyve16 regulates the proliferation of B-lymphoid cells

摘要:

Zfyve16 (a.k.a. endofin or endosome-associated FYVE-domain protein), a member of the FYVE-domain protein family, is involved in endosomal trafficking and in TGF-β, BMP, and EGFR signaling. The FYVE protein SARA regulates the TGF-β signaling pathway by recruiting Smad2/3 and accelerating their phosphorylation, thereby altering their susceptibility to TGF-β-mediated T cell suppression. Zfyve16 binds to Smad4 and their binding affects the formation of Smad2/3-Smad4 complex in TGF-β signaling. However, the in vivo function of Zfyve16 remains unknown. In this study, we generated a Zfyve16 knockout mouse strain (Zfyve16KO) and examined its hematopoietic phenotypes and hematopoietic reconstruction ability. The proportion of T cells in the peripheral blood of Zfyve16KO mice increases compared with that in wild-type mice. This finding is consistent with the role of Zfyve16 in facilitating TGF-β signaling. Unpredictably, B cell proliferation is inhibited in Zfyve16KO mice. The proliferation potential of Zfyve16KO B-lymphoid cells also significantly decreases in vitro. These results suggest that Zfyve16 inhibits the proliferation of T cells, possibly through the TGF-β signaling, but upregulates the proliferation of B-lymphoid cells.

Mutation profiling of 16 candidate genes in

摘要: This retrospective analysis aimed to investigate the mutation profile of 16 common mutated genes in acute myeloid leukemia (AML) patients. A total of 259 patients who were diagnosed of AML were enrolled in this study. Mutation profiling of 16 candidate genes were performed in bone marrow samples by using Sanger sequencing. We identified at least 1 mutation in 199 of the 259 samples (76.8%), and 2 or more mutations in 31.7% of samples. was the most common mutated gene (16.2%, 42/259), followed by (15.1%, 39/259), (14.7%, 38/259), and (13.5%, 35/259). Concurrence was observed in 97.1% of the mutated cases and in 29.6% of the double mutated cases. Distinct patterns of co-occurrence were observed for different hotspot mutations within the gene: mutations were associated with and/or mutations, whereas mutations co-occurred with mutations only. Concurrence was also observed in 86.6% of epigenetic regulation genes, most of which co-occurred with mutations. The results showed certain rules in the mutation profiling and concurrence of AML patients, which was related to the function classification of genes. Defining the mutation spectrum and mutation pattern of AML will contribute to the comprehensive assessment of patients and identification of new therapeutic targets.

Controlled synthesis of uniform silver nanowires with high aspect ratios in aqueous solutions of gemini surfactant

摘要: A simple solution-phase approach has been demonstrated for the large-scale synthesis of silver nanowires with diameters in the range of 15 25 nm, and lengths usually in the range of tens of micrometers. In the presence of gemini surfactant 1,3-bis(cetyldimethylammonium) propane dibromide (16-3-16), the growth of silver could be directed into a highly anisotropic mode to form uniform nanowires with aspect ratios up to about 2,000. X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM), energy-dispersive X-ray (EDX), X-ray powder diffraction (XRD), electron diffraction (ED), and UV-vis absorption spectroscopy, were used to characterize the as-prepared silver nanowires, indicating the formation of a highly pure phase, good crystallinity, as well as a uniform diameter.

摘要: We implemented a new protocol — multiphase dynamic helical scan to acquire CT angiography (CTA) and whole brain CT perfusion (CTP) images simultaneously with single scan on 16 multidetector CT (MDCT). A total of 90 patients who were randomly assigned into 3 groups were included in our study. Each group underwent CT scan by using the new protocol, traditional CTA and CTP protocol, respectively. The image quality of CTA, the CTP parameter values and the X-ray doses were measured and compared between the new protocol and the traditional protocols. There was no statistically significant difference in the CTA image quality between the above methods ( =0.55). For CTP parameters, the new protocol tended to overestimate the blood volume (BV) and blood flow (BF) value, and to underestimate the mean transit time (MTT) value compared with the traditional method. However, there was no statistically significant difference in BV, BF, and MTT value between the two methods except permeability surface (PS) ( >0.05). The volume CT dose index (CTDIvol) and dose length product (DLP) of our protocol were lower than the traditional one. The new protocol can obtain valuable diagnostic information in a shorter time without significant compromise in image quality. In addition, it reduces the radiation dose as well as contrast medium usage on the patient.

Biologic risk and source diagnose of 16 PAHs from Haihe River Basin, China

摘要: Surface sediments of rivers can exhibit spatial and temporal variations in contaminant concentrations that may significantly affect risk evaluations. As to pollution control and remediation of watershed, large-scale and further background data on PAHs in China were required urgently. Spatial distribution and compositional characteristics of 16 polycyclic aromatic hydrocarbons (PAHs) in surface sediments from Haihe River Basin were investigated. A method based on effects range (ER) was used to assess ecosystem risk of ∑PAHs (the total of 16 PAH) sensitively and accurately. The results indicated that ∑PAHs content levels ranged from 257 to 16901 μg·kg dry weight. The lower rings predominated in the samples, and 2-, 3-, 4-, 5- and 6-ring PAHs accounted for 12%, 21%, 30%, 30%, and 7% respectively in total PAHs. The ratio of Fl / (Fl+ Py) uniformly distributed in the interval 0.20–0.80, indicating that it may be affected by petroleum origin, oil combustion, biomass and coal combustion jointly. ∑PAHs in Cetian (S6), Dongwushi (S19), Handan (S20), Aixinzhuang (S21) and Tianjin (S37) exceeded effects range low (ERL), in which biologic effects were in a medium level with an adverse effect on biologic organisms. Thus, it is necessary to strengthen the PAHs monitoring and research of the Haihe River Basin.

Decentralized runtime enforcement for robotic swarms

摘要: s are usually designed in a bottom-up way, which can make s vulnerable to environmental impact. It is particularly true for the widely used control mode of s, where it is often the case that neither the correctness of the swarming tasks at the macro level nor the safety of the interaction among agents at the micro level can be guaranteed. To ensure that the behaviors are safe at runtime, it is necessary to take into account the property guard approaches for s in uncertain environments. is an approach which can guarantee the given properties in system execution and has no scalability issue. Although some methods have been studied and applied in different domains, they cannot effectively solve the problem of property enforcement on tasks at present. In this paper, an enforcement method is proposed on swarms which should satisfy multi-level properties in uncertain environments. We introduce a macro-micro property enforcing framework with the notion of agent shields and a discrete-time enforcing mechanism called -time enforcing. To realize this method, a domain specification language and the corresponding enforcer synthesis algorithms are developed. We then apply the approach to enforce the properties of the simulated in the robotflocksim platform. We evaluate and show the effectiveness of the method with experiments on specific unmanned aerial vehicle swarm tasks.

Zr66Al9Cu16Ni9块体非晶合金制备工艺研究

摘要:

运用真空电弧炉熔炼合金制备试样,采用三种不同的方法对金属熔体进行处理,试样的X射线衍射晶化相分析结果表明,B2O3和Na2SiO3的混合物与所选组分的熔体发生了反应,不适合作Zr基合金的包裹物;组分为CaF2(45%),AlF3(50%),CaO(5%)以及微量的TiO2的氟盐,由于该净化剂与所选的组分发生了反应,不适合Zr基合金的净化;设备的高真空度、原材料成分的高纯度是促进合金形成非晶态的重要因素。

Bacteria-mediated tumor-targeted delivery of tumstatin (54-132) significantly suppresses tumor growth in mouse model by inhibiting angiogenesis and promoting apoptosis

摘要: Tumor growth is an angiogenesis-dependent process and accompanied by the formation of hypoxic areas. Tumstatin is a tumor-specific angiogenesis inhibitor that suppresses the proliferation and induces the apoptosis of tumorous vascular endothelial cells. VNP20009, an attenuated Salmonella typhimurium strain, preferentially accumulates in the hypoxic areas of solid tumors. In this study, a novel Salmonella-mediated targeted expression system of tumstatin (VNP-Tum5) was developed under the control of the hypoxia-induced J23100 promoter to obtain anti-tumor efficacy in mice. Treatment with VNP-Tum5 effectively suppressed tumor growth and prolonged survival in the mouse model of B16F10 melanoma. VNP-Tum5 exhibited a higher efficacy in inhibiting the proliferation and inducing the necrosis and apoptosis of B16F10 cells in vitro and in vivo compared with VNP (control). VNP-Tum5 significantly inhibited the proliferation and migration of mouse umbilical vascular endothelial cells to impede angiogenesis. VNP-Tum5 downregulated the expression of anti-vascular endothelial growth factor A, platelet endothelial cell adhesion molecule-1, phosphorylated phosphoinositide 3 kinase, and phosphorylated protein kinase B and upregulated the expression of cleaved-caspase 3 in tumor tissues. This study is the first to use tumstatin-transformed VNP20009 as a tumor-targeted system for treatment of melanoma by combining anti-tumor and anti-angiogenic effects.

摘要: Abstract • Pig feces is the predominant excrement produced by animal husbandry in China. • The PF, Pig-1-BacTaqMan, and Pig-2-BacTaqMan MST assays showed better performance. • The pig-specific MST assays can contribute to managing the pig fecal pollution. In China, pig feces is the predominant source of excrement produced by animal husbandry. Improper use or direct discharge of pig feces can result in contamination of natural water systems. Microbial source tracking (MST) technology can identify the sources of fecal pollution in environmental water, and contribute to the management of pig fecal pollution by local environmental protection agencies. However, the accuracy of such assays can be context-dependent, and they have not been comprehensively evaluated under Chinese conditions. We aimed to compare the performance of five previously reported pig-specific MST assays (PF, Pig-Bac1SYBR, Pig-Bac2SYBR, Pig-1-BacTaqMan, and Pig-2-BacTaqMan, which are based on Bacteroidales 16S rRNA gene markers) and apply them in two rivers of North China. We collected a total of 173 fecal samples from pigs, cows, goats, chickens, humans, and horses across China. The PF assay optimized in this study showed outstanding qualitative performance and achieved 100% specificity and sensitivity. However, the two SYBR green qPCR assays (Pig-Bac1SYBR and Pig-Bac2SYBR) cross-reacted with most non-pig fecal samples. In contrast, both the Pig-1-BacTaqMan and Pig-2-BacTaqMan assays gave 100% specificity and sensitivity. Of these, the Pig-2-BacTaqMan assay showed higher reproducibility. Our results regarding the specificity of these pig-specific MST assays differ from those reported in Thailand, Japan, and America. Using the PF and Pig-2-BacTaqMan assays, a field test comparing the levels of pig fecal pollution in rivers near a pig farm before and after comprehensive environmental pollution governance indicated that pig fecal pollution was effectively controlled at this location.
