RNA m6A modification and its function in diseases

N6-methyladenosine (m6A) is the most common post-transcriptional RNA modification throughout the transcriptome, affecting fundamental aspects of RNA metabolism. m6A modification could be installed by m6A “writers” composed of core catalytic components (METTL3/METTL14/WTAP) and newly defined regulators and removed by m6A “erasers” (FTO and ALKBH5). The function of m6A is executed by m6A “readers” that bind to m6A directly (YTH domain-containing proteins, eIF3 and IGF2BPs) or indirectly (HNRNPA2B1). In the past few years, advances in m6A modulators (“writers,” “erasers,” and “readers”) have remarkably renewed our understanding of the function and regulation of m6A in different cells under normal or disease conditions. However, the mechanism and the regulatory network of m6A are still largely unknown. Moreover, investigations of the m6A physiological roles in human diseases are limited. In this review, we summarize the recent advances in m6A research and highlight the functional relevance and importance of m6A modification in in vitro cell lines, in physiological contexts, and in cancers.

Childhood obesity and obesity-related metabolic complications are induced by a high-fat postnatal diet. The lack of a suitable animal model, however, remains a considerable challenge in obesity studies. In the current study, we provided high-fat diet (HFD) to dams during lactation and to pups after weaning. We also developed a novel model of C57BL/6J mouse pups with HFD-induced postnatal obesity. Results showed that feeding with HFD induces fat deposition and obesity in pups. Furthermore, HFD more potently increased the body weight (BW) of male than female pups. HFD-fed female pups were obese, underwent precocious puberty, and showed increased kisspeptin expression in the hypothalamus. However, parental obesity and precocious puberty exerted no synergistic effects on the HFD-induced postnatal weight gain and puberty onset of the pups. Interestingly, some HFD-fed litters with normal BW also exhibited precocious puberty. This finding suggested that diet composition but not BW triggers puberty onset. Our model suggests good construction validity of obesity and precocious puberty. Furthermore, our model can also be used to explore the mutual interactions between diet–induced postnatal childhood obesity and puberty.

欧洲、美国、日本已开展蒸汽温度为700 °C 及以上的先进超超临界(A-USC) 发电厂用材料开发项目,以实现更高能效和低二氧化碳排放量。近年来中国、印度等也开展了上述项目的开发和研究。这些项目涉及采用镍基合金更换马氏体9%~12%Cr 钢以用于最高温度的锅炉和涡轮部件,从而在700 °C 及以上温度下提供充足的蠕变强度。为尽量减少对昂贵的镍基合金的需求,马氏体9%~12%Cr 钢可用于A-USC 电厂中的次高温部件(最高温度不超过650 °C)。

急性肝衰竭(acute liver failure, ALF)发病突然,常常导致死亡。既往动物实验发现,口服益生菌或抗生素都可以减轻药物引起的肝损伤,提示肠道菌群在药物性肝损伤的病理生理过程中发挥着重要作用。然而,其潜在的作用机制尚不完全清楚。本研究旨在通过多组学的方法,研究肠道菌群在ALF中的综合作用。广谱抗生素(broad-spectrum antibiotics, Abx)灌胃4周,明显改善了D-盐酸半乳糖胺/脂多糖[D-(+)-galactosamine hydrochloride/lipopolysaccharide, D-Gal/LPS]诱导的 ALF 小鼠的生存期。RNA 测序显示,在Abx干预的ALF小鼠中,肝脏的炎症反应受到抑制,而整体代谢途径上调。肠道菌群16S rRNA测序发现,Abx重塑了肠道菌群的结构和功能,伴随肠道菌群的色氨酸(tryptophan, Trp)代谢通路增强。此外,经超高效液相色谱串联质谱(ultraperformance liquid chromatography-mass spectrometry, UPLC‒MS)代谢谱检测提示,Abx 干预肠道菌群后,减少了 Trp 的排泄,释放了更多的 Trp 予宿主,犬尿氨酸(kynurenine, Kyn)水平升高,Kyn通路增强。Kyn作为一种内源性芳香烃受体(aryl hydrocarbon receptor, AhR)的配体, 可以通过结合AhR发挥抗炎和免疫抑制的作用。此外,AhR靶向干预了影响了经Abx预处理或未经Abx预处理的ALF小鼠的预后,表明AhR通路在一定程度上调节了机体对ALF的易感性。本研究表明,依赖肠道菌群调控的色氨酸代谢可以通过调节AhR的活性,影响宿主对ALF的易感性,为更好地管理ALF提供了一个有前景的靶点。
