Effects of 3-aminobenzamide on poly (ADP-ribose) polymerase expression, apoptosis and cell cycle progression

摘要: The aim of this paper is to study the changes of apoptosis and cell cycle progression in HeLa cells after the poly (ADP-ribose) polymerase (PARP) was inhibited by its inhibitor 3-aminobenzamide (3-AB) and the mechanisms of PARP action on HeLa cells damaged by irradiation. Flow cytometry (FCM) was used to examine the PARP expression and the percentage of apoptotic cells and cell cycle progression. The percentage of HeLa cells with positive expression of PARP protein 2, 4, 8 and 12 h after administrated with 3-AB was significantly lower than that of the control ( < 0.01). The percentages of apoptotic cells in the 3-AB plus irradiation group at the time points of 2, 8, 12 and 24 h after 2 Gy irradiation were higher than that in the irradiation group ( < 0.01 or < 0.05) and the percentage of G cells decreased significantly ( < 0.01 or < 0.05). It indicates that 3-AB can rapidly inhibit PARP expression of HeLa cells, promote cell apoptosis and block G arrest induced by irradiation.

Ternary phase behavior of water microemulsified diesel-palm biodiesel

摘要: This paper aims to develop a new microemulsions system comprising diesel and palm oil methyl ester (PME) that have the potential to be used as alternative fuels for diesel engines. The water-in-diesel-biodiesel microemulsions were prepared by applying PME mixed with diesel, non-ionic surfactants, co-surfactants and water to make the water-in-oil (W/O) microemulsion system. This microemulsified fuel was achieved through low-energy microemulsification by using the constant composition method. The diesel used was mixed with four different concentrations of PME, i.e., 10% (w/w) (B10), 20% (w/w) (B20), 30% (w/w) (B30) and neat diesel (B0). The amount of water was fixed at 20% (w/w). The phase behavior of the water/mixed non-ionic surfactant/diesel-PME system were studied by constructing pseudoternary phase diagrams with the goal of formulating optimized systems. The results showed that the microemulsions were formed and stabilized with a mixture of non-ionic surfactants at a weight ratio of 80:20 at 20% (w/w), and with mixed co-surfactants at a weight ratio of 25:75, 20:80 and 10:90 for B0, B10, B20 and B30 respectively. The particle size, kinematic viscosity at 40°C, refractive index, density, heating value, cloud point, pour point and flash point of the selected water-in-diesel microemulsion were 19.40 nm (polydispersity of 0.12), 2.86 mm /s, 1.435, 0.8913 g/mL, 31.87 MJ/kg, 7.15°C, 10.5°C and 46.5°C respectively. The corresponding values of the water-in-diesel-PME selected were 20.72 nm to 23.74 nm, 13.02 mm /s to 13.29 mm /s, 1.442, 0.8939 g/mL to 0.8990 g/mL, 31.45 MJ/kg to 27.34 MJ/kg, 7.2°C to 6.8°C, 8.5°C to 1.5°C and 47.5°C to 52.0°C. These preliminary findings were further studied as potential fuels for diesel engines.

摘要: 移动设备制造商在全球范围内快速开发各种Android版本。同时,网络罪犯也在实施各种恶意行为,例如跟踪用户活动、窃取个人数据以及实施银行诈骗。由于在日常生活中使用Android进行重要通信的人群数量庞大,这些网络罪犯从中获得了巨大非法收益。为此,安全从业者通过静态和动态分析对恶意软件进行识别。静态分析具有整体代码覆盖、低资源消耗和快速处理的优势。然而,静态分析需要最少量的特征才能对恶意软件进行有效分类。因此,我们采用基于遗传算法(GA)的遗传搜索(GS)在106个字符串中选择特征。为评估由GS确定的最佳特征,我们使用了5种机器学习分类器,分别是Naïve Bayes(NB)、功能树(FT)、J48、随机森林(RF)和多层感知器(MLP)。在这5种分类器中,FT仅使用6种特征,获得最高准确度(95%)和最高真正率(TPR)(96.7%)。

Synthesis of copolymers of 3-acryloyloxymethyl-3′-methyloxetane and 3-(2-(2-(2-Methoxyethylenoxy)ethylenoxy)ethylenoxy)-3′-methyloxetane and their ionic conductivity properties

摘要: An oxetane-derived monomer, 3-acryloyloxymethyl-3′-methyloxetane (AMO) was prepared from the reaction of 3-hydromethyl-3-methyloxetane with acryloyl chloride. The cationic ring-opening copolymerization of AMO with another oxetane-derived monomer, 3-(2-(2-(2-methoxyethylenoxy)ethylenoxy)ethylenoxy)-3′-methyloxetane (MEMO) was conducted in CHCl solution using BF3 ·OEt/1, 4-butanediol as a co-initiator. The resulting copolymers were characterized by FTIR, H NMR and Gel Permeation Chromatography (GPC) analyses, and it was found that the enchained ratio of AMO in the copolymers is far lower than its feed ratio. They were crosslinked via the radical polymerization of the vinyl group initiated by BPO after doping with lithium trifluoromethanesulfonimide (LiTFSI) to give rise to tough polymeric electrolyte films. The ionic conductivity was measured at varying content of AMO and different concentration of lithium salt LiTFSI by AC impedance, and a maximum ion conductivity of 1.44×10 S/cm at 30°C or 1.25×10 S/cm at 80°C was attained in the sample PAM 33 at the mole ratio of O : Li = 20. The DSC results indicated that decreases with the increase of the proportion of AMO in the copolymer, well consistent with the ion conductivity trend. The TGA (thermogravimetric analysis) measurement revealed that this kind of copolymer electrolytes is more thermostable than their liquid counterparts.

Heterologous expression of signal protein 14-3-3 in and the subsequent immune response in mice

摘要: Schistosomiasis japonica, a zoonosis caused by , is endemic to the Philippines and China. Several vaccine candidates have been identified and tested in different animal models, but it is still unclear which will be optimal for testing in the field. Therefore, new antigens and strategies are necessary for vaccine development against schistosomiasis japonica. The Sj14-3-3 gene was amplified and subcloned into the expression vector pPICZ?-B and transformed into X-33 by electroporation. Three transformants were induced with methanol. The cultural supernatant was collected and tested by SDS-PAGE and Western blotting. The protein of rSj14-3-3 was prepared and purified and BALB/c mice were immunized which was followed by a challenging infection. The immuno-protection was then evaluated. The Sj14-3-3 gene was expressed and secreted into the medium and its molecular weight was about 35000 as determined by SDS-PAGE. Western blotting showed that the protein had a high specificity against mouse-anti-Sj14-3-3 monoclonal antibody and rSj14-3-3 had a promising immune reactivity. The results of the immuno-protective experiments revealed that the worm reduction was 26.0%, 32.2%, and 36.8%, respectively. The number of eggs in liver tissue was reduced by 36.8%, 43.2%, and 46.1%, respectively. The recombinant Sj14-3-3 of eukaryotic expression in was successfully harvested. The molecular vaccine of Sj14-3-3 could partially induce resistance to the infection with in BALB/c mice. The recombinant protein Sj14-3-3 has promising immunological potentials for further approach to the diagnosis and development of molecular vaccine.

叉头框蛋白P3(FOXP3)是调节性T细胞(Tregs)的一个主要调节因子,调节性T细胞是能抑制抗原特异性免疫反应的T 细胞亚群,在增强宿主耐受性和维持免疫平衡方面发挥着重要作用。众所周知,FOXP3 与多种蛋白质形成复合物,并能通过乙酰化、磷酸化、泛素化和甲基化等各种翻译后修饰(PTM)进行调节。因此,翻译后修饰可改变FOXP3 的稳定性及其调节基因表达的能力,并最终影响调节性T细胞活性。虽然FOXP3 自身并非理想的药物靶点,但脱乙酰酶、乙酰转移酶、激酶和其他可调节FOXP3 的翻译后修饰的酶均为调控FOXP3 和调节性T细胞活性的潜在靶点。但FOXP3 并非这些酶的唯一底物;因此,当使用相关抑制剂时,必须考虑是否存在有害的“FOXP3脱靶”副作用。

Haploinsufficiency of Lipin3 leads to hypertriglyceridemia and obesity by disrupting the expression and

摘要: Lipin proteins including Lipin 1–3 act as transcriptional co-activators and phosphatidic acid phosphohydrolase enzymes, which play crucial roles in lipid metabolism. However, little is known about the function of Lipin3 in triglyceride (TG) metabolism. Here, we identified a novel mutation (NM_001301860: p.1835A>T/p.D612V) of Lipin3 in a large family with hypertriglyceridemia (HTG) and obesity through whole-exome sequencing and Sanger sequencing. Functional studies revealed that the novel variant altered the half-life and stability of the Lipin3 protein. Hence, we generated Lipin3 heterozygous knockout (Lipin3-heKO) mice and cultured primary hepatocytes to explore the pathophysiological roles of Lipin3 in TG metabolism. We found that Lipin3-heKO mice exhibited obvious obesity, HTG, and non-alcoholic fatty liver disorder. Mechanistic study demonstrated that the haploinsufficiency of Lipin3 in primary hepatocytes may induce the overexpression and abnormal distribution of Lipin1 in cytosol and nucleoplasm. The increased expression of Lipin1 in cytosol may contribute to TG anabolism, and the decreased Lipin1 in nucleoplasm can reduce PGC1α, further leading to mitochondrial dysfunction and reduced TG catabolism. Our study suggested that Lipin3 was a novel disease-causing gene inducing obesity and HTG. We also established a relationship between Lipin3 and mitochondrial dysfunction.

Influence and related mechanism of Retn gene expression on glucose uptake in 3T3-L1 cells

摘要: The aim of this article was to investigate the influence and the related mechanism of the gene on glucose uptake and insulin resistance in 3T3-L1 cells. Radioimmunoassay was used to determine glucose uptake in 3T3-L1 cells with different gene expression levels, whether cells were stimulated by insulin or not. RT-PCR and real-time RT-PCR analysis were used to determine the mRNA levels of several glucose transport proteins in 3T3-L1 cells with different gene expression levels, including insulin receptor substrate-1(IRS-1), phosphatidylinositol 3-kinase (PI-3K), AKT-2, glucose transporter-4 (GLUT-4), p38 mitogen-activated protein kinase (p38MAPK) and glycogen synthase kinase-3b (GSK-3). The glucose uptake decreased with the increase in gene expression in 3T3-L1 cells, which was independent of whether the cells were stimulated by insulin or not. The mRNA expression of two signal proteins PI-3K and AKT-2 decreased and the other two signal proteins, GSK-3 and p38MAPK, increased with overexpression in 3T3-L1 cells. Resistin could induce insulin resistance in adipocytes, which might be related to the changes of some proteins in PI-3K and Ras pathways.

Combustion and emissions of RP-3 jet fuel and diesel fuel in a single-cylinder diesel engine

摘要: The combustion characteristics and emission behaviors of RP-3 jet fuel were studied and compared to commercial diesel fuel in a single-cylinder compression ignition (CI) engine. Engine operational parameters, including engine load (0.6, 0.7, and 0.8 MPa indicating the mean effective pressure (IMEP)), the exhaust gas recirculation (EGR) rate (0%, 10%, 20%, and 30%), and the fuel injection timing (−20, −15, −10, and −5 ° crank angle (CA) after top dead center (ATDC)) were adjusted to evaluate the engine performances of RP-3 jet fuel under changed operation conditions. In comparison to diesel fuel, RP-3 jet fuel shows a retarded heat release and lagged combustion phase, which is more obvious under heavy EGR rate conditions. In addition, the higher premixed combustion fraction of RP-3 jet fuel leads to a higher first-stage heat release peak than diesel fuel under all testing conditions. As a result, RP-3 jet fuel features a longer ignition delay (ID) time, a shorter combustion duration (CD), and an earlier CA50 than diesel fuel. The experimental results manifest that RP-3 jet fuel has a slightly lower indicated thermal efficiency (ITE) compared to diesel fuel, but the ITE difference becomes less noticeable under large EGR rate conditions. Compared with diesel fuel, the nitrogen oxides (NOx) emissions of RP-3 jet fuel are higher while its soot emissions are lower. The NOx emissions of RP-3 can be effectively reduced with the increased EGR rate and delayed injection timing.

Overoxidized poly(3,4-ethylenedioxythiophene)-overoxidized polypyrrole composite films with enhanced

摘要: In this study, a simple and effective method was proposed to improve the electrocatalytic ability of overoxidized poly(3,4-ethylenedioxythiophene)-overoxidized polypyrrole composite films modified on glassy carbon electrode for rutin and luteolin determination. The composite electrode was prepared by cyclic voltammetry copolymerization with LiClO4-water as the supporting electrolyte. The peak current of rutin and luteolin on the composite electrode gradually decreased or even disappeared with the increase in the positive potential limit. After incubation in NaOH–ethanol solution with a volume ratio of 1:1, the composite electrodes prepared at positive potential limit greater than 1.5 V exhibited enhanced differential pulse voltammetry peak currents, reduced charge transfer resistance, larger effective specific surface area and higher electron transfer rate constant. The composite electrode prepared in the potential range of 0–1.7 V showed optimal electrocatalytic performance. The X-ray photoelectron spectroscopy results indicated that the content of –SO2/–SO and –C=N– groups in the composite film increased significantly after incubation. Further, the Raman spectra and Fourier transform infrared spectra revealed that the thiophene ring structure changed from benzene-type to quinone-type, and the quinone-type pyrrole ring was formed. The electrocatalytic mechanism of the composite film was proposed based on the experimental results and further verified by Density Functional Theory calculation.

Construction of digital 3D magic-cube organization structure for innovation-driven manufacturing

摘要: Owing to the heterogeneity between functional units and resource scarcity, manufacturing firms have been struggling with intra-organizational coordination for productivity and innovation. Traditional organizational structures, such as linear-functional and matrix, may cause responsibility division and independent goals (Shahani, 2020), and are more difficult to be adopted by large-sized innovative manufacturing firms for quantity production. This is based on a review of several new forms of organization (i.e., network organization, multiteam system, and platform organization) compared with the traditional forms of organization (i.e., linear, matrix, and business unit organization, among others). This study proposes a three-dimensional (3D) magic-cube organizational structure, considering the product dimensions, business, and administration. Moreover, the characteristics, propositions of system operation, system dynamic model, and working model of the 3D magic-cube organization are described. Finally, the 3D model is applied in a Chinese manufacturing firm to test its effectiveness. By redesigning the post and pay system, the pilot organization establishes a project-driven and cross-functional coordination mechanism, positively affecting the firm’s financial profit, output value, labor productivity, and income of per capita. The proposed 3D model can be adopted by large- or medium-sized manufacturing firms for product development and innovation. The implications of both practice and theory are also discussed in this study.
