大菱鲆免疫球蛋白M(IgM)单克隆抗体的制备与特性鉴定

摘要: 小鼠骨髓瘤细胞NS0 与经IgM免疫的BALB/C小鼠脾细胞融合,经过反复有限稀释法克隆,筛选获得4 株抗大菱鲆IgM的单克隆抗体杂交瘤细胞株,分别为B1D1、D5C2、E1B2 和F4A1。经小鼠腹水扩大生产后,单抗效价为1∶1.024×106检测灵敏度为32 ng/mL。Western blot 分析表明,获得的单抗与IgM 重链区特异性结合。

摘要: The immune mapped protein 1 (NcIMP1) was identified as a membrane protein, and a previous study indicated that NcIMP1 could be a promising vaccine candidate against neosporosis. In this study, the immune response and protection efficacy of NcIMP1 were evaluated. The coding sequence of NcIMP1 was inserted into the eukaryotic expression vector pcDNA 3.1(+), resulting in the recombination plasmid pcDNA-IMP1, which was used for the intramuscular immunization of BALB/c mice. After immunization, the immune response was evaluated using a lymphoproliferative assay and cytokine and antibody measurements. Quantification of the cerebral parasite burden of mice challenged with 2 × 10 was performed 14 days after the last immunization. The results showed that the mice immunized with pcDNA-IMP1 developed a high level of specific antibody responses against recombinant NcIMP1, with a mixed IgG1/IgG2a response and a predominance of IgG2a production. The cellular immune response was associated with the production of IFN-γ, IL-2, IL-4 and IL-10 cytokines. The experiment was terminated 30 days p.i., and the cerebral parasite burden in each mouse was assessed by quantitative PCR. The parasite burden was significantly reduced in the pcDNA-IMP1-vaccinated mice. These data suggest that IMP1 is a promising vaccine candidate against neosporosis.

表面张力辅助去核法对小鼠卵母细胞的去核研究

摘要:

论述了一种新的小鼠卵母细胞去核方法一一表面张力辅助去核法(STA),并同时与其他3种去核方 法(简称A法、B法、C法)进行比较。经STA法去核的卵母细胞质体用于电融合介导的小鼠耳成纤维细胞核移植实验,结果有 76.1 %细胞与细胞质体融合形成重组胚,85.4 %的重组胚形成原核,49.4%卵裂形成2-细胞。

摘要: Abx)灌胃4周,明显改善了D-盐酸半乳糖胺/脂多糖[D-(+)-galactosamine hydrochloride/lipopolysaccharide, D-Gal/LPS]诱导的 ALF 小鼠的生存期RNA 测序显示,在Abx干预的ALF小鼠中,肝脏的炎症反应受到抑制,而整体代谢途径上调。此外,AhR靶向干预了影响了经Abx预处理或未经Abx预处理的ALF小鼠的预后,表明AhR通路在一定程度上调节了机体对ALF的易感性。

基于人工智能的肺癌NOG/PDX模型驱动基因匹配预测 Article

摘要:

患者源性肿瘤异种移植物(PDX)是癌症药物发现和筛查的有力工具。然而,目前的研究对PDX的基因型错配知之甚少,导致PDX使用过程中产生巨大的经济损失。在此,本研究建立了53 例肺癌患者的PDX模型,基因型匹配率为79.2%(42/53)。此外,检查了17 个临床病理学特征,并基于最低赤池信息量准则(AIC)、最小绝对收缩和选择算子(LASSO)-逻辑回归(LR)、支持向量机(SVM)递归特征消除(SVM-RFE)、极端梯度增强(XGBoost)、梯度增强和分类特征(CatBoost),以及合成少数过采样技术(SMOTE)输入逐步逻辑回归模型。最后,通过100 个试验组的准确度、受试者工作特征曲线下面积(AUC)和F1 评分评价所有模型的性能。两个多变量 LR 模型显示,年龄、驱动基因突变的数量、表皮生长因子受体(EGFR)基因突变、既往化疗的类型、既往酪氨酸激酶抑制剂(TKI)治疗和样本来源是强有力的预测因素。此外,CatBoost (平均精度= 0.960;平均AUC = 0.939;平均F1 分数= 0.908)和八特征SVM-RFE(平均精度= 0.950;平均AUC = 0.934;平均F1 分数= 0.903)在算法中表现出最好的性能。同时,除CatBoost 外,SMOTE的应用提高了大多数模型的预测能力。基于SMOTE,单一模型的集成分类器达到了最高的准确度(平均值= 0.975)、AUC(平均值= 0.949)和F1 评分(平均值= 0.938)。总之,本文建立了一个最佳预测模型来筛选肺癌患者的NOD/Shi-scid白细胞介素-2受体(IL-2R) γnull(NOG)/PDX模型,并为建立预测模型提供了一种通用方法。

Valveless pumping using a two-stage impedance pump

Impedance pump is defined as a type of valveless pumping mechanism, where an elastic tube is joined with a tube of different impedance, a periodic asymmetrical compression on part of the elastic tube will produce a unidirectional flow. Numerical and experimental studies had been conducted in the past, but little experimental work was found to exploit the principle of multi-stage impedance pumping for fluid transportation. This paper concentrates on the fluid behavior for an open-loop two-stage system experimentally. The aim of this work is to have an insight on the deliverable head and pumping rate of an open-loop two-stage impedance pumping system. This paper generally investigates the effect of different parameters towards the generation of unidirectional flows and composite flow of two opposite directions. Results obtained are of significant values.

摘要: butterfly wings were replicated by a sol-gel method and a calcination process, which could take advantage of the spatial features of the wing to enhance their photocatalytic properties. Hierarchical structures of -carbon-TiO (PP-C-TiO ) were confirmed by SEM observations. By applying the Brunauer-Emmett-Teller method, it was concluded that in the presence of wings the product shows higher surface area with respect to the pure TiO made in the absence of the wings. The higher specific surface area is also beneficial for the improvement of photocatalytic property. Furthermore, the conduction and valence bands of the PP-C-TiO are more negative than the corresponding bands of pure TiO , allowing the electrons to migrate from the valence band to the conduction band upon absorbing visible light. That is, the presence of C originating from wings in the PP-C-TiO could extend the photoresponsiveness to visible light. This strategy provides a simple method to fabricate a high-performance photocatalyst, which enables the simultaneous control of the morphology and carbon element doping.

Antithrombin deficiency and decreased protein C activity in a young man with venous thromboembolism:

Antithrombin and protein C are two crucial members in the anticoagulant system and play important roles in hemostasis. Mutations in and lead to deficiency or dysfunction of the two proteins, which could result in venous thromboembolism (VTE). Here, we report a Chinese 22-year-old young man who developed recurrent and serious VTE in cerebral veins, visceral veins, and deep veins of the lower extremity. Laboratory tests and direct sequencing of and were conducted for the patient and his family members. Coagulation tests revealed that the patient presented type I antithrombin deficiency combined with decreased protein C activity resulting from a small insertion mutation c.848_849insGATGT in and a short deletion variant c.572_574delAGA in . This combination of the two mutations was absent in 400 healthy subjects each from southern and northern China. Then, we summarized all the mutations of the and gene reported in the Chinese Han population. This study demonstrates that the combination of antithrombin deficiency and decreased protein C activity can result in severe VTE and that the coexistence of different genetic factors may increase the risk of VTE.

Atomistic simulations of plasma catalytic processes

摘要: There is currently a growing interest in the realisation and optimization of hybrid plasma/catalyst systems for a multitude of applications, ranging from nanotechnology to environmental chemistry. In spite of this interest, there is, however, a lack in fundamental understanding of the underlying processes in such systems. While a lot of experimental research is already being carried out to gain this understanding, only recently the first simulations have appeared in the literature. In this contribution, an overview is presented on atomic scale simulations of plasma catalytic processes as carried out in our group. In particular, this contribution focusses on plasma-assisted catalyzed carbon nanostructure growth, and plasma catalysis for greenhouse gas conversion. Attention is paid to what can routinely be done, and where challenges persist.

Elevated C-reactive protein levels predict worsening prognosis in Chinese patients with first-onset stroke

摘要: The role of high sensitivity C-reactive protein (hsCRP) in predicting prognosis after stroke in the Asian population has not been investigated. We hypothesized that elevated levels of hsCRP were associated with worsening prognosis after stroke in Chinese patients. Two hundred and ninety consecutive patients with first-onset stroke and 290 age- and gender-matched control subjects without any cerebrovascular disease were enrolled for study. Plasma hsCRP level was detected and subsequent vascular events and death were recorded in both groups over a 5-year period. Compared to control group, patients presenting with stroke had higher plasma hsCRP level (3.3 ± 3.8 1.3 ± 2.2 mg/L, < 0.01). Furthermore, in the group of patients with stroke, the mean plasma hsCRP level was higher in patients who developed subsequent vascular diseases or died as compared with the patients without further complications (4.4 ± 4.3 2.7 ± 3.3 mg/L, < 0.01). Compared to the lowest tertile of hsCRP level, the relative risk for vascular events or death in stroke patients was 2.91 in the highest tertile of hsCRP (95% CI, 1.54–5.50, = 0.001). This increase in relative risk for vascular events or death in stroke patients continued after adjustment for age, sex and other cardiovascular risk factors such as hypertension and diabetes ( : 2.771, 95% CI: 1.367–5.617, = 0.005). These findings indicate that increased hsCRP level is associated with worsening prognosis after stroke in Chinese patients and suggests that inflammation is correlated with stroke outcome.

A simple, azulene-based colorimetric probe for the detection of nitrite in water

摘要: We describe the synthesis and evaluation of an azulene-based chemodosimeter for nitrite. The probe was found to undergo two distinct color changes upon introduction aqueous nitrite ion. A near-instant formation of a grey color provides a qualitative indication of the presence of nitrite, followed by the formation of a deep-yellow/orange color, the endpoint from which quantitative data can be derived. The azulene probe exhibits 1:1 stoichiometry of reaction with nitrite in water, and is selective for nitrite over other anions. The azulene probe was applied to determine nitrite content in cured meat, and compared with the British Standard testing procedure (Griess test). The value obtained from the azulene-based probe agreed closely with the standard test. Our procedure only requires the preparation of one standard solution, instead of the three required for the standard Griess test.
