快中子诱发U251神经胶质瘤细胞的生物效应研究

摘要:

目的:以14 MeV快中子照射体外培养的U251(人脑胶质瘤) 细胞,研究快中子诱发U251的相关生物效应;方法:快中子照射U251细胞,吸收剂量分别为1、3、5、7 Gy,辐照后检测细胞的存活率、凋亡率、生长曲线以及周期变化等相关指标;结果:随着吸收剂量的增加,细胞存活率下降,凋亡率上升,呈良好的剂量-效应关系。照射后24 h,细胞生长曲线无明显变化;照射后48 h,细胞增殖速度明显变缓;结论:快中子为高LET射线,与相关文献报道的γ射线相比,快中子具有较高的相对生物效应。

A total of 251 patients with epithelial ovarian cancer (EOC) treated between 2002 and 2008 was retrospectively analyzed to investigate the long-term outcomes and prognostic factors of these patients, particularly those who underwent primary debulking surgery followed by platinum-based chemotherapy. Clinico-pathological parameters, including progression-free survival (PFS) and overall survival (OS), were also analyzed. The median follow-up period from the end of initial treatment to June 2010 was 58 months. The three-year PFS rate was 61.7% for International Federation of Gynecology and Obstetrics (FIGO) I–II, 19.9% for FIGO III–IV, and 33.9% for all stages. By comparison, the five-year PFS rate was 44.6% for FIGO I–II, 17.7% for FIGO III–IV, and 28.3% for all stages. The three-year OS rate was 67.9% for FIGO I–II, 41.7% for FIGO III–IV, and 50.2% for all stages. The five-year OS rate was 52.7% for FIGO I–II, 30.8% for FIGO III–IV, and 39.2% for all stages. Univariate analysis revealed that advanced FIGO stage, serum CA125, and suboptimal debulking were significant factors affecting PFS and OS. In multivariate analysis, PFS was significantly influenced by FIGO stage and suboptimal debulking. However, OS was significantly influenced by advanced FIGO stage only. Our study confirms the efficacy of surgery followed by platinum-based chemotherapy for EOC. FIGO stage is considered as one of the most reliable predictors of the prognosis of patients with EOC.

Magnetic KIT-6 nano-composite and its amino derivatives as convenient adsorbent for U(VI) sequestration

摘要: Although mesoporous silica with magnetically hybridized two-dimensional channel structures has been well studied in recent years, it remains a challenge to fabricate the counterpart with macroporous three-dimensional cubic structures since the highly acidic preparation conditions lead to dissolution of magnetic particles. Herein, we successfully prepared magnetic KIT-6 nano-composite and its amino derivatives by bearing acid-resistant iron oxide. The prepared materials exhibited excellent properties for U(VI) ions removal from aqueous solutions under various conditions. The experimental data show that the U(VI) adsorption features fast adsorption kinetics, high adsorption capacity and ideal selectivity toward U(VI). The adsorption process is of spontaneous and endothermic nature and ionic strength independence, and the adsorbents can be easily regenerated by acid treatment. Compared to pristine KIT-6, the introduction of magnetism does not reduce the efficiency of the material to remove U(VI) while exerting its role as a recovery adsorbent. The findings of this work further demonstrate the potential broad application prospects of magnetic hybrid mesoporous silica in radionuclide chelation.

A building-based data capture and data mining technique for air quality assessment

摘要: Recently, a building-based air quality model system which can predict air quality in front of individual buildings along both sides of a road has been developed. Using the Macau Peninsula as a case study, this paper shows the advantages of building-based model system in data capture and data mining. Compared with the traditional grid-based model systems with input/output spatial resolutions of 1–2 km, the building-based approach can extract the street configuration and traffic data building by building and therefore, can capture the complex spatial variation of traffic emission, urban geometry, and air pollution. The non-homogeneous distribution of air pollution in the Macau Peninsula was modeled in a high-spatial resolution of 319 receptors·km . The spatial relationship among air quality, traffic flow, and urban geometry in the historic urban area is investigated. The study shows that the building-based approach may open an innovative methodology in data mining of urban spatial data for environmental assessment. The results are particularly useful to urban planners when they need to consider the influences of urban form on street environment.

Is there an inverted U-shaped curve?

摘要: As the largest contributor to water impairment, agriculture-related pollution has attracted the attention of scientists as well as policy makers, and quantitative information is being sought to focus and advance the policy debate. This study applies the panel unit root, heterogeneous panel cointegration, and panel-based dynamic ordinary least squares to investigate the Environmental Kuznets Curve on environmental issues resulting from use of agricultural synthetic fertilizer, pesticide, and film for 31 provincial economies in mainland China from 1989 to 2009. The empirical results indicate a positive long-run co-integrated relationship between the environmental index and real GDP per capita. This relationship takes on the inverted U-shaped Environmental Kuznets Curve, and the value of the turning point is approximately 10,000–13,000, 85,000–89,000 and over 160,000 CNY, for synthetic fertilizer nitrogen indicator, fertilizer phosphorus indicator and pesticide indicator, respectively. At present, China is subject to tremendous environmental pressure and should assign more importance to special agriculture-related environmental issues.

Effect of end-to-end invagination pancreaticojejunostomy with circle discontinuous U suture in pancreatic

摘要: The aim of this paper is to summarize the methods of pancreaticojejunostomy in the pancreatic operation and to study the safety and feasibility of a new operative method called end-to-end invagination pancreaticojejunostomy with circle discontinuous U suture to prevent fistula of pancreaticojejunostomy. Eight-three patients with pancreaticoduodenectomy in the 3rd Hospital, Jilin University from 2001 January to 2006 April were reviewed. The incidences of pancreatic fistula with different types of pancreaticojejunostomy were compared. The overall incidence rate of pancreatic fistula was 26.5% (22/83). No pancreatic fistula occurred in end-to-end invagination pancreaticojejunostomy with circle discontinuous U suture. The incidence rate of the fistula following end-to-end invagination pancreaticojejunostomy with circle discontinuous U suture was significantly lower than that of traditional end-to-end pancreaticojejunostomy [40%, (10/25), <0.01] and end-to-side pancreaticojejunostomy [27.3%, (12/44), <0.05], but no significant difference (>0.05) between traditional end-to-end pancreaticojejunostomy and end-to-side pancreaticojejunostomy was discovered. End-to-end invagination pancreaticojejunostomy with circle discontinuous U suture has a definite effect on avoiding pancreatic fistula following pancreaticojejunostomy and is worth being recommended. But the cases were limited, so this method would still need to be observed and confirmed further in the future.

Exploring adoption of augmented reality smart glasses: Applications in the medical industry

摘要: This study explores the use of augmented reality smart glasses (ARSGs) by physicians and their adoption of these products in the Turkish medical industry. Google Glass was used as a demonstrative example for the introduction of ARSGs. We proposed an exploratory model based on the technology acceptance model by Davis. Exogenous factors in the model were defined by performing semi-structured in-depth interviews, along with the use of an expert panel in addition to the technology adoption literature. The framework was tested by means of a field study, data was collected via an Internet survey, and path analysis was used. The results indicate that there were a number of factors to be considered in order to understand ARSG adoption by physicians. Usefulness was influenced by ease of use, compatibility, ease of reminding, and speech recognition, while ease of use was affected by ease of learning, ease of medical education, external influence, and privacy. Privacy was the only negative factor that reduced the perceived ease of use, and was found to indirectly create a negative attitude. Compatibility emerged as the most significant external factor for usefulness. Developers of ARSGs should pay attention to healthcare-specific requirements for improved utilization and more extensive adoption of ARSGs in healthcare settings. In particular, they should focus on how to increase the compatibility of ARSGs. Further research needs to be conducted to explain the adoption intention of physicians.

诱导性多潜能干细胞(iPS)研究:现状与展望

摘要:

通过特定的基因组合与转染可以将已分化的体细胞诱导重编程为多潜能干细胞(iPS),是近年来干细胞研究领域最令人瞩目的一项新的干细胞制造技术。与胚胎干细胞(ES)不同,iPS细胞的制造不需要毁损胚胎,因而不会涉及更多的伦理学问题。iPS的出现不仅为体细胞重编程去分化机制的研究注入了新的活力,而且为疾病发生发展相关机制研究与特异的细胞治疗,特别是再生医学带来新的曙光。

Causality between energy consumption and GDP in the U.S.: evidence from wavelet analysis

摘要: This study investigates the dynamic causal relationship between energy consumption and economic growth in the U.S. at different time scales. The main novelty of the study is that this paper complements the existing studies on the nexus between energy consumption and economic growth by employing the wavelet transformation to obtain different time scales in order to investigate causality between energy consumption and economic growth. This method is first developed by Ramsey and Lampart. Their approach consists of first decomposing the series into time scales by wavelet filters and testing causality of each time scale with the pertinent time scale of the other series separately. The data span from 1973q1 to 2012q1 on a quarterly basis. The main empirical insight is that the causal relationship is stronger at finer time scales, whereas the relationship is less and less apparent at longer time horizons. The results indicate that energy consumption causes economic growth, while the reverse is not true at the original frequency of the data. At the very finest scale the same result arises. However, at coarser scales feedback is observed. In particular, at intermediate time scales the evidence indicates that energy consumption causes economic growth, while the reverse is also true. These empirical findings are expected to be of high importance in terms of the effective design and implementation of energy and environmental policies, especially when a number of countries in the pursuit of high economic growth targets do not pay any serious attention on environmental issues.

Effect of arsenic trioxide on proliferation and apoptosis of U266 cells and its relationship with the

摘要: The aim of this article is to explore the effect of arsenic trioxide (AsO) on the proliferation and apoptosis of myeloma cell line U266 and its relationship with the expression variation of vascular endothelial growth factor (VEGF). The viability and apoptosis of U266 cells were observed by methylthiazolyl- tetrazolium (MTT) assay and terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL). The effect of AsO on the VEGF expression of U266 cells were tested by enzyme linked immunosorbent assay (ELISA) and reverse transcription-polymerase chain reaction (RT-PCR) analysis. We found that AsO could significantly inhibit the growth of U266 cells, and the concentration for 50% growth inhibition (IC) was 2 ?mol/L. After treatment with 2, 5, 10 ?mol/L AsO for 36 hours, dose-dependent apoptosis of U266 cells was observed. After treatment with 2, 5, 10 ?mol/L AsO for 72 hours, a dose-dependent reduction of VEGF in the supernatant of U266 cells culture was found. As far as single cells are concerned, nevertheless, the expression of VEGF mRNA did not vary. So we draw the conclusion that AsO could induce the apoptosis of U266 cells and inhibit their proliferation, decrease the tumor load, and lead to the reduction of VEGF in the culture supernatant, but not change the expression of VEGF in single U266 cells.

诱导性多能干细胞(iPSC)在许多方面,如再生医学和药物筛选方面,是理想的细胞来源,其开发前景广阔但是,目前还没有一种有效的大规模生产系统来确保诱导性多能干细胞的稳定供应。本文综述了近年来为了实现诱导性多能干细胞的稳定生产所采取的方法。本文首先讨论了诱导性多能干细胞培养过程中需要控制的因素,如养分供应、废物清除和氧气供应。然后,介绍了基于黏附、悬浮和支架的诱导性多能干细胞培养系统的最新研究。本文还讨论了细胞培养阶段之后进行的过程,如灌装和冻存过程,由于细胞悬浮液在超低温保存培养基中存活率降低,所以其生产规模受到了限制。本文表明为了获得最佳且稳定的大规模生产诱导性多能干细胞系统,运用多学科研究手段对研究影响细胞生长和质量的复杂因素是至关重要的。

摘要: 细胞核移植技术是动物克隆的核心,其操作过程非常复杂,不可避免地造成细胞内损伤。通常来说,只有不到1%的重构胚胎能够发育成克隆动物,克隆的低成功率是制约其广泛应用的主要原因之一。为此,本文提
出了一种基于细胞内应变评估的卵母细胞去核方法,以减少体细胞核移植中的潜在细胞内损伤,从而提高克隆成功率。首先,根据细胞内速度场计算细胞内应变,进而将内应变作为评价标准来改进去核操作;在此基础上,开发了一套机器人批量体细胞核移植系统,用于实现机器人化克隆。实验结果表明,本文方法将体细胞核移植后的胚胎囊胚率从10.0%提高到20.8%,并成功获得17 头克隆小猪。这是世界上首批由机器人完成克隆操作获得的克隆动物。与此同时,基于细胞内应变的去核方法有望应用于其他生物操作,并建立一种通用的减少细胞内损伤的细胞操作规范。
