Abstract:

Transcription factor networks have evolved in order to control, coordinate, and separate, the functions of distinct network modules spatially and temporally. In this review we focus on the MYC network (also known as the MAX-MLX Network), a highly conserved super-family of related basic-helix-loop-helix-zipper (bHLHZ) proteins that functions to integrate extracellular and intracellular signals and modulate global gene expression. Importantly the MYC network has been shown to be deeply involved in a broad spectrum of human and other animal cancers. Here we summarize molecular and biological properties of the network modules with emphasis on functional interactions among network members. We suggest that these network interactions serve to modulate growth and metabolism at the transcriptional level in order to balance nutrient demand with supply, to maintain growth homeostasis, and to influence cell fate. Moreover, oncogenic activation of MYC and/or loss of a MYC antagonist, results in an imbalance in the activity of the network as a whole, leading to tumor initiation, progression and maintenance.

Abstract:

Finite element models are often used to simulate impact and contact dynamics responses of multibody dynamical systems. However, such a simulation remains very inefficient because very small integration time step must be used when solving the involved differential equations, especially when the involved contact stiffness is high. Although many model reduction techniques have been available to improve the efficiency of finite element based simulations, these techniques cannot be readily applied to contact dynamics simulations due to the high nonlinearity of the contact dynamics models. This paper presents a model reduction approach for finite-element based multibody contact dynamics simulation, based on a modified Lyapunov balanced truncation method. An example is presented to demonstrate that, by applying the model reduction the simulation process is significantly speeded up and the resulting error is bounded within an acceptable level. The performance of the method with respect to some influential factors such as element size, shape and contact stiffness is also investigated.

Abstract:

The funding gap of public infrastructure networks (roads, railways, ports, electricity, and energy lines) can be solved partly by introducing private capital for investments, i.e., public-private partnerships (PPP). This paper introduces an integrated model of a PPP project and investigates its implications on PPP policies and strategies regarding appropriate project appraisal and selection. The model has different resolution levels, namely, project level, business ecosystem level, and market and societal levels. The integrated model suggests that investing in merely financially viable projects is insufficient to realize economically and socially sustainable and acceptable projects.

On Balancing Mechanism Between Supply of Medical Education and Demand for Clinicians in China

Abstract:

The imbalance between supply and demand in terms of talent cultivation scale, structure, and quality is one of the key issues facing the cultivation of medical personnel in China. The supply of medical talents in China cannot meet the growing demand for health, the quality of personnel training is uneven, and the talent supply structure does not match social needs. Through the research and discussion on the forms and contents of medical education in the United States and the United Kingdom, this paper proposes the enlightenment on how to maintain the balance between supply and demand of medical education in China and how to improve the efficiency of medical training. It is suggested to gradually establish a talent supply and demand balance mechanism where the number and structure of post-graduate education posts determines the enrollment scale and structure of medical colleges and universities, and establish a dynamic monitoring and early warning mechanism for the demand for medical talents. Furthermore, medical personnel training requires cross-departmental macro-coordination and regulation, and quality requirements such as a high threshold for doctors should be set to limit the cultivation scale of medical talents.

Artificial Bear Bile: A Novel Approach to Balancing Medical Requirements and Animal Welfare

Abstract: Bear bile has been a valuable and effective medicinal material in traditional Chinese medicine (TCM) for over 13 centuries. However, the current practice of obtaining it through bear farming is under scrutiny for its adverse impact on bear welfare. Here, we present a new approach for creating artificial bear bile (ABB) as a high-quality and sustainable alternative to natural bear bile. This study addresses the scientific challenges of creating bear bile alternatives through interdisciplinary collaborations across various fields, including resources, chemistry, biology, medicine, pharmacology, and TCM. A comprehensive efficacy assessment system that bridges the gap between TCM and modern medical terminology has been established, allowing for the systematic screening of therapeutic constituents. Through the utilization of chemical synthesis and enzyme engineering technologies, our research has achieved the environmentally friendly, large-scale production of bear bile therapeutic compounds, as well as the optimization and recomposition of ABB formulations. The resulting ABB not only closely resembles natural bear bile in its composition but also offers advantages such as consistent product quality, availability of raw materials, and independence from threatened or wild resources. Comprehensive preclinical efficacy evaluations have demonstrated the equivalence of the therapeutic effects from ABB and those from commercially available drained bear bile (DBB). Furthermore, preclinical toxicological assessment and phase Ⅰ clinical trials show that the safety of ABB is on par with that of the currently used DBB. This innovative strategy can serve as a new research paradigm for developing alternatives for other endangered TCMs, thereby strengthening the integrity and sustainability of TCM.
