摘要:

S100A9, a calcium-binding protein, participates in the inflammatory process and development of various tumors, thus attracting much attention in the field of cancer biology. This study aimed to investigate the regulatory mechanism of and its function involvement in APL. We used real-time quantitative PCR to determine whether PML/RARα affects the expression of in NB4 and PR9 cells upon ATRA treatment. ChIP-based PCR and dual-luciferase reporter assay system were used to detect how PML/RARα and PU.1 regulate promoter activity. CCK-8 assay and flow cytometry were employed to observe the viability and apoptosis of NB4 cells when was overexpressed. Results showed that was an ATRA-responsive gene, and PML/RARα was necessary for the ATRA-induced expression of in APL cells. In addition, PU.1 could bind to the promoter of , especially when treated with ATRA in NB4 cells, and promote its activity. More importantly, overexpression of induced the apoptosis of NB4 cells and inhibited cell growth. Collectively, our data indicated that PML/RARα and PU.1 were necessary for the ATRA-induced expression of in APL cells. Furthermore, promoted apoptosis in APL cells and affected cell growth.

Insight into the role of cholesterol in modulation of morphology and mechanical properties of CHO-K1

Cholesterol plays a significant role in the organization of lipids and modulation of membrane dynamics in mammalian cells. However, the effect of cholesterol depletion on the eukaryotic cell membranes seems controversial. In this study, the effects of cholesterol on the topography and mechanical behaviors of CHO-K1 cells with manipulated membrane cholesterol contents were investigated by atomic force microscopy (AFM) technique. Here, we found that the depletion of cholesterol in cell membranes could increase the membrane stiffness, reduce the cell height as well as promote cell retraction and detachment from the surface, whereas the cholesterol restoration could reverse the effect of cholesterol depletion on the membrane stiffness. Increased methyl-β-cyclodextrin levels and incubation time could significantly increase Young’s modulus and degree of stiffing on cell membrane and cytoskeleton. This research demonstratede importance of cholesterol in regulating the dynamics of cytoskeleton-mediated processes. AFM technique offers excellent advantages in the dynamic monitoring of the change in membranes mechanical properties and behaviors during the imaging process. This promising technology can be utilized in studying the membrane properties and elucidating the underlying mechanism of distinct cells in the near-native environment.

摘要: China s accelerator-driven sub-critical system (ADS) sub-critical experimental assembly Venus-1 and the preliminary experiment is presented. The core of Venus-1 is a coupled one of a fast neutron zone and a thermal neutron zone. The fast neutron zone is at the centre of the core and formed by natural uranium fuel. A fast neutron spectrum field can be produced in the fast neutron zone and used for the transmutation of minor actinides (MAs). The thermal neutron zone surrounds the fast neutron zone and is formed by low-enriched uranium fuel. It is a fission zone. An epithermal neutron zone between the fast neutron zone and the thermal neutron zone can be established for the transmutation of long-lived fission products (LLFP). On July 18, 2005, the first fuel element was loaded into the Venus-1 sub-critical assembly and some preliminary experiments about the sub-critical neutronics were performed. The Venus-1 can be driven by an Am-Be source or other steady neutron source (Cf-252, D-D reaction and D-T reaction) to study the effect of the external neutron source with different energies or a D-T pulsed neutron source on the dynamic characteristics.

基于专家示教聚类经验池的高效深度强化学习 Research Article

摘要: 作为强化学习领域最基本的主题之一,样本效率对于深度强化学习算法的部署至关重要。与现有大多数从不同类型的后验分布中对动作进行采样的探索方法不同,我们专注于策略的采样过程,提出一种有效的选择性采样方法,通过对环境的内部层次结构建模来提高样本效率。具体来说,首先在策略采样过程中使用聚类方法生成动作候选集,随后引入一个用于对内部层次结构建模的聚类缓冲区,它由同轨数据、异轨数据以及专家数据组成,用于评估探索阶段动作候选集中不同类别动作的价值。通过这种方式,我们的方法能够更多地利用专家示教数据中的监督信息。在6种不同的连续运动环境中进行了实验,结果表明选择性采样方法具有卓越的强化学习性能和更快的收敛速度。特别地,在LGSVL任务中,该方法可以减少46.7%的收敛步数和28.5%的收敛时间。代码已开源,见https://github.com/Shihwin/SelectiveSampling。

Association of miRNA-122-binding site polymorphism at the interleukin-1 α gene and its interaction with

This study was designed to investigate the contribution of miRNA-122-binding site polymorphism at the IL-1A gene and its multiplicative interactions with hepatitis B virus (HBV) mutations in the risk of hepatocellular carcinoma (HCC). A total of 1021 healthy controls, 302 HBV surface antigen (HBsAg) seroclearance subjects, and 2011 HBsAg-positive subjects (including 1021 HCC patients) were enrolled in this study. Quantitative PCR was used to genotype rs3783553. HBV mutations were determined by direct sequencing. Multivariate logistic regression analyses were performed to test the associations of rs3783553, mutations, and their interactions with the risk of HCC. No significant association was found between rs3783553 and the risk of HCC among healthy controls, HBsAg seroclearance subjects, HBsAg-positive subjects without HCC, and all controls. Additionally, rs3783553 was not significantly associated with chronic HBV infection, liver cirrhosis, HBV e antigen seroconversion, abnormal alanine aminotransferase, and high viral load (>104 copies/ml). However, the TTCA insertion allele of rs3783553 was significantly associated with an increased frequency of HBV C7A mutation compared with homozygous TTCA deletion carriers [(del/ins+ ins/ins) vs. del/del, adjusted odds ratio (OR)=1.48, 95% confidence interval (CI)=1.09-2.02, P=0.013]. Multiplicative interaction of rs3783553 with HBV preS deletion significantly reduced the risk of HCC in males, with an adjusted OR of 0.64 (95% CI=0.42-0.98; P=0.041) after age and HBV genotype were adjusted. Although rs3783553 did not significantly affect genetic susceptibility to HBV-related HCC, its variant allele may predispose the host to selecting HBV C7A mutation during evolution and significantly reduce the risk of HCC caused by HBV preS deletion. This study provides an insight into the complex host-virus interaction in HBV-induced hepatocarcinogenesis and is helpful in determining HBsAg-positive subjects who are likely to develop HCC.

自监督脓毒症治疗推荐算法 Research Articles

摘要: 由于每个脓毒症患者治疗反应可能不同,为病人提供量身定制的治疗建议来帮助医生有效、准确地做出决定,并采取有效治疗方案,是降低医院重症监护病房死亡率的一项极具挑战性的工作。本文将强化学习应用于个人治疗推荐,采用对样本不确定性进行建模并评估的方法,根据患者对治疗的反应和状态,将患者样本分为两个域,然后使用辅助迁移学习任务重建两个域的样本,使用特权学习的蒸馏方法与用于迁移学习的变分自动编码器框架关联低质量域和高质量域间的任务。通过结合自监督方式获得更好的状态和动作表示,本文提出一种针对引起较高风险的不确定性进行控制的深度强化学习方法;模型提供一定的灵活性使之可以在不同场景对模糊样本做出保守预测或明确判断,并降低预期死亡率。在大规模公开可用的真实医疗数据集MIMIC-III上的实验表明,所提模型将总体估计死亡率降低了2.3%,并将主要估计死亡率降低到9.5%。

摘要: 提出一种新颖的分数阶记忆性神经突触加权电路。与以往大多数整数阶神经突触加权电路不同,该分数阶记忆性神经突触加权电路具有许多重要优点。由于忆阻的概念已从经典的整数阶忆阻推广到分数阶忆阻(分忆抗),分忆抗能否实现分数阶记忆性突触成为一个具有挑战性的理论问题。本文研究利用脉冲分忆抗桥电路实现的分数阶记忆性神经突触加权电路的特点。首先,利用基于脉冲的分忆抗桥电路设计分数阶记忆性神经突触加权电路结构。其次,从数学上证明分数阶记忆性神经突触加权电路的分数阶学习能力。最后,通过实验研究分数阶记忆性神经突触加权电路的电特性。分数阶记忆性神经突触加权电路在解释学习和记忆基础的细胞机制方面具有很强的能力,优于传统的整数阶神经突触加权电路,是该电路的主要优势。

格上高效的身份基签名

摘要: 随着电子信息技术的飞速发展,数字签名已成为人们生活中不可或缺的一部分。由于证书管理的局限性,传统的公钥证书密码系统无法满足现有需求。基于身份的密码系统避免了证书管理问题。量子计算机的发展给传统密码学带来严峻挑战。后量子密码学研究势在必行。目前,几乎所有后量子基于身份的签名方案都是利用高斯采样技术或陷门技术构建的。但是,这两种技术对计算效率有很大影响。为克服该问题,采用Lyubashevsky签名方案构造了格上基于身份的签名方案。基于格上的最短向量问题,该方案既不使用高斯采样技术也不使用陷门技术。在随机谕言机模型中,可以证明该方案对适应性选择的消息和身份攻击是不可伪造的。其安全性级别是强不可伪造的,比其他方案存在性不可伪造的安全性更高。与其他有效方案相比,所提方案在计算复杂度和安全性方面具有优势。

New Challenges, New Opportunities and New Measures for Oil and Gas Cooperation in the Belt and Road

摘要: First-mover advantage and solid foundation of oil and gas cooperation is very important to the Belt and Road Initiative. Deeply studied opportunities and challenges of oil and gas cooperation have far-reaching significance to the cooperation in other industries in the Belt and Road. In this article, based on the systematic analysis of the oil and gas supply security, investment environment, regional win-win situation in oil and gas cooperation, new opportunities and challenges about upstream, pipeline, refining, trade, warehousing, engineering technology and equipment are studied. Enhancing capacities in four areas and implementing the 16 measures are given for deepening oil and gas cooperation in the future. Meanwhile, this article also gives suggestions and cooperation directions for countries and companies.

Industrial robots are becoming increasingly vulnerable to cyber incidents and attacks, particularly with the dawn of the Industrial Internet-of-Things (IIoT). To gain a comprehensive understanding of these cyber risks, vulnerabilities of industrial robots were analyzed empirically, using more than three million communication packets collected with testbeds of two ABB IRB120 robots and five other robots from various Original Equipment Manufacturers (OEMs). This analysis, guided by the confidentiality–integrity–availability (CIA) triad, uncovers robot vulnerabilities in three dimensions: confidentiality, integrity, and availability. These vulnerabilities were used to design Covering Robot Manipulation via Data Deception (CORMAND2), an automated cyber–physical attack against industrial robots. CORMAND2 manipulates robot operation while deceiving the Supervisory Control and Data Acquisition (SCADA) system that the robot is operating normally by modifying the robot’s movement data and data deception. CORMAND2 and its capability of degrading the manufacturing was validated experimentally using the aforementioned seven robots from six different OEMs. CORMAND2 unveils the limitations of existing anomaly detection systems, more specifically the assumption of the authenticity of SCADA-received movement data, to which we propose mitigations for.

Experimental study of nitrite accumulation in pre-denitrification biological nitrogen removal process

摘要: The effect of dissolved oxygen (DO) concentration on nitrite accumulation was investigated in a pilot-scale pre-denitrification process at room temperature for 100 days. In the first 10 days, due to the instability of the system, the DO concentration fluctuated between 1.0 and 2.0 mg/L. In the next 14 days, the DO concentration was kept at 0.5 mg/L and nitrite accumulation occurred, with the average nitrite accumulation rate at 91%. From the 25th day, the DO concentration was increased to 2.0 mg/L to destroy the nitrite accumulation, but nitrite accumulation rate was still as high as 90%. From the 38th day the nitrite accumulation rate decreased to 15%–30% linearly. From the 50th day, DO concentration was decreased to 0.5 mg/L to resume nitrite accumulation. Until the 83rd day the nitrite accumulation rate began to increase to 80%. Dissolved oxygen was the main cause of nitrite accumulation, taking into account other factors such as pH, free ammonia concentration, temperature, and sludge retention time. Because of the different affinity for oxygen between nitrite oxidizing bacteria and ammonia oxidizing bacteria when DO concentration was kept at 0.5 mg/L, nitrite accumulation occurred.

摘要: 拓扑绝缘体Sb2Te3具有从可见光到红外波段的宽谱可饱和吸收的优点。在本研究中,首先将拓扑绝缘体家族中的二维材料Sb2Te3可饱和吸收体应用在宽谱可调谐被动调Q掺镱光纤激光器中。高质量的Sb2Te3晶体通过选区熔炼法合成,进一步通过改进的机械剥离方法制备少层Sb2Te3可饱和吸收体。基于此可饱和吸收体,在掺镱光纤环形腔中获得稳定的波长可调谐被动调Q脉冲,其中心波长可从1040.89 nm连续调节到1092.85 nm。实验结果表明,Sb2Te3具有宽谱可饱和吸收特性,这个波长可调谐脉冲激光可以为实际应用提供一个方便简单的光源。
